A carrier-free photodynamic nanodrug to enable regulation of dendritic cells for boosting cancer immunotherapy

Researchers have developed a photodynamic nanodrug that can simultaneously regulate multiple steps to enhance the immune response of dendritic cells (DCs), effectively suppressing tumor growth and postoperative recurrence.
Immune response is initiated by dendritic cells (DCs), where the cross-presentation of antigens by DCs determines the activating of cytotoxic T cells. However, the efficacy of DCs-initiated immune response is governed by multiple (cascade) steps of immunogenic cell death (ICD), recruitment of DCs, and cross-presentation of DCs. It is urgent but challenging to achieve a platform for simultaneously regulating these multiple steps, amplifying the immune response against tumors. Herein, we reported a photodynamic nanodrug enabling simultaneous regulation of these multiple steps for realizing powerful immune response. The nanodrug was designed by the co-assembling of chlorin e6 (Ce6), celecoxib and 6-thio-2'-deoxyguanosine (6-thio-dG). In our nanodrug, Ce6 enables induction of ICD, while celecoxib down-regulates the prostaglandin E2 (PGE2) for promoting recruitment of DCs enabled by chemokine CCL5 produced from natural killer (NK) cells. Moreover, 6-thio-dG triggers DNA damages in the tumor cells, which in turn activates STING/interferon I pathway for enhancing the cross-presentation ability of DCs. Therefore, an amplified immune therapeutic effect against tumors is achieved, thanks to the simultaneous regulation of these multiple steps. The nanodrug effectively inhibits tumor growth and postoperative recurrence, demonstrating a new approach for boosting immune response initiated by DCs in cancer therapy. Statement of significance The dendritic cells (DCs)-initiated immune response against tumors is dominated by multiple (cascade) steps including the process of (I) immunogenic cell death (ICD), (II) recruitment of DCs, and (III) cross-presentation of antigens by DCs. Based on this, it is urgent to design a nanoplatform enabling simultaneous regulation of these multiple steps for achieving a potent therapeutic efficacy. A carrier-free photodynamic nanodrug, engineered by a co-assembling approach, was designed to regulate DCs for realizing a powerful DCs-initiated immune response against tumors, thanks to the simultaneous regulation of the above multiple steps. Our nanodrug demonstrated a boosted immune response against tumors, powerfully suppressing primary/abscopal tumor growth and postoperative recurrence, which offers a conceptually innovative strategy for amplifying immunity against tumors. Time-varying additive model with autoregressive errors for locally stationary time series

Summary: This article studies the time-varying additive model with time-varying autoregression error in the locally stationary context and proposes a two-step estimation for it. The B-spline method is used to obtain the initial estimator of trend function and additive components, and ULASSO is used to estimate the structure of autoregression error. The improved estimator of trend function and additive components is derived using local linear smoothing. Simulation studies and a real data application demonstrate the effectiveness and applicability of the proposed model.

Thermogravimetric kinetic analysis of the Ce-Zr composite oxygen carrier prepared by ball milling

Summary: In this study, a series of zirconium-doped cerium-based composite oxygen carriers were prepared using the ball milling method. The addition of carbon nanotubes was found to greatly increase the specific surface area and activity of the oxygen carriers. The kinetics of H2 reduction of the oxygen carriers were best described by a three-dimensional diffusion model. Additionally, the activation energies of the oxygen carriers corresponded with their reactivity sequence.

Influence of post-thaw culture duration on pregnancy outcomes in frozen blastocyst transfer cycles

Summary: This study aimed to evaluate the impact of post-thaw culture duration on the clinical outcomes of frozen blastocyst transfer. The results showed no statistical differences in the main outcomes among the groups following D5 blastocyst transfer. However, patients in the long culture group following D6 blastocyst transfer had significantly lower implantation, clinical pregnancy, and live birth rates, but a higher abortion rate compared to those in the short culture group. No superiority was found among the different short culture durations for D6 blastocyst vitrification in terms of live birth outcomes.

Integrated heat and power optimal dispatch method considering the district heating networks flow rate regulation for wind power accommodation

Summary: This paper proposes a new method of improving wind power accommodation by integrating district heating networks flow rate regulation into power dispatch. The results show that wind power accommodation increases with increasing flow rate due to increased thermal inertia. However, the flow rate cannot be too high as the electricity consumption of water pumps becomes the main reason for improving wind power accommodation.

Impacts of Ce dopants on the hydrogen storage performance of Ti-Cr-V alloys

Summary: Vanadium-based alloys are promising materials for hydrogen storage due to their high capacity at ambient conditions. However, the challenges of complex activation and poor stability at high temperatures limit their large-scale applications. In this study, TiCr3V16Cex (x = 0, 0.1, 0.2, 0.4, 1) hydrogen storage alloys with different Ce contents were developed and investigated for their hydrogen storage performance, activation mechanism, and absorption mechanism. The results showed that Ce doping increased the plateau pressure while slightly decreasing the hydrogen storage capacity. Ce doping also affected the kinetics and thermodynamics of the alloy. The TiCr3V16Cex (x = 0.2, 0.4, 1) alloys exhibited hydrogen absorption and desorption at room temperature without activation. The optimum alloy, TiCr3V16Ce0.2, showed a high hydrogen absorption rate and effective hydrogen desorption capacity. After cycling, the alloy maintained its original capacity. The Ce-doped BCC alloy developed in this study offers a new route for achieving high hydrogen storage performance under mild conditions.

Summary: This study investigated the effects of liquid co-cultivation of Arthrobacter ureafaciens DnL1-1 and Trichoderma harzianum LTR-2 on the metabolome of fermentation liquors. The results showed significant changes in the abundance of metabolites in the fermentation liquors compared to single-cultured strains, and the co-cultivation had significant promoting effects on wheat germination and radicle length.
