PD-1 signalling defines and protects leukaemic stem cells from T cell receptor-induced cell death in T cell acute lymphoblastic leukaemia

“T cell acute lymphoblastic leukemia (T-ALL) is an aggressive malignancy. In this study, a rare inhibitory receptor called programmed cell death 1 (PD-1) was identified as a marker for leukemia stem cells (LSCs) in T-ALL. Blocking PD-1 significantly eradicated LSCs and suppressed disease progression, and combination therapy with PD-1 blockade and chemotherapy extended survival. Mechanistically, PD-1+ LSCs had high NOTCH1-MYC activity and were protected against apoptosis. These findings provide a therapeutic approach for eliminating LSCs in T-ALL.”
T cell acute lymphoblastic leukaemia (T-ALL) is an aggressive malignancy with poor prognosis, but a decisive marker and effective treatment for leukaemia stem cells (LSCs) remain unclear. Here, using lineage tracing, limiting dilution assays and in vivo live imaging approaches, we identify rare inhibitory receptor programmed cell death 1 (PD-1)-expressing cells that reside at the apex of leukaemia hierarchy for initiation and relapse in T-ALL. Ablation of PD-1-expressing cells, deletion of PD-1 in T-ALL cells or blockade of PD-1 or PD-1 ligand 1 significantly eradicated LSCs and suppressed disease progression. Combination therapy using PD-1 blockade and chemotherapy substantially extended the survival of mice engrafted with mouse or human T-ALL cells. Mechanistically, PD-1+ LSCs had high NOTCH1-MYC activity for disease initiation. Furthermore, PD-1 signalling maintained quiescence and protected LSCs against T cell receptor-signal-induced apoptosis. Overall, our data highlight the hierarchy of leukaemia by identifying PD-1(+) LSCs and provide a therapeutic approach for the elimination of LSCs through PD-1 blockade in T-ALL.

The average deviation of the mileage positioning is 4.06 m, meeting the requirement of road inspection. The significance of this paper lies in improving the automatic method of mileage positioning with the rapid disposal of abnormal targets on roads.

Variety and quantity of dietary insoluble fiber intake from different sources and risk of new-onset hypertension

Summary: This study investigated the associations between the variety and quantity of insoluble fibers from different food sources and new-onset hypertension. The results showed an L-shaped association between dietary insoluble fibers from vegetables, beans, tubers, and fruits with new-onset hypertension; a reversed J-shaped association between insoluble fibers from whole grains and new-onset hypertension; and no obvious association between insoluble fibers from refined grains and new-onset hypertension. More importantly, a higher variety of insoluble fibers from different sources was associated with a lower risk of new-onset hypertension.

Fasting plasma glucose and alanine aminotransferase on the risk of hepatocellular carcinoma: A nested case-control study

Summary: Based on a prospective cohort study, we found that abnormal fasting plasma glucose (FPG) and elevated alanine aminotransferase (ALT) are independent risk factors for hepatocellular carcinoma (HCC), and they have a synergistic effect on the risk of HCC. Therefore, serum FPG and ALT levels should be monitored to prevent the development of HCC.

MARVELD1 stabilizes PARP1 through PARylation and enhances NAA50-mediated acetylation, forming a positive feedback loop. This interaction between MARVELD1 and PARP1 contributes to therapeutic resistance and disrupts the effectiveness of PARP inhibitors in colorectal cancer models.

Achieving multi-cycle regeneration of activated carbon and Cr(VI) removal over a wide pH range by hydrothermal converting quinonimine dye into difunctional pyrrolic-N: Implication for carbon capture in printing and dyeing wastewater treatment

Summary: Activated carbon (AC) adsorption is commonly used for printing and dyeing wastewater (PDW) treatment, but its efficacy is limited by active site depletion. This study presents a sustainable hydrothermal regeneration method for AC exhausted with methylene blue (MB). The absorbed MB is converted to pyrrolic-N-containing hydrochar attached to the regenerated AC, allowing for multiple adsorption-regeneration cycles. The pyrrolic-N-loaded AC counters the negative impact of high pH on Cr(VI) reductive removal, enabling recycling of the regenerated AC. This approach provides a green methodology for simultaneous removal of dye and Cr(VI) in PDW.

