Summary: In this study, the therapeutic potential of GGsTop, a γ-glutamyl transferase inhibitor, on acute asthma exacerbations was investigated using a murine model. The results showed that GGsTop significantly attenuated hallmark features of asthma exacerbation, including airway hyperresponsiveness, mucus hypersecretion, collagen deposition, and expression of inflammatory cytokines. Furthermore, GGsTop restored the level of glutathione and inhibited the activation of the LPS/NFκB signaling pathway.

Summary: This study evaluated the IL-17A and IFN-gamma modulatory effect of two new oxazolidine derivatives on mononucleated cells of patients with moderate and severe asthma. The results showed that the compounds reduced IFN-gamma and IL-17 production in a concentrated manner in peripheral blood mononucleated cells from asthmatic patients. In silico analysis revealed the neurokinin-1 receptor as a common target for both compounds, indicating a potential mechanism for their effects.

Summary: Tip-DCs are key immune effector cells involved in fighting off various infections, but their excessive development can lead to tissue damage. This study reveals that IL-27 plays a crucial role in inhibiting Tip-DC development during parasitic infections by interfering with the CD4(+) T cell-IFN-gamma axis and preserving the balance between Ly6C(-) and Ly6C(+) monocytes. Targeting IL-27 signaling may offer a promising approach for therapeutic intervention to control Tip-DC development.

Summary: Mucosal delivery of IL-27 has been shown to have therapeutic benefit in murine models of inflammatory bowel disease (IBD). However, murine colonoids and primary intact colonic crypts were unresponsive to IL-27 in vitro and lacked detectable IL-27 receptors. On the other hand, macrophages in inflamed colon tissue were responsive to IL-27 in vitro and had potent effects on colonic epithelium.

Summary: Deployment related asthma-like symptoms have been observed in military personnel who served in Iraq and Afghanistan. This study aimed to investigate the effect of respiratory exposure to particulate matter from Afghanistan (PMa) on distal airway hyperresponsiveness (AHR) and the role of IL-13 and mitochondrial dysfunction in this process. The results suggest that PMa, in the presence of IL-13, increases AHR through oxidative stress and ATP signaling.

Summary: Due to the failure of target-specific strategies, a novel multifunctional biomaterial based on peptide nanonets has been developed to selectively trap lipopolysaccharide (LPS), tumor necrosis factor-alpha (TNF-alpha), and interleukin-6 (IL-6) in order to suppress inflammation and restore the antimicrobial activity of the last-resort antibiotic colistin. In addition to its ability to simultaneously trap and kill bacteria, the peptide nanonets have shown remarkable potential as a holistic anti-infective and anti-septic biomaterial.
