The Circadian Biology of Heart Failure

Summary: Cardiac physiology is regulated by autonomous molecular clocks and the suprachiasmatic nucleus, exhibiting diurnal rhythms. Disrupting molecular clocks can lead to heart failure, and the time of day can condition myocardial response to injury. Human studies support the involvement of molecular clocks and circadian rhythms in the pathogenesis of heart failure. Certain circadian rhythms are maintained in heart failure patients, which can guide optimal timing of therapy. Manipulation of circadian rhythms and molecular clocks show promise in the prevention and treatment of heart failure.

Modulation of the Immune Response to Severe Acute Respiratory Syndrome Coronavirus 2 Vaccination by Nonsteroidal Anti-Inflammatory Drugs

Summary: In this small pilot study, it was found that the immune response elicited by the SARS-CoV-2 vaccine was not affected by concomitant use of nonsteroidal anti-inflammatory drugs (NSAIDs). The absence of significant immunomodulation signals suggests that NSAIDs do not impact vaccine efficacy. The gathered information may be useful for designing a larger clinical trial.

Sexual dimorphism in the response to chronic circadian misalignment on a high-fat diet

Summary: Longitudinal studies suggest a relationship between shiftwork and cardiometabolic disorders, but the causes and mechanisms are not clear. We developed a mouse model to study circadian misalignment in both sexes and found that female mice showed preserved rhythmicity and were protected from the cardiometabolic effects of misalignment on a high-fat diet. Differences were observed in the liver transcriptome and proteome, as well as gut microbiome dysbiosis, which only occurred in male mice. Antibiotic treatment reduced the impact of misalignment. In the UK Biobank, female shiftworkers demonstrated stronger circadian rhythmicity and a lower incidence of metabolic syndrome compared to males. Our findings suggest that females are more resilient to chronic circadian misalignment, and this difference is conserved in humans.

Cardiovascular Diseases and Marine Oils: A Focus on Omega-3 Polyunsaturated Fatty Acids and Polar Lipids

Summary: Cardiovascular diseases are the leading cause of death globally. Dietary modification, specifically the supplementation of omega-3 polyunsaturated fatty acids (n-3 PUFAs) and polar lipids (PLs), has shown potential in preventing and reducing the burden of cardiovascular diseases. Clinical trials, such as the REDUCE-IT and STRENGTH trials, provide insights into the effectiveness of n-3 PUFAs in primary and secondary prevention of cardiovascular diseases. However, further research is needed to determine the potential benefits of PL supplementation in reducing cardiovascular risk.
