Summary: Con A is a T-cell mitogen that has been shown to induce hepatitis in mice by activating conventional T cells and NKT cells. This study used an immunodeficient mouse model and found that Con A can rapidly induce liver injury without the involvement of immunocytes. In vitro experiments showed that hepatocytes underwent dose-dependent caspase-independent apoptosis in response to Con A stimulation. Transcriptome RNA-sequencing analysis revealed the activation of apoptosis pathways in both in vivo and in vitro models. This study provides new insights into the mechanism of Con A-induced hepatitis.
