Approaches to Quality Risk Management When Using Single-Use Systems in the Manufacture of Biologics

Biologics manufacturing technology has made great progress in the last decade. One of the most promising new technologies is the single-use system, which has improved the efficiency of biologics manufacturing processes. To ensure safety of biologics when employing such single-use systems in the manufacturing process, various issues need to be considered including possible extractables/leachables and particles arising from the components used in single-use systems. Japanese pharmaceutical manufacturers, together with single-use suppliers, members of the academia and regulatory authorities have discussed the risks of using single-use systems and established control strategies for the quality assurance of biologics. In this study, we describe approaches for quality risk management when employing single-use systems in the manufacturing of biologics. We consider the potential impact of impurities related to single-use components on drug safety and the potential impact of the single-use system on other critical quality attributes as well as the stable supply of biologics. We also suggest a risk-mitigating strategy combining multiple control methods which includes the selection of appropriate single-use components, their inspections upon receipt and before releasing for use and qualification of single-use systems. Communication between suppliers of single-use systems and the users, as well as change controls in the facilities both of suppliers and users, are also important in risk-mitigating strategies. Implementing these control strategies can mitigate the risks attributed to the use of single-use systems. This study will be useful in promoting the development of biologics as well as in ensuring their safety, quality and stable supply.

Violacein improves recombinant IgG production by controlling the cell cycle of Chinese hamster ovary cells

Summary: This study evaluated the potency of violacein for cell cycle control and improvement of IgG production in CHO cells. Results showed that violacein increased IgG production by inducing G2/M phase arrest and without affecting product quality. This suggests that violacein has potential as a mAb production enhancer in CHO cells.

Summary: GRP94 is a chaperone protein involved in folding of unfolded proteins in the endoplasmic reticulum, and the truncated form without the KDEL motif can be naturally secreted into the medium in response to ER stress. This secretion phenomenon may shed light on the functional role of GRP94 as a trafficking molecule during the unfolded protein response.

A chemically-defined plastic scaffold for the xeno-free production of human pluripotent stem cells

Summary: In this study, we optimized the protocol for GMP-compatible CRISPRCas9 genome editing to simultaneously deplete three gene loci in HLA homozygous iPSCs. The successful depletion of HLAs was confirmed by RNA-seq and flow cytometry analyses, and the differentiation into cardiomyocytes was verified. However, unexpected genomic changes were observed in some clones, highlighting the importance of multidimensional analyses to ensure the safety and quality of genome-edited cells.

Effect of co-overexpression of the cargo receptor ERGIC-53/MCFD2 on antibody production and intracellular IgG secretion in recombinant Chinese hamster ovary cells

Summary: Therapeutic antibodies are highly effective biopharmaceuticals with fewer side effects and longer half-life in the blood. This study focused on the intracellular trafficking of antibody-producing CHO cells and identified the accumulation of antibodies in the endoplasmic reticulum (ER) due to insufficient cargo receptors. Overexpression of the cargo receptor complex, ERGIC-53-MCFD2, improved antibody production but also increased intracellular antibody retention.

Induced hepatic stem cells are suitable for human hepatocyte production

Artificial induction of chromosome aneuploidy in CHO cells alters their function as host cells

Summary: This study investigated the effects of artificial changes in the chromosome number on productivity in CHO cells. It was found that cell populations with a diverse chromosome number distribution exhibited higher antibody productivities. Additionally, inducing aneuploid cells using 3-aminobenzamide increased antibody productivity. Furthermore, using host cells with an average chromosome number of 37 significantly improved antibody productivity and increased the integration of exogenous genes into the genome.

Semi-3D cultures using Laminin 221 as a coating material for human induced pluripotent stem cells

Summary: Previous belief regarding the compatibility of human induced pluripotent stem cells (hiPSCs) and the coating material Laminin 221 has been challenged by this study. It was found that hiPSCs derived from human mononuclear cells can form peninsular-like colonies when cultured on Laminin 221, and these colonies can be passaged multiple times. The expression of undifferentiated markers tends to be higher in hiPSCs cultured on Laminin 221, and these cells also showed the ability to differentiate into cardiomyocytes on Laminin 221.
