Systematic analysis of key miRNAs and related signaling pathways in colorectal tumorigenesis

The development of colorectal cancers (CRC) is accompanied with the acquisition and maintenance of specific genomic alterations. These alterations can emerge in premalignant adenomas and faithfully maintained in highly advanced tumors. miRNAs are a class of small non-coding RNAs that are frequently deregulated in human cancers and negatively regulate a wide variety of protein coding genes. To identify the sequential alterations of miRNAs and its regulatory networks during CRC development and progression, we detected the miRNA expression profiles of tissue samples from normal colon, colorectal adenoma and CRC using miRNA microarray. qRT-PCR assay was used to validate and select the miRNAs with differential expression among the three groups, and the computer-aided algorithms of TargetScan, miRanda, miRwalk, RNAhybrid and PicTar were used to search for the possible targets of the selected 8 miRNAs (miR-18a, miR-18b, miR-31, miR-142-5p, miR-145, miR-212, miR-451, and miR-638) with continuous alterated expression. These potential target genes were enriched in several key signal transduction pathways (KEGG pathway analysis), which have been proved to be closely related to colorectal tumorigenesis. To confirm the reliability of the analyses, we identified that the metastasis-related gene ZO-1 is a certain target of miR-212 in CRC and keeps declining during CRC progression. By following these analyses, we might gain an in-depth understanding of the molecular regulatory networks of colorectal tumorigenesis and provide new potential targets for the diagnostic and therapeutic interventions of this disease. Colorectal Cancer-Derived Small Extracellular Vesicles Promote Tumor Immune Evasion by Upregulating PD-L1 Expression in Tumor-Associated Macrophages

Summary: This study reveals that colorectal cancer cells can induce macrophage polarization and PD-L1 expression through the release of small extracellular vesicles (sEVs), leading to suppressed CD8(+) T cell activity and increased tumor growth. The miR-21-5p and miR-200a derived from sEVs play key regulatory roles in this process. Inhibiting the secretion of specific sEV-miRNAs from CRC and targeting PD-L1 in TAMs may serve as novel methods for CRC treatment.

GLTSCR1 coordinates alternative splicing and transcription elongation of ZO1 to regulate colorectal cancer progression

Summary: This study evaluated the HER2 status in a large cohort of Chinese colorectal cancer patients and found a positive correlation between HER2 expression and female gender. Furthermore, HER2 protein expression was significantly negatively correlated with RAS/BRAF mutations according to the HERACLES criteria.

Summary: In this study, we developed a dual-targeting non-IgG format of bispecific antibodies (BsAbs) that can simultaneously target EGFR and HER2 on tumor cells. The BsAbs retained dual-targeting activity and exerted potent anticancer effects through the engagement of endogenous antibodies. The optimized BsAbs also showed improved pharmacokinetics and efficient inhibitory effects against tumor growth. This research provides a general and cost-effective platform for generating new BsAbs for cancer immunotherapy.

Up-regulation of STRA6 predicts poor prognosis and contributes to oxaliplatin resistance in colorectal cancer

Summary: This study found that the signaling receptor and transporter STRA6 is highly up-regulated in colorectal cancer (CRC) and promotes CRC cell proliferation. It was also discovered that STRA6 suppresses apoptosis by controlling BCL2 expression, leading to resistance to LOHP treatment in CRC. These findings demonstrate that STRA6 is a potential prognostic factor and oncogene in CRC by promoting tumor growth and chemoresistance.

Brain-Targeting Emodin Mitigates Ischemic Stroke via Inhibiting AQP4-Mediated Swelling and Neuroinflammation

Summary: In this study, emodin was encapsulated in a PEG/cRGD-modified liposome to achieve brain-targeted delivery and maximize therapeutic efficacy. Furthermore, the critical target of emodin responsible for alleviating ischemic stroke was identified, providing a potential drug delivery strategy for ischemic stroke and other brain injuries.

ELOA promotes tumor growth and metastasis by activating RBP1 in gastric cancer

Summary: In this study, we found that ELOA is upregulated in gastric cancer and is associated with malignant characteristics and poor prognosis. Functional experiments showed that ELOA promotes gastric cancer proliferation and metastasis. SIRT5-mediated ME2 desuccinylation promotes cancer growth by enhancing mitochondrial respiration

Summary: In this study, researchers found that ME2 is highly expressed in human colorectal cancer (CRC) tissues and its knockdown inhibits CRC cell proliferation. They also discovered that SIRT5 can desuccinylate ME2, activating its enzymatic activity. Activated ME2 enhances mitochondrial respiration, supporting cell proliferation and tumor development. Additionally, the levels of succinylated ME2 and SIRT5 are associated with patient prognosis.

Comparative alternative polyadenylation profiles in differentiated adipocytes of subcutaneous and intramuscular fat tissue in cattle

Genome-wide regulation of Pol II, FACT, and Spt6 occupancies by RSC in Saccharomyces cerevisiae

Summary: RSC is an essential ATP-dependent chromatin remodeling complex in Saccharomyces cerevisiae. It regulates nucleosome-depleted regions (NDRs) by sliding flanking nucleosomes away from NDRs. Depletion of RSC leads to nucleosome encroachment in NDRs and transcription initiation defects. The study compared the effects of catalytic-dead Sth1 and rapid depletion of Sth1 on transcription. Rapid depletion of Sth1 reduces recruitment of TBP and Pol II, while the catalytic-dead mutant exhibits a severe reduction in TBP binding but accumulates Pol II in coding regions. The results suggest a role for RSC in transcription elongation and termination processes.

Mapping the genetic architecture of idiopathic pulmonary fibrosis: Meta-analysis and epidemiological evidence of case-control studies

Age-different BMSCs-derived exosomes accelerate tendon-bone interface healing in rotator cuff tears model

The exosomes from young BMSCs have better effects compared to those from aged BMSCs.
