Background: Pituitary adenylate cyclase-activating polypeptide (PACAP) is associated with migraine phase; however, whether PACAP levels could be used to distinguish between migraine and tension-type headache (TTH) remains unknown. We compared interictal plasma PACAP levels among healthy controls, migraineurs, and patients with TTH. Methods: Interictal plasma levels of PACAP were measured in 133 migraineurs, 106 patients with TTH, and 50 controls using enzyme-linked immunoassays. We further evaluated the relationships between interictal PACAP plasma concentrations and clinical parameters, such as headache severity, attack frequency, and duration. Results: We found that migraineurs had significantly lower interictal plasma PACAP levels than patients with TTH and healthy controls. However, there were no significant differences between patients with TTH and healthy controls. Plasma PACAP levels were significantly lower in patients with episodic migraine (EM) than in patients with episodic tension-type headache (ETTH) and in patients with chronic migraine (CM) than in patients with chronic tension-type headache (CTTH). Interictal PACAP levels were negatively correlated with duration in the CM group. Conclusions: The results of this study demonstrated differences in interictal PACAP levels in migraine and TTH, suggesting that PACAP is involved in the pathogenesis of migraine rather than TT-I. Summary: Mutations in ATP1A2 gene can lead to phenotypes ranging from pure FHM to FHM with epilepsy and intellectual disability due to varying degrees of deficits in the biochemical and electrophysiological properties of Na+/K+-ATPase. Mutations associated with intellectual disability result in severe impairment of Na+/K+-ATPase, while the presence of epilepsy or the type of epilepsy does not seem to affect the degree of pump function impairment.

Furthermore, the levels of cytokines CCL7, CCL12, and IL-10 may be modulated by astrocytic activation, contributing to the pathology of chronic migraine.

Targeted Metabolomics Based on LC-MS/MS Revealing Alteration of Bile Acids in Male Migraine Patients

Summary: In this study, the levels of bile acid metabolites were compared in the serum of migraine patients and healthy controls, and it was found that there were differences between male patients and male controls, suggesting a gender-related variation in bile acids. This highlights the importance of gender-stratified analysis in studying diseases with varying morbidity in males and females. Five differential metabolites may serve as potential serum biomarkers for male migraine patients.

Accurate identification of HLA-B*15:02 allele by two-dimensional polymerase chain reaction

Summary: This study successfully established a 2D-PCR method for identifying HLA-B*15:02 through a two-tube reaction. This method can distinguish HLA-B*15:02 from 16 highly homologous HLA-B*15 alleles. Among 1830 samples from the clinical general population, 3 HLA-B*15:02 homozygotes and 84 HLA-B*15:02 heterozygotes were detected.
