The proteome characterization of fish muscle tissues, together with the relative expression of each individual protein, provides knowledge on the biochemical response of the organisms and allows to assess the effect of different types of feeding, growth site and nutritional quality of the investigated species. This type of study is usually performed by gel-based proteomics approaches, however shotgun proteomics can serve as well, reducing analysis time and improving sample high-throughput. In this work, a shotgun proteomics method was thus developed and then applied to the characterization of gilthead sea bream edible muscle. The sarcoplasmic protein fraction was extracted, in-solution digested by trypsin and finally analyzed by nanoHPLC high resolution tandem mass spectrometry. Two different quantification strategies were also tested. One was based on chemical dimethyl labeling and the other one on label free quantification. A comparison between these two analytical workflows was performed, to evaluate their individual performance in the analysis of fish samples and assess the differences induced by farming practice on the final commercial product with respect to wild gilthead sea bream. Quantitative differences were detected, and the most relevant one regarded the common fish allergen parvalbumin, found overexpressed in farmed fish samples. (C) 2015 Elsevier B.V. Summary: This study provides in-depth characterization of the perinuclear theca (PT) of boar spermatozoa through subcellular fractionation, revealing 1802 identified proteins. The PT was found to be enriched in proteins involved in sperm-egg fusion and egg activation, as well as endoplasmic reticulum (ER) proteins and the ER-stress response. Protein-protein interaction analysis visualized an intricate network of PT protein complexes, including proteasome subunits.

Investigating the Short Peptidome Profile of Italian Dry-Cured Ham at Different Processing Times by High-Resolution Mass Spectrometry and Chemometrics

Summary: In this study, a suspect screening approach based on high-resolution mass spectrometry was used to comprehensively characterize the short endogenous peptidome in dry-cured ham samples. A total of 236 short peptide sequences, including 173 natural short peptides and 63 sequences containing non-proteinogenic amino acids, were tentatively identified. Several modified short peptides, such as pyroglutamination and lactoyl conjugation, were also observed, suggesting reactions occurring over time.

New hydrophilic material based on hydrogel polymer for the selective enrichment of intact glycopeptides from serum protein digests

Summary: The paper presents the preparation and characterization of a new HILIC material for the enrichment of N-linked glycopeptides. The material was prepared using specific monomer and cross-linker, and by fine-tuning the polymer properties, it displayed superior performance in terms of the number of enriched glycopeptides and selectivity. These results not only provide a new HILIC material for glycopeptide enrichment but also pave the way for the use of hydrogel materials in this field.

Summary: Sperm motility is crucial for successful reproduction, but the ornamentation of axonemal microtubules and its support for motility in different fertilization environments are unclear. This study investigates native axonemal doublet microtubules (DMTs) in sea urchin and bovine sperm and identifies over 60 proteins decorating DMTs, including those linked to infertility. Through analyzing DMTs across species and cell types, the study defines core microtubule inner proteins (MIPs) and examines the evolution of the tektin bundle. Furthermore, a testis-specific serine/threonine kinase that connects DMTs to outer dense fibers in mammalian sperm is identified. The study provides structural insights into sperm evolution, motility, and dysfunction at a molecular level.

Summary: Industrial hemp is being widely used for pharmaceutical and nutraceutical purposes, but the detailed composition of hemp remains poorly understood. In this study, the carotenoids and fat-soluble vitamins in the inflorescences of seven Italian industrial hemp varieties were analyzed. The most abundant carotenoids were found to be phytoene, phytofluene, and all-trans -a-carotene. In addition, several other carotenoids were tentatively identified using liquid chromatography coupled with photodiode-array detection.

Biotic transformation products of sulfonamides in environmental water samples: High-resolution mass spectrometry-based tentative identification by a suspect screening approach

Summary: The presence of pharmaceuticals in the aquatic environment is mainly due to their release from wastewater treatment plants. Sulfonamides, a class of synthetic antibacterials, have often been found in surface water, contributing to antibiotic resistance. Monitoring their transformation products (TPs) is important, as they may still exert pharmaceutical activity. In this study, three commonly used sulfonamides were incubated with activated sludge, and their TPs were identified using mass spectrometry. The identified TPs were then used to analyze wastewater treatment plant samples. Acetylation was found to be the predominant transformation pathway.

The mechanism of how these membrane proteins are transported is poorly understood. Summary: This study elucidated the molecular mechanism underlying the hypocholesterolemic effects of Lupin protein hydrolysates (LPHs) in an in vivo model. The results showed that LPH treatment reduced de novo cholesterol synthesis through the regulation of HMGCoAR and LDLR protein levels. LPHs also controlled the PSCK9 signal pathway, leading to decreased cholesterol absorption. These findings highlight LPHs as a potential ingredient for developing new functional foods.

Breakthrough curve analysis of phosphorylated hazelnut shell waste in column operation for continuous harvesting of lithium from water

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

An integrated approach for studying the metabolic profiling of herbal medicine in mice using high-resolution mass spectrometry and metabolomics data processing tools

Summary: Analysis of in vivo exposure to traditional Chinese medicine using mass spectrometry is helpful for identifying effective ingredients and developing new drugs. This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles

Summary: A quantitative analytical method was developed and validated in this study to determine the concentrations of 81 aroma-relevant wine volatiles. The method utilizes stir bar sorptive extraction and multidimensional gas chromatography with flame ionization detector or mass spectrometer. The method demonstrated good linearity, reproducibility, and recovery, and was successfully applied to analyze 20 wine samples.
