Summary: In this study, novel compounds were synthesized from furanone derivatives and tested for their anti-inflammatory activities against COX enzymes, 15-LOX, and TNF-alpha. The majority of the compounds showed significant to high activities, with three pyridazinone derivatives identified as the most promising anti-inflammatory agents with dual COX-2/15-LOX inhibition activities and high TNF-alpha inhibition activity.

Summary: Drug delivery to ocular tissues is hindered by anatomical and physiological barriers, resulting in reduced bioavailability. In this study, layered double hydroxides (LDHs) intercalated with non-steroidal anti-inflammatory drugs (NSAIDs) were combined with carboxymethylcellulose (CMC) to adjust rheological properties and achieve sustained release and mucoadhesive properties. The obtained LDH-NSAID particles showed high drug loading, aggregation, and positive surface charges. When combined with CMC, the dispersions exhibited viscous, shear-thinning behavior, with ketoprofen leading to more gel-like behavior. The CMC/LDH-NSAID dispersions demonstrated more sustained drug release compared to pure drug formulations, mainly determined by the anion exchange of the intercalated NSAID. Additionally, the formulations showed strong mucoadhesive capacity. Further optimization of particle size and surface properties is needed to modulate the release profile and enhance the rheological properties of these CMC/LDH-based formulations.

Summary: Two series of twenty benzothiazole amide derivatives were synthesized and screened for their anti-inflammatory activity and gastric ulcerogenicity. Compounds 3h and 6a showed the most potent anti-inflammatory activity with high gastrointestinal tolerability. In vitro selectivity testing demonstrated that compound 6a had the best selectivity index among the investigated compounds.

The agent, Gd-DO3A-fen, showed higher and longer-lasting contrast-to-noise ratio (CNR) in vivo compared to the control group, Gd-DO3A-BT. In addition, Gd-DO3A-fen demonstrated a stronger binding affinity for the COX-2 enzyme and exhibited anti-inflammatory activity in two animal models.

In this study, self-assembling nanoparticles made from gelatin-epigallocatechin gallate (EGCG) and surface-decorated with hyaluronic acid (HA) were successfully used for drug delivery to the retinal/choroidal area. The nanoparticles, characterized by a positive surface and HA decoration, showed high drug delivery efficiency to the posterior eye through eye drops and subconjunctival injection.
