INO80 governs superenhancer-mediated oncogenic transcription and tumor growth in melanoma

Superenhancers (SEs) are large genomic regions with a high density of enhancer marks. In cancer, SEs are found near oncogenes and dictate cancer gene expression. However, how oncogenic SEs are regulated remains poorly understood. Here, we show that INO80, achromatin remodeling complex, is required for SE-mediated oncogenic transcription and tumor growth in melanoma. The expression of Ino80, the SWI/SNFATPase, is elevated in melanoma cells and patient melanomas compared with normal melanocytes and benign nevi. Furthermore, Ino80 silencing selectively inhibits melanoma cell proliferation, anchorage-independent growth, tumorigenesis, and tumor maintenance in mouse xenografts. Mechanistically, Ino80 occupies >90% of SEs, and its occupancy is dependent on transcription factors such as MITF and Sox9. Ino80 binding reduces nucleosome occupancy and facilitates Mediator recruitment, thus promoting oncogenic transcription. Consistently, genes co-occupied by Ino80 and Med1 are selectively expressed in melanomas compared with melanocytes. Together, our results reveal an essential role of INO80-dependent chromatin remodeling in SE function and suggest a novel strategy for disrupting SEs in cancer treatment.

A comprehensive review of calcium and ferrous ions chelating peptides: Preparation, structure and transport pathways

Summary: Calcium and iron are essential for human health, but deficiencies in these minerals pose public health risks worldwide. Existing mineral supplements have limitations in terms of solubility, bioavailability, and gastrointestinal irritation. Mineral chelating peptides, as a new type of supplement, have gained attention due to their stability, absorptivity, and safety. Most calcium and iron chelating peptides are isolated from by-products of food processing. Enzymatic hydrolysis combined with affinity chromatography and gel filtration are common methods to obtain peptides with high affinity for calcium and iron. Peptides with small molecular weight and specific functional groups, such as carboxyl and amino groups, can effectively chelate metallic ions. Various chelating modes, including unidentate, bidentate, tridentate, bridging, and alpha mode, have been identified. The stability of peptide-mineral complexes in the gastrointestinal tract and potential transport pathways have also been summarized.

Post-discharge care needs of the older people with hospital-associated disability: A longitudinal study

Summary: This study investigated the change and factors associated with care needs in older people with hospital-associated disability within 6 months post-discharge, and explored the relationship between time-varying care needs and physical function. The study found that the care needs declined unevenly, with daily care and rehabilitation needs being consistently important. Socio-demographic factors and physical function had different effects on care needs at different time points, with physical function being the main factor. These findings are useful for healthcare professionals and caregivers in providing accurate care.

Efficacy and safety of chimeric antigen receptor T-cells treatment in central nervous system lymphoma: a PRISMA-compliant single-arm meta-analysis

Summary: This meta-analysis examines the effectiveness and safety of CAR-T cell therapy in the treatment of central nervous system lymphoma (CNSL). Long-term follow-up data with large sample sizes are needed to further evaluate the effectiveness and safety of CAR-T cell therapy.

Summary: This study examines how reward type, reward characteristic, and product involvement affect the design of reward programs. The findings suggest that reward type is an important determinant of customers' preference towards reward programs. Plural rewards are preferred over single rewards, particularly when the level of product involvement is low. Additionally, the characteristic of the reward interacts with the reward type, positively influencing customer preference towards reward programs.

Summary: This article reports the first direct measurement of the absolute branching fraction of Sigma(+) -> Lambda e(+)nu(e). The measurement is based on an e(+) e(-) annihilation sample collected with the BESIII detector at root s = 3.097 GeV. The branching fraction is determined to be B(Sigma(+)-> Lambda e(+)nu(e))=2.93 +/- 0.74 (stat) +/- 0.13(syst)] x 10(-5), making it the most precise measurement obtained in a single experiment to date and the first result obtained at a collider experiment. The ratio Gamma (Sigma(-) ->Lambda e(-) (nu) over bar (e) /Sigma(+) -> Lambda e(+) (nu) over bar (e) is found to be 1.06 +/- 0.28, within 1.8 standard deviations of the expected value in the absence of second-class currents.

MCANet: Multiscale Cross-Modality Attention Network for Multispectral Pedestrian Detection

Summary: This paper introduces a Multiscale Cross-Modality Attention (MCA) module for multispectral pedestrian detection that efficiently extracts and fuses features. The transformer architecture is used to extract features of two modalities and a novel spatial attention mechanism is designed to enhance object details and suppress background. Extensive experimental results demonstrate that MCANet achieves state-of-the-art detection accuracy on the challenging KAIST multispectral pedestrian dataset.

Summary: This paper proposes a bidirectional narrow pulse modulation of semiconductor optical amplifier (SOA) to enhance the extinction ratio of optical pulse. By reflecting the optical pulse back to the SOA for the second modulation, the bidirectional modulation achieves extra gain and weakens the leakage light intensity.
