Summary: This study investigated the predictors of eGFR changes and other safety and efficacy outcomes during long-term TAF therapy in patients with chronic hepatitis B. The results showed that eGFR improved in most switched patients, especially in those with baseline eGFR 30-60 ml/min. Additionally, TAF may also improve low serum phosphate, BMD, and ALT, while maintaining or inducing virological suppression.

Vitamin D-VDR Novel Anti-Inflammatory Molecules-New Insights into Their Effects on Liver Diseases

Ioanna Aggeletopoulou, Konstantinos Thomopoulos, Athanasia Mouzaki, Christos Triantos

Summary: There is strong evidence linking vitamin D deficiency to liver dysfunction and poor prognosis in liver disease. Vitamin D and its receptor play important roles in immune response regulation. Genetic variations may influence the progression of liver disease. The association of the vitamin D-VDR complex with liver dysfunction has great potential for clinical application.

Molecular Mechanisms Underlying IL-33-Mediated Inflammation in Inflammatory Bowel Disease

Ioanna Aggeletopoulou, Efthymios P. P. Tsounis, Christos Triantos

Summary: Interleukin-33 (IL-33) is a pleiotropic cytokine with both extracellular and nuclear functions, and its receptor ST2 plays a critical role in regulating inflammatory disorders. The IL-33/ST2 axis is involved in maintaining intestinal homeostasis and mucosal defenses. This review focuses on the pro- and anti-inflammatory effects of the IL-33/ST2 axis in ulcerative colitis and Crohn's disease, as well as the molecular mechanisms underlying its association with IBD pathogenesis. Understanding the IL-33/ST2-mediated mechanisms in IBD has potential for clinical applications as therapeutic targets.

COVID-19 Vaccination in Patients with Chronic Liver Disease

Georgios Schinas, Eleni Polyzou, Fevronia Mitropetrou, Aristotelis Pazionis, Charalambos Gogos, Christos Triantos, Karolina Akinosoglou

Summary: Vaccination against SARS-CoV-2 is crucial for individuals with Chronic Liver Disease (CLD), as they have a decreased immune response and are vulnerable to severe COVID-19. However, the incidence of liver injury following vaccination is relatively rare. Timely vaccination and booster shots are necessary according to guidelines.

Summary: The rate of underdiagnosis of Hepatitis D virus (HDV) in Greece is still high, with wide variations in screening rates among tertiary liver centres. Factors such as patient age, risk group, ALT levels, and location and size of the liver centres are found to affect the diagnosis of HDV.

Summary: The aim of this study was to validate the content of IBD-Disk in a Greek cohort of IBD patients. The results showed good correlation and consistency between the scores of IBD-Disk and IBD-DI at baseline and follow-up. Female gender and extraintestinal manifestations were significantly associated with a higher IBD-Disk total score. Therefore, the Greek version of IBD-Disk was proven to be a reliable and valid tool for assessing IBD-related disability.

Creeping Fat in Crohn's Disease-Surgical, Histological, and Radiological Approaches

Ioanna Aggeletopoulou, Efthymios P. Tsounis, Athanasia Mouzaki, Christos Triantos

Summary: During the progression of Crohn's disease, mesenteric adipose tissue responds to inflammatory mediators and bacterial invasion, resulting in the formation of creeping fat. This review discusses the features of creeping fat and the histological evidence of its importance. Recent advances in radiological assessment of creeping fat in Crohn's disease are also reported, with improved visualization and quantification of adipose tissue depots. Visceral fat has shown significant correlation with various outcomes in Crohn's disease.

Gene Polymorphisms and Biological Effects of Vitamin D Receptor on Nonalcoholic Fatty Liver Disease Development and Progression

Evanthia Tourkochristou, Athanasia Mouzaki, Christos Triantos

Summary: Nonalcoholic fatty liver disease (NAFLD) is a common chronic liver disease with a rising global prevalence. The vitamin D/vitamin D receptor (VDR) axis plays a significant role in the development and progression of NAFLD. Gene polymorphisms affecting the regulation of the VDR gene are associated with the presence and severity of NAFLD, as they influence adipose tissue activity, fibrosis, and hepatocellular carcinoma (HCC) development. Vitamin D binds to the hepatic VDR, activating transcriptional activity and regulating gene expression related to inflammation and fibrosis, as well as inducing intracellular signal transduction through VDR-mediated activation of Ca2+ channels. Elucidating the genetic and molecular background of VDR in NAFLD pathophysiology can lead to the development of VDR agonists as potential therapeutic targets.

The Influence of Single Nucleotide Polymorphisms on Vitamin D Receptor Protein Levels and Function in Chronic Liver Disease

Evanthia Tourkochristou, Efthymios P. Tsounis, Haralampos Tzoupis, Ioanna Aggeletopoulou, Aggeliki Tsintoni, Theoni Lourida, Georgia Diamantopoulou, Konstantinos Zisimopoulos, Theodora Kafentzi, Anne-Lise de Lastic, Maria Rodi, Theodore Tselios, Konstantinos Thomopoulos, Athanasia Mouzaki, Christos Triantos

Summary: This study investigated the association between single nucleotide polymorphisms (SNPs) in the vitamin D receptor (VDR) gene and chronic liver disease. The expression levels and function of VDR protein were examined in patients with chronic liver disease. The results suggest that VDR SNPs are associated with VDR expression levels and may contribute to the progression of chronic liver disease.
