Abrocitinib versus Placebo or Dupilumab for Atopic Dermatitis

Summary: The study demonstrated that both 200 mg and 100 mg doses of abrocitinib showed significant efficacy in treating moderate-to-severe atopic dermatitis compared to placebo, with the 200 mg dose being superior to dupilumab in improving itch response. Neither dose of abrocitinib showed significant differences from dupilumab in most other key secondary endpoints at week 16.

Summary: This retrospective study analyzed the data of 10 patients with moderate to severe atopic dermatitis who were treated with dupilumab and subcutaneous immunotherapy (SCIT). The results showed that the combination therapy could safely improve the treatment response of patients with moderate to severe atopic dermatitis who were resistant to dupilumab or SCIT monotherapy.

Treatment of Severe Atopic Dermatitis with Dupilumab in Patients with Advanced Cancer

Summary: Patients with moderate-to-severe atopic dermatitis or with difficult-to-treat areas may benefit from systemic therapy, including targeted biological therapy such as dupilumab. A case series involving three patients with advanced cancer and severe atopic dermatitis treated with dupilumab for an average of 17 months showed a good response towards atopic dermatitis without cancer recurrence. These findings suggest that dupilumab can safely control atopic dermatitis in patients with advanced cancer.

Rapid response to dupilumab treatment in children with moderate-to-severe atopic dermatitis: A case series

Summary: Atopic dermatitis (AD), a chronic and inflammatory skin disease, can have a significant impact on the quality of life for children and their caregivers. Severe cases of AD that are not effectively controlled with topical therapy often require systemic corticosteroids or phototherapy, but these treatments can have unfavorable adverse effects. Dupilumab, a fully human monoclonal antibody, has shown effectiveness in treating type 2 inflammatory diseases, including AD. This study reports successful treatment of severe AD in three pediatric cases using dupilumab.

Management of inadequate response and adverse effects to dupilumab in atopic dermatitis

Summary: Atopic dermatitis is a chronic inflammatory skin disease, and dupilumab is the only approved systemic therapy for moderate-to-severe cases. Limited literature is available on treatment failure with dupilumab, and individual goals and quality of life should be considered. Possible confounding dermatoses should be ruled out.

Summary: This study found that BDD symptoms were fivefold more prevalent in dermatological patients than in healthy skin controls, and were associated with young age, female sex, psychological stress, and feelings of stigmatization. Certain patient groups had a significantly higher chance of BDD symptoms compared with controls. Doctors should consider BDD more often in patients with dermatological conditions and refer them when necessary for assessment and treatment.

Transcriptomic, Epigenomic, and Neuroanatomic Signatures Differ in Chronic Prurigo, Atopic Dermatitis, and Brachioradial Pruritus

Summary: This study aimed to link gene expression, DNA methylation, and neuroanatomy in different chronic pruritis conditions. Results showed that specific signatures of gene expression and DNA methylation distinguished pruritic lesional skin from nonpruritic skin and healthy skin. Neuroanatomy analysis revealed differential branching patterns among the different conditions, indicating that chronic itching conditions are primarily caused by nerve injury and subsequent sprouting, while genuine neuropathy is expected in brachioradial pruritus.

Summary: Patients with prurigo nodularis often have sleep disturbances due to itchy skin in the evening and at night. This study evaluates the Sleep Disturbance Numerical Rating Scale (SD NRS) as a tool for measuring sleep in prurigo nodularis patients. Results show that the SD NRS is easy to use and can be useful for assessing sleep on a daily basis.
