Vanillic acid Zn(II) complex: a novel complex with antihyperglycaemic and anti-oxidative activity

The study synthesized a new Zn-vanillic acid complex with enhanced antioxidative and antihyperglycaemic properties compared to vanillic acid alone. The complex demonstrated significant bioactivity in cell-based and ex vivo models.
Objectives Our aim was to synthesize, characterize and evaluate the antihyperglycaemic and antioxidative properties of a new Zn(II) complex of vanillic acid. Methods The complex was synthesized using ZnSO4 center dot 7H(2)O and vanillic acid as precursors. NMR and FTIR techniques were used to characterize the synthesized complex. The cytotoxicity of the complex was measured.The antihyperglycemic and anti-oxidative properties of the complex were evaluated using in vitro, cell-based and ex vivo models and compared with those of its precursors. Key findings Zn(II) coordinated with vanillic acid via a Zn(O-6) coordination, with the complex having three moieties of vanillic acid. The radical scavenging, Fe3+ reducing and hepatic antilipid peroxidative activity of the complex were, respectively, 2.3-, 1.8- and 9.7-folds more potent than vanillic acid. Complexation increased the ci-glucosidase and glycation inhibitory activity of vanillic acid by 3- and 2.6-folds, respectively. Zn(II) conferred potent L-6 myotube (EC50 = 20.4 mu m) and muscle tissue (EC50 = 612 mu m) glucose uptake effects on vanillic acid. Cytotoxicity evaluation showed that the complex did not reduce the viability of L-6 myotubes and Chang liver cells. Conclusions The data suggest that Zn(II)-vanillic acid complex had improved bioactivity relative to vanillic acid.Thus, Zn(II) may be further studied as an antihyperglycaemic and anti-oxidative adjuvant for bioactive phenolic acids.

Bioactive synergism between zinc mineral and p-coumaric acid: A multi-mode glycemic control and antioxidative study

The complex exhibited strong inhibitory effects on lipid peroxidation and GSH depletion, and enhanced glucose uptake and hexokinase activity. Effect of solvent extraction on the antioxidant and phytochemical profiles of ellagitannins from wonderful pomegranate peel: an advanced chemometrics analysis

Summary: Ellagitannins are the main bioactive tannins in pomegranate peel. This study used advanced chemometrics models to determine the solvent extraction (methanol, ethanol, acetone, water) that yields the highest antioxidant and ellagitannins profiles. Acetone extraction had the highest total phenol and flavonoid contents, while methanol extraction had the strongest radical scavenging activity. The models adequately characterized and predicted the variation and patterns in the antioxidant and ellagitannins datasets.

Modeling the Influence of Extraction Temperature on the Ellagitannin and Antioxidant Profiles of Wonderful Pomegranate Peel Using Advanced Chemometrics Analysis

Summary: This study used advanced chemometrics models to determine the influence of different extraction temperatures on the antioxidant profile and ellagitannins recovery of pomegranate peel extract. Extraction at 78 degrees Celsius showed the highest antioxidant activity and ellagitannins content. Pearson's bivariate correlation showed a strong positive correlation between ellagitannins and the anti-lipid peroxidative profile of the extract. The PCA and OPLS models adequately characterized and predicted the variation and patterns in the antioxidant and phytochemical datasets. OPLS biplot showed a distinct clustering pattern around 78 degrees Celsius extraction temperature, indicating the influence of ellagitannins on the antioxidant profile at this temperature.

Therapeutic Antidiabetic and Antioxidative Synergism of Zn(II)-Syringic Acid Complexation

The complex of zinc(II) and syringic acid reduced polyphagia, polydipsia, and weight loss in diabetic rats. The complex enhanced the anti-hyperglycemic action of zinc(II) and syringic acid, reduced lipid peroxidation, and increased antioxidant enzyme activity.

Betulinic Acid Modulates Redox Imbalance and Dysregulated Metabolisms, While Ameliorating Purinergic and Cholinergic Activities in Iron-Induced Neurotoxicity

Summary: The neuroprotective effect of betulinic acid against oxidative neurotoxicity induced by iron was investigated in isolated rat brains. The induction of oxidative neurotoxicity resulted in suppressed antioxidant levels and increased levels of reactive oxygen species. Treatment with betulinic acid reversed these effects and restored the depleted compounds. Pathway analysis revealed the activation of neuroprotective metabolic pathways upon betulinic acid treatment. Molecular docking showed a strong binding affinity between betulinic acid and ATPase. These findings suggest that betulinic acid confers neuroprotection by restoring redox balance and modulating various metabolic pathways.

Temperature-dependent extraction and chromatographic recovery and characterisation of ellagitannins with potent antioxidant and glycaemic control properties from 'Wonderful' pomegranate peel

Summary: This study optimized the extraction of bioactive ellagitannins and punicalagin from pomegranate peel using temperature-dependent extraction. The extract obtained at 78 degrees C had the highest content of phenols, flavonoids, tannins, and punicalagin, showing the strongest antioxidant and glycaemic control properties. The purified tannin from the extract at 78 degrees C exhibited better anti-lipid peroxidation and glucose modulatory effects compared to the crude extract and purified punicalagin.
