Inhibiting SARS-CoV-2 infection in vitro by suppressing its receptor, angiotensin-converting enzyme 2, via aryl-hydrocarbon receptor signal

Understanding the molecular mechanisms of SARS-CoV-2 infection is crucial for developing effective therapies against COVID-19. This study found that cigarette smoke extract, known to be harmful, surprisingly suppressed expression of ACE2 in HepG2 cells. Treatment with AHR agonists decreased ACE2 expression via AHR activation, resulting in the suppression of SARS-CoV-2 infection.
Since understanding molecular mechanisms of SARS-CoV-2 infection is extremely important for developing effective therapies against COVID-19, we focused on the internalization mechanism of SARS-CoV-2 via ACE2. Although cigarette smoke is generally believed to be harmful to the pathogenesis of COVID-19, cigarette smoke extract (CSE) treatments were surprisingly found to suppress the expression of ACE2 in HepG2 cells. We thus tried to clarify the mechanism of CSE effects on expression of ACE2 in mammalian cells. Because RNA-seq analysis suggested that suppressive effects on ACE2 might be inversely correlated with induction of the genes regulated by aryl hydrocarbon receptor (AHR), the AHR agonists 6-formylindolo(3,2-b)carbazole (FICZ) and omeprazole (OMP) were tested to assess whether those treatments affected ACE2 expression. Both FICZ and OMP clearly suppressed ACE2 expression in a dose-dependent manner along with inducing CYP1A1. Knock-down experiments indicated a reduction of ACE2 by FICZ treatment in an AHR-dependent manner. Finally, treatments of AHR agonists inhibited SARS-CoV-2 infection into Vero E6 cells as determined with immunoblotting analyses detecting SARS-CoV-2 specific nucleocapsid protein. We here demonstrate that treatment with AHR agonists, including FICZ, and OMP, decreases expression of ACE2 via AHR activation, resulting in suppression of SARS-CoV-2 infection in mammalian cells.

Meta-Analysis of Transcriptomes in Insects Showing Density-Dependent Polyphenism

Summary: Population density can induce changes in insect morphology, physiology, and behavior, a phenomenon known as density-dependent plasticity. Aphids and locusts are classic examples of insects exhibiting density-dependent plasticity. This study conducted a meta-analysis of public transcriptomes to gain insights into the molecular mechanisms behind this phenomenon. The analysis revealed that processes such as DNA replication, DNA metabolic processes, and mitotic cell cycle were enriched in response to crowded conditions. These findings provide important understanding of the regulatory mechanisms of density-dependent plasticity and highlight the significance of oxidative stress response and neurological system modifications under isolated conditions.

Meta-Analysis of the Public RNA-Seq Data of the Western Honeybee Apis mellifera to Construct Reference Transcriptome Data

Exploratory meta-analysis of hypoxic transcriptomes using a precise transcript reference sequence set

Summary: Gene expression studies can be biased due to external information, but this can be overcome using data-driven analysis. The FANTOM CAGE-Associated Transcriptome project provides a comprehensive meta-assembly of the human transcriptome and was used to evaluate the differential response of various transcripts to hypoxia. The study revealed down-regulation of genes associated with ncRNA metabolism in response to hypoxia.

Genome Sequence of the Edible Green Alga Ulva prolifera, Originating from the Yoshinogawa River in Japan

Summary: We report the genome sequence of Ulva prolifera, a type of seaweed, which was isolated from the Yoshinogawa River in Japan. The genome assembly size is 103.8 Mbp, obtained using Oxford Nanopore Technologies MinION and Illumina sequencing reads, consisting of 142 scaffolds with an N-50 value of 4.11 Mbp.

A highly contiguous genome assembly of red perilla (Perilla frutescens) domesticated in Japan

Summary: Perilla frutescens is an important herbal plant with perillaldehyde and rosmarinic acid as its major bioactive compounds. By using genome editing technology, the production of valuable metabolites can be enhanced, improving the medicinal and nutritional uses of this plant. A high-quality genome assembly of red perilla domesticated in Japan was generated, which can serve as a valuable reference for selecting target genes in genome editing.

Ribosomal Stress Couples with the Hypoxia Response in Dec1-Dependent Orthodontic Tooth Movement

Summary: This study investigated the impact of a deficiency of the Dec1 gene on orthodontic tooth movement (OTM) and explored the role of ribosomal proteins in the hypoxic environment during OTM. Results showed that HIF-1 alpha transgenic mice had higher expression levels of Dec1, HSP105, and ribosomal proteins. Knockout of Dec1 resulted in slower OTM and reduced expression of ribosomal proteins. These findings suggest that the hypoxic environment in OTM plays a crucial role in the functions of Dec1 and ribosomal proteins in maintaining periodontal tissue homeostasis.

Reference Genome Sequences of the Oriental Armyworm, Mythimna separata (Lepidoptera: Noctuidae)

Summary: In this study, a reference genome of the oriental armyworm was constructed, and the quality of the genome was found to be sufficient for use as a reference. The number of predicted genes and coding sites were also determined. Through functional gene annotation, important genes such as the Toll and Immune Deficiency pathways and C-type lectins were identified. These research findings will contribute to molecular and genome-level research on the oriental armyworm and lepidopteran insects.

Inflammatory Cytokine-Induced HIF-1 Activation Promotes Epithelial-Mesenchymal Transition in Endometrial Epithelial Cells

Summary: The endometrium undergoes repeated changes during the menstrual cycle, and chronic endometritis can lead to implantation failure and miscarriage. In this study, the effects of inflammatory cytokines, hypoxia-inducible factor (HIF), and inflammation on endometrial epithelial cells were investigated. The results showed that pro-inflammatory factors and hypoxia activated HIF-1 and promoted epithelial-mesenchymal transition (EMT) in endometrial epithelial cells.

Meta-Analysis of Public RNA Sequencing Data Revealed Potential Key Genes Associated with Reproductive Division of Labor in Social Hymenoptera and Termites

Summary: Eusociality in insects, characterized by reproductive division of labor, has evolved independently many times. Despite transcriptome analyses of queens and workers in various eusocial species, the genes that regulate the reproductive division of labor across multiple eusocial species remain elusive. A meta-analysis of publicly available RNA-sequencing data from four major groups of social insects identified 20 differentially expressed genes in queens or workers, of which 12 genes are newly discovered to be involved in reproductive division of labor. Functional annotation reveals that these genes may regulate behaviors and physiological states associated with reproductive division of labor. These 20 genes, uncovered through extensive datasets of numerous eusocial insects, may serve as key regulators of reproductive division of labor.
