Summary: Chemotherapy is widely used in gastric cancer treatment as an adjuvant when surgery is not feasible or in the presence of metastasis. Chemoresistance is a major challenge in cancer therapy, leading to tumor recurrence and treatment failure. Despite known mechanisms of chemoresistance, a universal marker for chemoresistant cancer cells has not yet been identified.

Summary: Resistance to anticancer therapeutics is a major challenge in cancer treatment, with altered cancer metabolism, such as the Warburg effect, playing a pivotal role. The use of antiglycolytic agents, such as catechin, may help overcome resistance to 5FU in gastric cancer cells by limiting LDHA expression and lactate production, providing a potential novel adjuvant drug option.

Summary: This study demonstrates that MeCP2 confers 5-FU resistance in gastric cancer cells by upregulating the NOX4/PKM2 pathway. Knockdown of MeCP2 enhances the sensitivity of cells to 5-FU, and silencing NOX4 can rescue the inductive effect of MeCP2 overexpression on 5-FU sensitivity. In vivo experiments also show that MeCP2 knockdown enhances 5-FU sensitivity in tumors.

Summary: The formulation of niosomes is influenced by various variables, including the preparation method and type of non-ionic surfactants used. This study evaluated the effects of different preparation methods and surfactants on the physico-chemical properties, drug release, and cytotoxicity of 5-fluorouracil-loaded niosomes. Thin film hydration method produced the smallest particle sizes and highest drug entrapment efficiency. Real-time cell assay showed sustained cell death caused by the loaded niosomes.

The study found that the expression levels of Nrf2 and ME-1 were decreased in resistant cells, while the expression level of MDR1 was increased. The combined application of Nrf2 inhibitors can be considered as a novel strategy to overcome 5-FU resistance.
