Simultaneous determination of dorzolamide and timolol by first-order derivative UV spectroscopy in simulated biological fluid for in vitro drug release testing

Dorzolamide hydrochloride and timolol maleate is a well-established fixed-dose combination for the treatment of glaucoma worldwide. The utilization of simulated biological fluids can give a superior understanding of the release mechanisms and practicable in vivo nature of a dosage form that can improve the predictive potential of in vitro drug release testing.
Dorzolamide hydrochloride and timolol maleate is a well-established fixed-dose combination for the treatment of glaucoma worldwide. The utilization of simulated biological fluids can give a superior understanding of the release mechanisms and practicable in vivo nature of a dosage form that can improve the predictive potential of in vitro drug release testing. No method has been reported so far for the simultaneous estimation of dorzolamide and timolol in simulated tear fluid. In the present study, a simple, precise, and accurate first-order derivative ultraviolet spectrophotometric method for the rou-tine analysis of dorzolamide and timolol in simulated tear fluid is proposed for in vitro drug release test-ing. The developed method was validated as per International Conference on Harmonization guidelines Q2 (R1). First-order derivative UV spectrophotometry was successfully applied to separate the overlap-ping peaks of dorzolamide and timolol by measuring peak amplitude at 251.80 nm and absorbance at 295.00 nm, respectively. The method was found to be accurate and precise, with a recovery range of 98.0 -101.0% and low relative standard deviations (<2.0%). The developed method was successfully applied for in vitro drug release testing of in-house in situ gel and marketed eye drops containing dorzo-lamide and timolol. Various mathematical models were adopted to fit the in vitro drug release profile. It was observed that the drug release of both drugs from the in situ gel and the marketed solution were best fitted by the Higuchi and first-order kinetic models, respectively. Moreover, the analysis of variance (ANOVA) provision was used for the validation of results. Overall, with the advantages of simple and fast operations, as well as reliability, the proposed method offers an ideal platform for routine analysis as compared to other high-cost and time-consuming chromatographic techniques. Having access to such a robust method will encourage the use of simulated tear fluid for in vitro drug release testing of ocular products and help to predict the in vivo performances of ophthalmic preparations. Formulation performance window for manufacturing cellulose-based sustained-release mini-matrices of highly water-soluble drug via hot-melt extrusion technology

Summary: The aim of this study was to develop sustained-release mini-matrices of tramadol hydrochloride using hot-melt extrusion (HME) process. By using the Design of Experiment (DoE) method, a feasible formulation window with the desired quality target product profile was successfully developed by determining the ratios of hydrophobic and hydrophilic components, as well as the processing temperature. FTIR chemical imaging, SEM analysis, and FTIR results confirmed the homogeneous drug distribution and drug-matrix interaction. Hydroalcoholic dissolution studies demonstrated safe and sustained-release of tramadol. The developed mini-matrices followed the Peppas-Sahlin model, indicating a combination of Fickian diffusion and swelling mechanisms.

Summary: Flavonoids, a major group of polyphenolic compounds, have shown significant potential in inhibiting tumor proliferation, cell cycle, angiogenesis, migration, and invasion in lung cancer. However, their clinical application is limited due to poor solubility, low permeability, and quick blood clearance. Nanoengineered systems such as liposomes, nanoparticles, micelles, dendrimers, and nanotubes can greatly enhance the targeted action of flavonoids with improved efficacy and pharmacokinetic properties, making them suitable for treating lung cancer. This review highlights the therapeutic potential of flavonoids and the cutting-edge progress in nanoengineered systems incorporating flavonoids for lung cancer treatment.

Summary: Lung cancer is a major cause of cancer-related deaths worldwide, and non-small-cell lung cancer is the most common type. Nanotechnology-based drug-delivery systems show great potential in lung cancer therapy due to their unique characteristics. However, traditional chemotherapeutics have toxicity issues, so polyphenols from diet could be used as a replacement. Targeted nanotherapeutics based on polyphenols have shown interactions with protein targets and cellular signaling pathways, affecting key cellular processes. This review summarizes the molecular mechanisms and targeted therapeutic potentials of nanoengineered dietary polyphenols in lung cancer management.

Hot-melt extruded in situ gelling systems (MeltDrops Technology): Formulation development, in silico modelling and in vivo studies

Summary: In this study, a novel ophthalmic solution called MeltDrops was successfully developed using hot-melt extrusion technology. MeltDrops showed improved retention time and bioavailability compared to marketed eyedrops. It also exhibited better physical and chemical uniformity and sustained drug release for over 12 hours in vitro. In vivo studies in rabbits confirmed the superior effectiveness and safety of MeltDrops.

Infrared characterization of hydrated products of glyoxal in aqueous solution

Techno-economic assessment of waste mandarin biochar as a green adsorbent for binary dye wastewater effluents of methylene blue and basic fuchsin: Lab- and large-scale investigations

Summary: The recycling of waste mandarin peels into biochar (MRBC) has been used as a high performance and cost-effective adsorbent for treating polluted wastewater effluents. Batch adsorption studies were conducted to analyze the adsorption competency of MRBC on two dyes, methylene blue (MB) and basic fuchsin (BF), either in individual solutions or binary combinations. The results showed that MRBC was capable of effectively removing a high percentage of both dyes. However, the adsorption capacity decreased when the dyes were combined. The cost estimation of MRBC production and wastewater treatment indicated that both were relatively low.

Lead-free double perovskite Cs2NaInCl6 nanocrystals doped with Sb3+and Tb3+for copper ions detection in lubricating oil

Summary: This study synthesized Cs2NaInCl6 nanocrystals co-doped with Sb3+ and Tb3+ ions as probes for copper ions detection in lubricating oil. The introduction of Sb3+ effectively reduced the band gap of the host material and enabled energy transfer pathway for Tb3+ emission. The doped Tb3+ ions resulted in the suppression of emission due to electron transfer. The Cs2NaInCl6: 2.5 %Sb3+, 40 %Tb3+ NCs exhibited superior sensitivity and selectivity for copper ions detection.

Non-invasive analysis of reference glass and historical mosaic tesserae by means of reflectance infrared spectroscopy

Summary: Raman spectroscopy is commonly used for studying glassy materials in cultural heritage, but it is more difficult to interpret the spectra and apply the technique with portable instruments. In contrast, diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS) can be used in archaeometric investigations as it is portable and non-invasive. However, there is limited application of this technique to historical glasses. This exploratory work demonstrates the potential of DRIFTS, in combination with portable X-ray Fluorescence (pXRF) and EDS microanalyses, for studying the composition and alteration of glass samples in cultural heritage.
