We previously reported the discovery of a CXCL12-mimetic cyclic peptide (2) as a selective CXCR4 antagonist showing promising in vitro and in vivo anticancer activity. However, further development of this peptide was hampered by its degradation in biological fluids as well as by its low micromolar affinity for the receptor. Herein, extensive chemical modifications led to the development of a new analogue (10) with enhanced potency, specificity, and plasma stability. A combined approach of Ala-amino acid scan, NMR, and molecular modeling unraveled the reasons behind the improved binding properties of 10 vs 2. Biological investigations on leukemia (CEM) and colon (HT29 and HCT116) cancer cell lines showed that 10 is able to impair CXCL12-mediated cell migration; ERK-phosphorylation, and CXCR4 internalization. These outcomes might pave the way for the future preclinical development of 10 in CXCR4 overexpressing leukemia and colon cancer.

Summary: Synthetic nucleic acid interactors have exciting biotechnological and potential therapeutic applications. This study focuses on the synthesis of homo-thymine nucleopeptides and investigates the influence of underivatized amino acid residues and the distance between the nucleobase and peptide backbone on nucleic acid recognition. The results show that two of the nucleopeptides can efficiently bind DNA and RNA targets and cross membranes.

Summary: The study optimized the extraction process of Ursolic acid (UA) from Annurca apple (AA) using sunflower oil as a solvent, and identified twenty-three phenolic and terpenoid molecules in the extract. The optimized extract, named Optimized Annurca Apple Oleolyte (OAAO), showed significant antioxidant activity and could be considered a safe and functional ingredient for cosmetic formulations.

Natural Polyphenols for Prevention and Treatment of Urinary Tract Infections

Summary: Urinary tract infections (UTIs) are a common bacterial infection, especially in women. The main etiological agents are E. coli, Pseudomonas aeruginosa, and Proteus mirabilis. Due to the increase in antimicrobial resistance, there is a need for natural alternatives for UTI treatment. This review summarizes the potential therapeutic effects of natural polyphenol-based nutraceuticals and foods through in vitro and in vivo studies.

Two newly developed compounds, 19 and 34, are selective KAT8 inhibitors with low-micromolar activity and show antiproliferative effects in cancer cell lines, while not affecting nontransformed cells. These compounds provide valuable tools for studying KAT8 biology and have potential for further optimization studies.

Streamlining Large Chemical Library Docking with Artificial Intelligence: the PyRMD2Dock Approach

Summary: This research introduces a novel computational protocol called PyRMD2Dock, which combines the Ligand-Based Virtual Screening (LBVS) tool PyRMD with the popular docking software AutoDock-GPU (AD4-GPU), to enhance the throughput of virtual screening campaigns for drug discovery. By implementing PyRMD2Dock, it is possible to rapidly screen massive chemical databases and identify compounds with the highest predicted binding affinity to a target protein. The study showcases the value of combining AI-powered LBVS tools with docking software for effective and high-throughput virtual screening of ultralarge molecular databases in drug discovery. PyRMD and the PyRMD2Dock protocol are freely available on GitHub (https://github.com/cosconatilab/PyRMD) as an open-source tool.
