Summary: The synthesis and comprehensive pharmacological evaluation of M4 mAChR PAMs structurally related to 1e, Me-C-c, [11C]MK-6884 and [18F]12 were reported. Small structural changes to the PAMs resulted in significant differences in baseline, potency, and maximum effect measures in cAMP assays compared to the endogenous ligand acetylcholine (ACh). The discovery of novel PAMs with improved allosteric properties, 6k and 6l, and their ability to cross the blood-brain barrier make them more suitable for future preclinical assessment.

Through continued optimization, two compounds, 8b (VU6005610) and 20a (VU6005852), were found to have robust selectivity for the M-1 mAChR with no agonism. Additionally, compound 8b showed high brain exposure in a rodent IV cassette model.

Summary: Recent evidence suggests that inhibition of the M5 muscarinic acetylcholine receptor (mAChR) may provide a novel non-opioid mechanism for the treatment of opioid use disorder (OUD). In this study, the effects of the M5 negative allosteric modulator (NAM) VU6008667 on oxycodone self-administration and reinstatement behaviors were evaluated. The results showed that VU6008667 decreased oxycodone self-administration and attenuated cue-induced reinstatement of lever pressing, while having minimal effects on other behaviors.

Summary: CBX proteins are cell-type-specific paralogous chromobox proteins that repress developmental genes. We have identified a potent positive allosteric modulator (PAM), UNC7040, for CBX8, which disrupts its binding to H3K27me3 and enhances interactions with nucleic acids. Treatment with UNC7040 efficiently and selectively removes CBX8-containing PRC1 from chromatin, resulting in gene desilencing and reduced proliferation in cancer cells.

Summary: Arc is a protein crucial for cognitive function and memory formation, able to self-assemble into virus-like capsids for mRNA transportation. The study shows that TNIK kinase can modify Arc at Serine 67 and Threonine 278, influencing its subcellular distribution and self-assembly. Understanding the interaction between Arc and TNIK in neuronal cells could offer new insights into psychiatric disorders.

Summary: This study found that diabetic animals exhibit severe xerostomia characterized by reduced saliva flow rate, diminished total protein content, and decreased amylase activity. The impaired saliva production in diabetes is associated with reduced and delayed intracellular Ca2+ signals in submandibular acinar cells, caused by malfunctioning mitochondria. Targeting malfunctioning mitochondria may be a potential strategy for the treatment of diabetic xerostomia.

Summary: The coexistence of pain and depression is frequently observed in patients with chronic pain and depression. Oxytocin, a neuropeptide, has been reported to relieve chronic pain and depressive symptoms. This study investigated the effect of intranasal oxytocin on neuropathic pain and comorbid depressive symptoms, and found that oxytocin attenuated depression-like behavior but did not alleviate mechanical hyperalgesia. The results suggest that intranasal oxytocin may have the potential to treat depressive symptoms in neuropathic pain patients.
