This article provides an overview of the presence and potential health impacts of microplastics in the biosphere, with a focus on inhalation and ingestion as the major exposure pathways. It discusses the absorption, distribution, metabolic toxicity, and excretion of microplastics in the human body, as well as the potential risks associated with them.
Microplastics (MPs; <5 mm) in the biosphere draws public concern about their potential health impacts. Humans are potentially exposed to MPs via ingestion, inhalation, and dermal contact. Ingestion and inhalation are the two major exposure pathways. An adult may consume approximately 5.1 x 103 items from table salts and up to 4.1 x 104 items via drinking water annually. Meanwhile, MP inhalation intake ranges from 0.9 x 104 to 7.9 x 104 items per year. The intake of MPs would be further distributed in different tissues and organs of humans depending on their sizes. The excretion has been discussed with the possible clearance ways (e.g., urine and feces). The review summarized the absorption, distribution, metabolic toxicity and excretion of MPs together with the attached chemicals. Moreover, the potential implications on humans are also discussed from in vitro and in vivo studies, and connecting the relationship between the physicochemical properties and the potential risks. This review will contribute to a better understanding of MPs as culprits and/or vectors linking to potential human health hazards, which will help outline the promising areas for further revealing the possible toxicity pathways.

Multicellular tumor spheroids bridge the gap between two-dimensional cancer cells and solid tumors: The role of lipid metabolism and distribution

Peisi Xie, Jinghui Zhang, Pengfei Wu, Yongjiang Wu, Yanjun Hong, Jianing Wang, Zongwei Cai

Summary: In this study, the differences in lipid metabolism and distribution among 2D cancer cells, multicellular tumor spheroids (MCTS), and solid tumors were explored. The results showed that MCTS had more shared lipid species with solid tumors compared to 2D cells. In addition, MCTS exhibited more similar characteristics to solid tumors in terms of lipid abundance and spatial distribution. This study provides evidence that MCTS are a more suitable in vitro model for mimicking solid tumors and can contribute to the understanding of tumor metabolism and microenvironment.

New Evidence of Rubber-Derived Quinones in Water, Air, and Soil

Guodong Cao, Wei Wang, Jing Zhang, Pengfei Wu, Xingchen Zhao, Zhu Yang, Di Hu, Zongwei Cai

Summary: The study found that rubber-derived quinones are widely present in urban environments and are closely related to human activities. One of the identified quinones was found to be highly toxic to coho salmon. Mass spectrometry and liquid chromatography-mass spectrometry were used to investigate the distribution and concentration of these compounds. The results showed that the quinones had distinct distribution and concentration patterns in different environments. The estimated daily intake of these compounds by the population in Hong Kong exceeded the exposure levels of their parent compounds. Therefore, further toxicological studies are recommended to explore the potential ecological and human health risks of these newly discovered quinones.

Summary: This study aims to investigate the immunotoxic effect and mechanism of dimethyl phthalate (DMP) on blood immune system. The results showed that DMP caused oxidative damage in rats, suppressed immune defense, and disrupted inflammatory regulation, leading to significantly decreased blood cell counts and disordered antibody and cytokine secretion. Flow cytometry revealed a positive correlation between DMP exposure level and apoptosis/necrosis in lymphocytes, especially CD3(+)CD4(+)T cells. Transcriptomics uncovered an oxidative stress-related mechanism.

Particulate matter-induced metabolic recoding of epigenetics in macrophages drives pathogenesis of chronic obstructive pulmonary disease

Summary: This study reveals that chronic exposure to PM induces chronic inflammation and development of COPD by dysregulating NAD+ metabolism and subsequent SIRT1 deficiency in pulmonary macrophages. Activation of SIRT1 by resveratrol effectively mitigates PM-induced inflammation and COPD development.
