Exploratory study of the association of steroid profiles in stimulated ovarian follicular fluid with outcomes of IVF treatment

Steroid concentrations in stimulated follicular fluid (sFF) samples have been linked to the quality of oocytes used in IVF treatments. Most of the published studies focused on evaluating the association of the IVF outcomes with only a few of the steroids, measured by immunoassays (IA). We performed a treatment outcome, prospective cohort study using stimulated FF sampled from 14 infertile women undergoing IVF treatment; single oocyte was used per IVF cycle. Fourteen endogenous steroids were analyzed in 22 ovarian follicle aspirations, which corresponded to the embryos used in the IVF. Ten oocytes were associated with live birth (LB) and 12 with no pregnancy (NP). Steroids were analyzed using liquid chromatography-tandem mass spectrometry (LC-MS/MS) methods. Differences in distribution of concentrations in association with the pregnancy outcome (LB or NP), and receiver operating characteristic (ROC) curves analysis were performed for the entire cohort and for within-women data. The predominant androgen and estrogen in stimulated sFF were androstenedione (A4) and estradiol (E2), respectively. Lower concentrations of pregnenolone (Pr), lower ratios of A4/ dehydroepiandrosterone (DHEA), testosterone (Te)/DHEA, and greater ratios of E2/Te, and estrone/A4 were observed in sFF samples associated with LB. Among the oocytes associated with NP, in four out of 12 samples total concentration of androgens was above the distribution of the concentrations in the oocytes corresponding to the LB group. Observations of the study indicated increased consumption of precursors and increased biosynthesis of estrogens in the follicles associated with LB. Our data suggest that potentially steroid profiles in sFF obtained during oocyte retrieval may serve as biomarkers for selection of the best embryo to transfer after IVF. (C) 2015 Elsevier Ltd. Further exploration of LKYN's therapeutic potential in stroke and epilepsy in human studies is warranted.

Summary: CPSE concentration is associated with future growth of the prostate in dogs, while concentrations of corticosterone and corticosteroids are positively associated with the change in relative prostatic size over time. Concentrations of nine endogenous steroid hormones in 70-year-old men and women

Summary: This study examined sex-specific plasma concentrations of endogenous sex and adrenal steroids in elderly Swedish Caucasians, finding that estradiol, estrone, and testosterone levels were higher in 70-year-old men compared to women. Body mass index was positively associated with estradiol and estrone in women, while negatively associated with testosterone in men. Apparently healthy individuals showed differences in hormone concentrations compared to all individuals.

Endogenous Levels of Gamma Amino-Butyric Acid Are Correlated to Glutamic-Acid Decarboxylase Antibody Levels in Type 1 Diabetes

Summary: Gamma-aminobutyric acid (GABA) is an important inhibitory neurotransmitter found in the central nervous system and immune cells. Research has shown that GABA has immune-modulatory and beta-cell stimulatory effects and could potentially be used to treat type 1 diabetes. However, a study measuring the levels of GABA in T1D patients found that the levels were unchanged compared to healthy controls, suggesting that GABA secretion from beta-cells has minimal impact on systemic levels. The study also found correlations between GABA levels and certain cytokines and GAD-autoantibodies, indicating potential alterations in local GABA levels within pancreatic islets.

Silencing DTX3L Inhibits the Progression of Cervical Carcinoma by Regulating PI3K/AKT/mTOR Signaling Pathway

Summary: A study found that there were 46 differentially expressed proteins between cervical carcinoma (CC) and normal tissues, with Integrin beta 6, PPP1CB, TMPO, PTGES3, and DTX3L significantly upregulated in CC, while Desmin was significantly downregulated. Silencing DTX3L could suppress CC cell proliferation, migration, invasion, and tumorigenesis, and enhance cell apoptosis. Furthermore, DTX3L silencing inhibited the activity of the PI3K/AKT/mTOR signaling pathway. These findings suggest that DTX3L may regulate CC progression through the PI3K/AKT/mTOR signaling pathway and could serve as a potential biomarker and therapeutic target for CC.

Glycosylation Profiling of the Neoplastic Biomarker Alpha Fetoprotein through Intact Mass Protein Analysis

Summary: AFP serves as a serum biomarker for liver cirrhosis and hepatocellular carcinoma, with different glycosylation patterns distinguishing these conditions. Core fucosylated AFP-L3 is a marker for malignancy risk in HCC. A method combining immune enrichment, LC-HRMS, and protein analysis was developed to detect endogenous AFP proteoforms and quantify AFP-L3% in serum. The method showed reproducibility and agreement with a commercial assay, and identified AFP glycoforms without core fucosylation (AFPL1) and common AFP-L3 glycoforms (G2FS1 and G2FS2).

Summary: A LC-MS/MS method was developed for the simultaneous identification and quantification of cyanotoxins in edible bivalves. The method successfully detected and resolved two previously detected cyanotoxin congeners separately. The method showed limitations for certain cyanotoxins but overall was specific and robust for the parameters investigated. The method was applied to analyze mussel and oyster samples collected along the Swedish coast, and the results can contribute to future regulatory monitoring for cyanotoxins to ensure seafood safety.

Temporal pathway analysis of cerebrospinal fluid proteome in herpes simplex encephalitis

Summary: This study investigated the temporal changes of the CSF proteome in patients with herpes simplex encephalitis (HSE) and its association with factors such as anti-NMDAR serostatus, corticosteroid treatment, brain MRI, and neurocognitive performance. The results showed that the composition and activation pathways of CSF proteins vary at different stages of the disease. Additionally, it was found that several proteins were less abundant in patients with anti-NMDAR seropositivity compared to seronegativity.
