ZnAl nano layered double hydroxides for dual functional CRISPR/Cas9 delivery and enhanced green fluorescence protein biosensor

Evaluation of the effect of different parameters for designing a non-viral vector in gene delivery systems has great importance. In this manner, 2D crystals, precisely layered double hydroxides, have attracted the attention of scientists due to their significant adjustability and low-toxicity and low-cost preparation procedure. In this work, the relationship between different physicochemical properties of LDH, including pH, size, zeta potential, and synthesis procedure, was investigated and optimized for CRISPR/Cas9 delivery and reverse fluorescence response to the EGFP. In this manner, ZnAl LDH and ZnAl HMTA LDH were synthesized and characterized and applied in the HEK-293 cell line to deliver CRISPR/Cas9. The results were optimized by different characterizations as well as Gel Electrophoresis and showed acceptable binding ability with the DNA that could be considered as a promising and also new gold-standard for the delivery of CRISPR/Cas9. Also, the relationship of the presence of tertiary amines (in this case, hexamethylenetetramine (HMTA) as the templates) in the structure of the ZnAl LDH, as well as the gene delivery application, was evaluated. The results showed more than 79% of relative cell viability in most of the weight ratios of LDH to CRISPR/Cas9; fully quenching the fluorescence intensity of the EGFP/LDH in the presence of 15 mu g mL(-1) of the protoporphyrins along with the detection limit of below 2.1 mu g mL(-1), the transfection efficiency of around 33% of the GFP positive cell for ZnAl LDH and more than 38% for the ZnAl LDH in the presence of its tertiary amine template.

The challenges and opportunities for translating miRNA-based strategies into clinical applications are also discussed.

A multifunctional bioresponsive and fluorescent active nanogel composite for breast cancer therapy and bioimaging

Summary: Bio-responsive nanocomposites with facile fabrication and rational design are utilized for the diagnosis and treatment of cancer in this study. A traceable and bioresponsive fluorescent active nanogel composite, integrating creatinine-functionalized carbon dots (QD) and lecithin-inulin nanogel, was fabricated for targeted therapy of HER2-positive breast cancer. The nanogel composite exhibited effective cellular uptake, boosted reactive oxygen species production, and regulated expression of apoptosis-related genes. It also enhanced the cellular uptake of herceptin, inhibited cancer cell proliferation, and reduced tumor growth in vivo. This herceptin-adorned PEGylated lecithin-inulin nanogel composite holds potential as a theranostic candidate for HER-2-positive breast cancer.

Summary: In this study, a new 2D-2D nanosheet photocatalyst was developed with NiCuInS2: In2S3/g-C3N4. The heterojunction exhibited significantly improved hydrogen production due to the synergistic effects of Ni-Cu-In trimetallic active sites and the dual S-scheme heterojunction, enabling efficient charge separation and transfer.

Anthocyanins extract as a non-toxic and green fluorescent label for bioimaging of HER2-positive breast cancer cells

Summary: This study presents a green and inexpensive method for extracting natural anthocyanin fluorophore from Red Cabbage and demonstrates its application for fluorescent bioimaging of HER2 positive breast cancer cells. The extracted anthocyanins exhibit proper fluorescence intensity, negligible photobleaching and no cytotoxicity, making them a potential fluorescent probe for biological diagnosis and bio-imaging techniques.

This study analyzed their total and risk-attributable burden across different regions and identified disparities based on Socio-demographic Index (SDI). The findings highlight the substantial impact of tobacco and alcohol use on these types of cancers.

Toward preparation of large scale and uniform mesoporous BiVO4 thin films with enhanced photostability for solar water splitting

Summary: The preparation of high-performance BiVO4 thin films using a straightforward and cost-effective hot spin coating method is described in this work. The optimized parameters resulted in uniform mesoporous films with high adhesion. The method was also capable of preparing large-scale superhydrophilic BiVO4 layers, exhibiting stable photo-current density in a photoelectrochemical cell.
