Summary: The hypusinated form of EIF5A was found in human myometrium, leiomyoma, and leiomyosarcoma, with increasing levels in neoplastic tissues. Inhibition of EIF5A hypusination reduced cell proliferation and fibronectin expression in myometrium, leiomyoma, and leiomyosarcoma cell lines. These findings suggest that EIF5A may play a role in the pathogenesis of myometrial benign and malignant pathologies.

Summary: This study explored the impact of fibrillar collagens on uterine leiomyosarcoma (uLMS) and found that uLMS tumors have low collagen density and increased expression of collagen-remodeling genes compared to uterine leiomyomas (LM).The overexpression of matrix metalloproteinase-14 (MMP14) in uLMS was found to support uLMS cell proliferation, and uLMS cells were found to be less sensitive to changes in collagen substrate stiffness. Additionally, enhanced basal yes-associated protein 1 (YAP) activity was observed in uLMS cells, which sustains their growth in low-stiffness substrates. These findings suggest that matrix remodeling and YAP may be potential therapeutic targets for uLMS.

Summary: In this meta-analysis, we examined the quantity and dose of opioids dispensed during the perioperative period for benign hysterectomy, as well as the predictors of persistent opioid use after the procedure. A total of 8 studies involving 377,569 women were included. The findings showed that 83% of women were dispensed opioids during the perioperative period, and the rate of persistent opioid use after benign hysterectomy was 5%.

Summary: The aim of this study was to determine the cellular and molecular mechanisms by which adipocytes contribute to the initiation and promotion of uterine leiomyoma tumors. The results showed that adipocytes promote inflammation, fibrosis, and angiogenesis in leiomyoma cells by upregulating associated factors and transferring free fatty acids. Adipocyte-leiomyoma cell interaction increases the phosphorylation of NF-.B and enhances cell proliferation.

Patient, perioperative, and hospital characteristics were included as predictor variables in a multiple linear regression model. The results showed that perioperative and hospital factors significantly influenced the total hospital charges for benign hysterectomy.

Summary: This retrospective cohort study aimed to evaluate the association between surgical start time and clinical and financial outcomes of hysterectomies for benign indications. The study found that an afternoon start time was associated with increased blood loss, longer length of stay, and decreased likelihood of same-day discharge. However, there was no significant difference in the rates of adverse perioperative events between the morning and afternoon groups. Interestingly, an afternoon start time was associated with decreased total hospital charges. Awareness of the relationship between surgical start time and outcomes can guide surgical scheduling and optimize same-day discharge.

Summary: This study investigated the effects of adipocyte coculture and leptin treatment on human myometrium and leiomyoma cells. The results showed that coculture and leptin treatment increased the expression of JAK2/STAT3, MAPK/ERK, and PI3k/AKT signaling pathways, as well as promoting the upregulation of tumor-related inflammatory, fibrotic, and angiogenic factors, and increasing leiomyoma cell growth. These findings suggest that adipocytes or their secreted hormones may contribute to leiomyoma development and growth by activating leptin receptor signaling pathways.

Summary: In this prospective cohort study, predictors of patient satisfaction with pain control, including opioid prescribing practices, were investigated. The analysis showed that patient satisfaction with pain control was primarily influenced by subjective assessment of shared decision-making with the gynecologist, rather than opioid prescription status. These findings suggest the need for a larger cohort to further explore the impact of opioid use on pain control satisfaction after minor gynecologic surgery.

It is now known that uterine fibroids have different aspects of their genesis and oxidative stress plays an important role in their development. The influence of oxidative stress on fibroid development is mediated through genetic, epigenetic, and profibrotic mechanisms. This knowledge has led to clinical implications for diagnosis and treatment using biomarkers and antioxidants.

Summary: This study aims to determine how adipocytes and adipocyte-secreted factors affect changes in myometrial stem cells (MSCs) in a manner that promotes the growth of uterine leiomyomas. The results showed that co-culture of MSCs with adipocytes increased proliferation and extracellular matrix (ECM) deposition, and adipocyte-secreted factors induced oxidative damage and DNA damage in MSCs. These findings suggest that obesity may mediate the initiation of tumorigenesis in MSCs, leading to the development of leiomyomas.

Summary: The objective of this study was to assess the influence of the test-to-delivery interval on the performance of ultrasound and MRI in predicting birthweight. The results showed that MRI performed best in predicting birthweight greater than gestational age if delivery occurred within two weeks of the examination, with a slight decrease thereafter, while ultrasound performance decreased drastically over time.
