Identification and Validation of a Prognostic Signature for Prostate Cancer Based on Ferroptosis-Related Genes

This study investigated ferroptosis related genes in prostate cancer and identified seven FRGs to establish a risk model. The high-risk group was associated with various clinical characteristics, and overexpression of TFRC gene was found to be related to PCa cell proliferation and invasion.
Ferroptosis, an iron-dependent form of selective cell death, is involved in the development of many cancers. However, ferroptosis related genes (FRGs) in prostate cancer (PCa) are not been well studied. In this study, we collected the mRNA expression profiles and clinical information of PCa patients from TCGA and MSKCC databases. The univariate, LASSO, and multivariate Cox regression analyses were performed to construct a prognostic signature. Seven FRGs, AKR1C3, ALOXE3, ATP5MC3, CARS1, MT1G, PTGS2, and TFRC, were included to establish a risk model, which was validated in the MSKCC dataset. The results showed that the high-risk group was apparently correlated with copy number alteration load, tumor burden mutation, immune cell infiltration, mRNAsi, immunotherapy, and bicalutamide response. Moreover, we found that TFRC overexpression induced the proliferation and invasion of PCa cell lines in vitro. These results demonstrate that this risk model can accurately predict prognosis, suggesting that FRGs are promising prognostic biomarkers and potential drug targets in PCa patients.

To address this issue, we propose a novel LDP mechanism that splits and perturbs input numerical data using digital bits, without requiring a fixed input range and large data volume. Our adaptive privacy budget allocation model significantly reduces the deviation of the perturbation function and provides high data utility while maintaining privacy.

Biomechanical analysis of vertebral wedge deformity in elderly women with quantitative CT-based finite element analysis

Summary: The study explored the VD angle of L1 in elderly women and its impact on vertebral stiffness using QCT-FEA. Results showed a slight increase in VD angle with age, moderate negative correlation between age and VS, and no correlation between VD angle and vBMD. Vertebral stiffness gradually decreased with higher VD angle, and fracture risk increased when load was perpendicular to the lower edge of the vertebral body.

Quantitative assessment of zonal trabecular volumetric bone mineral density in middle-aged and elderly women using quantitative computed tomography

Summary: The study aims to explore the feasibility of the 9 zonal trabecular vBMD method for evaluating the first lumbar vertebral body (L1) in women aged 50-80 years and assess the distribution of trabecular vBMD in different zones of L1. The results showed no significant differences in vBMDs among the 9 zones and a negative correlation between age and global vBMD as well as the 9 zonal vBMDs. In terms of vBMD, the posterior region had higher density than the anterior region, and the foot and head regions had higher density than the middle region.

Monitoring early responses to neoadjuvant chemotherapy and the factors affecting neoadjuvant chemotherapy responses in primary osteosarcoma

Summary: This study aimed to predict early responses to neoadjuvant chemotherapy (NACT) in patients with primary conventional osteosarcoma (COS) using the apparent diffusion coefficient (ADC) and evaluate factors affecting the tumor necrosis rate (TNR). Data from 41 patients who underwent magnetic resonance imaging (MRI) and diffusion-weighted imaging scans were prospectively collected. The results showed that ADC2, ADC3, ADC3-1, ADC3-2, and alkaline phosphatase (ALP) before NACT were significantly higher in the good-response group than in the poor-response group. ADC2, ADC3, and ADC3-1 showed good diagnostic performance. However, multivariate analysis showed no significant correlation between these parameters and TNR.

Comparison of single-postlabeling delay and seven-delay three-dimensional pseudo-continuous arterial spin labeling in the assessment of intracranial atherosclerotic disease

Rap1 regulates lumen continuity via Afadin in renal epithelia

Summary: The presence of a continuous lumen in renal tubules is essential for their proper function. In this study, we found that the protein Afadin, which binds to Factin, is required for lumen formation and continuity in renal tubules derived from the nephrogenic mesenchyme in mice. We also discovered that the small GTPase Rap1 plays a crucial role in this process, as its absence leads to severe defects in lumen formation. Furthermore, we observed that Rap1 is necessary for the correct localization of Afadin to adherens junctions. These findings suggest a model in which Rap1 localizes Afadin to junctional complexes, thereby regulating lumen formation and positioning.

Macrophage autophagy contributes to immune liver injury in trichloroethylene sensitized mice: Critical role of TNF-α mediating mTOR pathway

Summary: Trichloroethylene (TCE) induces occupational medicamentosa-like dermatitis due to TCE (OMDT) with immune liver injury, and TNF-α plays an important role in macrophage polarization and liver injury. This study aimed to investigate the regulatory effects of TNF-α on macrophage polarization and autophagy in TCE-induced liver injury. The results showed that TNF-α/TNFR1 promoted M1 polarization of macrophages and suppressed macrophage autophagy via the mTOR pathway.