Fipronil and its transformation products in the Yangtze River: Assessment for ecological risk and human exposure

Summary: This study aims to fill the data gap regarding the contamination of the Yangtze River by Fipronil (FP) and its transformation products (FPs). FPs concentrations significantly increased from the upper to the lower reaches of the river, with an increase of 13-15% for every 100 km. The urban region and wet season had higher FPs contamination. Human exposure risk through water ingestion was acceptable, but ecological risk assessment showed a moderate to high risk posed by FPs. Regular monitoring of FPs in the Yangtze River is necessary due to their high ecological risk.

Summary: In this study, a search for the heavier partner of the recently observed Z(cs)(3985)(-) state, denoted as Z'(-)(cs), was conducted using e(+)e(-) collision data collected at the BESIII detector. The Z'(-)(cs) is of interest as a potential candidate for a hidden-charm and open-strange tetraquark. The results suggest an excess of Z'(-)(cs) -> D-s(*-) D-*0 (c.c) candidates, indicating the presence of this state with a mass of (4.123.5 +/- 0.7(stat.) +/- 4.7(syst.)) MeV/c(2). Further studies with larger data samples are needed to confirm and clarify the nature of this state.

Predictive nomogram models for unfavorable prognosis after aneurysmal subarachnoid hemorrhage: Analysis from a prospective, observational cohort in China

Summary: This study aimed to identify predictors for 3-month poor functional outcome or death after aSAH and develop precise and easy-to-use nomogram models. The study was conducted at Beijing Tiantan Hospital, involving a total of 310 aSAH patients as a derivation cohort and 208 patients as an external validation cohort. The results showed that the constructed nomogram models could effectively predict poor functional outcome and death, demonstrating good performance in both internal and external validation cohorts.

Summary: This study compared the efficacy and safety of hyperfractionation with standard fractionation in intensity-modulated radiotherapy. Ratiometric mechanoluminescence in single Pr3+-activated LiGa5O8

Summary: This study proposes a new method for stress sensing using mechanoluminescence (ML) materials. The relationship between ML intensity and the change in positional symmetry under stress is determined. The reliability of ML intensity ratio under different factors is systematically analyzed, and a ratiometric ML-based strategy is found to improve stress sensing.

A NIR-mediated MXene@CuS composite for eliminating osteosarcoma and promoting osteogenesis

Summary: Increasing attention is being focused on malignant bone tumors originating from postoperative recurrence and bone defect problems. The urgent need for a multi-functional platform to achieve efficient tumor treatment and bone regeneration has prompted the design of a multifunctional nanoparticle of copper sulfide-modified Ti3C2TX MXene decorated with dopamine (PDA). These nanoparticles demonstrate excellent photothermal properties and anti-tumor ability, rapidly killing osteosarcoma cells (Saos-2 cells) within 10 minutes under 808 nm laser irradiation by combining photothermal and photodynamic effects. Furthermore, the PDA modification provides the nanoparticles with favorable osteogenic activity.

A Kinematic-Decouple Model for Cable-Driven Manipulators with Series-Parallel Coupling Relationship

Summary: This article proposes a kinematic-decouple model that considers the series and parallel relationships of cable-driven manipulators. The parallel relationship between cables and joints is analyzed based on the structure characteristics. The series relationship of the cable-driven manipulator is solved using the Jacobi pseudo-inverse iterative method. The mapping relationship among task space, joint space, and driving space is established. Finally, the configuration and end-trajectory of cable-driven manipulators are simulated, and the experiment confirms the effectiveness of the kinematic-decouple model.

Summary: Pancreaticoduodenal artery aneurysm (PDAA) is rare and can have high rupture risks. This case report describes a 55-year-old female patient with abdominal pain initially diagnosed as acute pancreatitis, but later found to have a rupture and hemorrhage of a small pancreaticoduodenal aneurysm. Interventional treatment was performed, successfully occluding the pseudoaneurysm. This report helps improve understanding of PDAA and provides a basis for clinical treatment.

Study on in situ modification mechanism of mg-Ce-Y based hydrogen storage alloy by ZnF2

Summary: The Mg90Ce5Y5 + x wt% ZnF2 (x = 3, 6, 10, 15) composites were prepared by mechanical ball-milling, and their effects on the hydrogen absorption and desorption properties of Mg90Ce5Y5 alloy were investigated. The addition of ZnF2 introduced MgF2 and MgZn2 phases, which acted as catalysts and improved the hydriding-dehydriding characteristics. Mg90Ce5Y5 + 6 wt% ZnF2 composites exhibited the fastest hydrogen adsorption/desorption kinetics and lower decomposition temperatures.
