Currently, there is a growing focus on a holistic approach to health, specifically in relation to disease prevention through diet. Functional food, which not only provides essential nutrition but also has the potential for preventing disorders, has gained increasing interest. Soybean, as a promising functional food, contains high levels of the isoflavone genistein. Genistein has shown potential in preventing and treating various disorders such as psoriasis, cataracts, cystic fibrosis, non-alcoholic fatty liver disease, and type 2 diabetes. However, the achievable concentrations of genistein in humans are low, and there are concerns about potential side effects related to its estrogenic and goitrogenic effects.
Nowadays, increasingly more attention is being paid to a holistic approach to health, in which diet contributes to disease prevention. There is growing interest in functional food that not only provides basic nutrition but has also been demonstrated to be an opportunity for the prevention of disorders. A promising functional food is soybean, which is the richest source of the isoflavone, genistein. Genistein may be useful in the prevention and treatment of such disorders as psoriasis, cataracts, cystic fibrosis, non-alcoholic fatty liver disease and type 2 diabetes. However, achievable concentrations of genistein in humans are low, and the use of soybean as a functional food is not devoid of concerns, which are related to genistein's potential side effects resulting from its estrogenic and goitrogenic effects.

Hot Melt Extrusion as an Effective Process in the Development of Mucoadhesive Tablets Containing Scutellariae baicalensis radix Extract and Chitosan Dedicated to the Treatment of Oral Infections

Magdalena Paczkowska-Walendowska, Andrzej Miklaszewski, Daria Szymanowska, Krystyna Skalicka-Wozniak, Judyta Cielecka-Piontek

Summary: Hot Melt Extrusion (HME) technology was developed to improve the rheological properties of blends containing lyophilized Scutellariae baicalensis root extract and chitosan through the use of different ratios of (Hydroxypropyl)methyl cellulose (HPMC) as amorphous matrix formers. The systems were characterized and tablets were prepared from the extrudates. HPMC-based systems showed delayed release of baicalin due to the swelling behavior of HPMC, and the best tabletability properties were observed in the formulation containing lyophilized extract HPMC 50:50 w/w, which maintained good mucoadhesive properties.

Improving Solubility and Permeability of Hesperidin through Electrospun Orange-Peel-Extract-Loaded Nanofibers

Magdalena Paczkowska-Walendowska, Andrzej Miklaszewski, Judyta Cielecka-Piontek

Summary: The optimization of hesperidin extraction from orange peel was conducted to obtain its highest content, by investigating the influence of extraction mixture and temperature. The results showed that methanol and temperature were key parameters for efficient extraction of bioactive compounds. Furthermore, electrospinning technology was used to prepare nanofibers loaded with hesperidin-rich orange peel extract, which exhibited improved solubility and permeability. These nanofibers demonstrated significant antioxidant and anti-inflammatory activities, suggesting their potential buccal applications.

Do Rutin and Quercetin Retain Their Structure and Radical Scavenging Activity after Exposure to Radiation?

Natalia Rosiak, Judyta Cielecka-Piontek, Robert Skibinski, Kornelia Lewandowska, Waldemar Bednarski, Przemyslaw Zalewski

Summary: The influence of ionizing radiation on the physicochemical properties of quercetin and rutin in the solid state was investigated. The results showed that irradiation caused the formation of a small number of free radicals, but did not change the chemical structure or antioxidant properties of the tested flavonoids. Spectroscopic and chromatographic analyses confirmed the radiostability of quercetin and rutin.

Hot-Melt Extrusion as an Effective Technique for Obtaining an Amorphous System of Curcumin and Piperine with Improved Properties Essential for Their Better Biological Activities

Kamil Wdowiak, Robert Pietrzak, Ewa Tykarska, Judyta Cielecka-Piontek

Summary: To overcome the poor bioavailability of curcumin and piperine, their solubility can be enhanced and bioenhancers can be used to inhibit metabolic transformation processes. The amorphous systems of curcumin and piperine produced using hot-melt extrusion showed improved solubility, permeability, and biological activities. The best system significantly increased dissolution rate, apparent solubility, permeability, and antioxidant and anti-butyrylcholinesterase activities.

Screening of the Anti-Neurodegenerative Activity of Caffeic Acid after Introduction into Inorganic Metal Delivery Systems to Increase Its Solubility as the Result of a Mechanosynthetic Approach

Anna Stasilowicz-Krzemien, Natalia Rosiak, Andrzej Miklaszewski, Judyta Cielecka-Piontek

Summary: To improve the bioavailability of caffeic acid, delivery systems were developed using ball milling and freeze-drying techniques to prepare solid dispersions of caffeic acid and magnesium aluminometasilicate. The 1:1 mass ratio solid dispersion obtained from ball milling was the most effective, and its identity was confirmed using X-ray powder diffraction and Fourier-transform infrared spectroscopy techniques. Screening tests demonstrated the improved anti-neurodegenerative activity of caffeic acid with enhanced solubility, inhibiting acetylcholinesterase, butyrylcholinesterase, tyrosinase, and showing antioxidant potential. In silico studies identified the domains of caffeic acid that interacted with enzymes relevant to neuroprotective activity. Furthermore, improved permeability through gastrointestinal tract and blood-brain barrier mimicking membranes validated the in vivo screening tests for anti-neurodegenerative activity.

Zein as an Effective Carrier for Hesperidin Delivery Systems with Improved Prebiotic Potential

Szymon Sip, Anna Sip, Andrzej Miklaszewski, Marcin Zarowski, Judyta Cielecka-Piontek

Summary: This study aimed to develop a zein-based hesperidin system with improved solubility and sustained release profile. Triple systems enriched with solubilizers were utilized, and the best system was found to be hesperidin-zein-Hp & beta;-CD, with solubility improved by more than six times. Additionally, the solubility improvement resulted in enhanced antioxidant activity and inhibition of & alpha;-glucosidase. Release profile analysis confirmed the sustained release of hesperidin while enhancing solubility. Furthermore, the study investigated the ability of selected probiotic bacteria to metabolize hesperidin and its impact on their growth.

The Antioxidant Potential of Resveratrol from Red Vine Leaves Delivered in an Electrospun Nanofiber System

Magdalena Paczkowska-Walendowska, Andrzej Miklaszewski, Bozena Michniak-Kohn, Judyta Cielecka-Piontek

Summary: Despite the wide range of effects of polyphenols, their low oral bioavailability limits their usefulness. In this study, red vine leaf extract with optimal antioxidant and anti-inflammatory properties was obtained using the Design of Experiment approach. Nanofibers containing the extract were then developed to improve the release and buccal penetration of resveratrol. This innovative strategy enhances the utilization of active compounds for the development of nutraceuticals.
