Summary: This study examined the neurobiological consequences of cocaine abstinence in rhesus monkeys. The results showed that global rates of cerebral metabolism were significantly lower in monkeys 1 day and 30 days after cocaine self-administration, compared to food-reinforced controls. The effects were more pronounced in the prefrontal, parietal, and cingulate cortex, as well as the dorsal striatum and thalamus, after 30 days of cocaine abstinence. The study suggests that these changes in glucose utilization are a result of cocaine exposure rather than conditioned stimuli associated with cocaine.

Summary: The study assessed the modulating effects of chronically administered levetiracetam on the reinforcing strength of cocaine in monkeys. Lower doses of levetiracetam caused non-systematic fluctuations in cocaine injections received, while the highest tested dose significantly increased cocaine reinforcement. After termination of levetiracetam treatment, sensitivity to cocaine was increased in two monkeys.

Summary: Identifying neurobiological characteristics that predict the development of cocaine use disorder would be valuable in prevention efforts. Brain dopamine receptors, specifically D2R and D3R, play important roles in mediating the abuse-related effects of cocaine. Analysis of data from studies on rhesus monkeys suggests that D2R availability in the caudate nucleus may be related to initial sensitivity to cocaine, while D3R sensitivity is associated with the dose at which monkeys acquired cocaine self-administration. These findings suggest the potential utility of D3R sensitivity as a biomarker for vulnerability and resilience to cocaine.

Summary: The cyclin D-CDK4/6 complexes are important in controlling the cell cycle, and their deregulation can lead to uncontrolled cell proliferation. While CDK4/6 inhibitors have shown impressive results in treating hormone positive advanced breast cancer, about 30% of breast cancer cases are intrinsically resistant to these inhibitors and prolonged treatment leads to acquired resistance. The mechanisms of resistance involve both cell-cycle and non-cell-cycle related strategies, and understanding these mechanisms may lead to the development of therapeutic strategies to overcome resistance.
