Cytotoxicity and Anticancer Activity of Donkioporiella mellea on MRC5 (Normal Human Lung) and A549 (Human Lung Carcinoma) Cells Lines

Polypores are mushrooms which are rich in bioactivities and for generations, they have been widely used as herbal remedies. Despite their significant importance in treatments of various health issues, only a few local species have been reported for their pharmacological potentials. The present study was carried out to establish cytotoxicity potentials of Donkioporiella mellea, a local polypore species collected from forested areas in Malaysia at cellular levels on normal human lung (MRC5) and human lung carcinoma (A549) cell lines. Survival and inhibition rates were analyzed by 3-(4, 5)-dimethylthiahiazo (-z-y-l)-2,5-diphenyltetrazoliumbromide (MTT) while monitoring changes on cellular shapes by inverted phase contrast microscopy. Survival rates of MRC5 cells were observed to be significantly higher than A549 after treatments with various concentrations of polypore extracts. MRC5 cells showed excellence in survival performance when treated with hot and cold aqueous extracts. Cold aqueous extract showed higher cytotoxicity activities compared to hot aqueous extract (p<0.0001) with inhibitory concentration (IC50) values of 414.29 mu g/ml and >1000 mu g/ml, respectively. Treatments with tamoxifen as a control exhibited necrotic features in both cell lines. The results suggest that D. mellea possesses pharmacological potentials that can be utilized for human consumption as a new bioresource alternative, thus encouraging research advancement in mycological and nutraceutical studies.

Zero waste management of spent mushroom compost

Summary: Edible mushrooms are commercially grown using lignocellulosic waste through a biological process. After harvesting season, about 70% of the substrate remains as spent mushroom compost (SMC). SMC can be utilized for value-added products, supporting a zero waste approach. The major uses of SMC include in agriculture, as mushroom media, animal feed, plant compost, and fertilizer, with potential for extended usage in second cultivation and renewable energy production.

Summary: The study found that factors such as bed humidity, bed temperature, harvesting week, harvesting month, and replication significantly affected the production of V. volvacea, while bed pH and orientation did not have an impact. Variations in temperature and humidity, especially during incubation, were found to be crucial for mycelial growth and fruiting body development, affecting mushroom production. Further research is needed to explore the potential of cultivating V. volvacea in different tropical agroforestry settings and its ability to contribute to climate change mitigation and circular economy by recycling oil palm wastes.

Ultrasound-Assisted Extraction of Antioxidants from Melastoma malabathricum Linn.: Modeling and Optimization Using Box-Behnken Design

Summary: This study presents modeling and optimization of ultrasound-assisted extraction (UAE) of Melastoma malabathricum to evaluate its phytochemical properties. Through an OFAT procedure and response surface methodology, optimum conditions were established for maximum antioxidant extraction. The results showed that UAE at 37 kHz, X-ET 32 degrees C for X-UT 16 min, X-SC 70% ethanol concentration, and X-SLR 1:10 ratio yielded high scavenging effects on DPPH, as well as significant levels of total phenolic and flavonoid contents. GC-MS analysis confirmed the presence of important phytochemical compounds. This study provides valuable insights for further validation of the medicinal and cosmeceutical uses of Melastoma malabathricum.

Summary: In this study, two entomopathogenic fungal species, Isaria javanica and Purpureocillium lilacinum, were found to act as endophytes in tomato plants, benefiting the plants and causing significant mortality of the whitefly pests. This indicates that they have potential as biocontrol agents and plant growth promoters.

Summary: This article reports the temperature influence on the growth and efficacy of M. anisopliae in Malaysia. The results showed that the isolates of M. anisopliae had growth and virulence capabilities against B. tabaci on vegetables planted in a tropical climate. However, further research is still needed.

Identification and virulence of entomopathogenic fungi, Isariajavanica and Purpureocilliumlilacinum isolated from the whitefly, Bemisiatabaci (Gennadius) (Hemiptera: Aleyrodidae) in Malaysia

Summary: Three entomopathogenic fungi, Isaria javanica (Cjc-03 and Cjw-01) and Purpureocillium lilacinum (TS-01), were discovered infecting whiteflies naturally on chili and tomato plants. The virulence of these isolates against nymphs and adults of Bemisia tabaci was evaluated at different concentrations in laboratory and glasshouse conditions. Isaria javanica showed the highest virulence against both nymphs and adults of B. tabaci.

Effect of feeding Pleurotus pulmonarius-treated empty fruit bunch on nutrient digestibility and milk fatty acid profiles in goats

Summary: This study evaluated the effect of feeding P. pulmonarius-treated empty fruit bunch (FTEFB) on the nutrient intakes, digestibility, milk yield and milk profiles of lactating Saanen goats. The results showed that the replacement of Napier grass with FTEFB did not adversely affect nutrient intake, fiber digestibility, milk yield, milk composition and milk fatty acid profiles in lactating goats.

Impact of Storage Temperatures on the Quality of Oven-Dried Black Jelly Mushroom, Auricularia nigricans (Agaricomycetes)

Summary: The effects of different storage temperatures (4, 25, and 35 degrees C) on the quality and microbial counts of oven-dried black jelly mushroom were investigated. The results showed that a storage temperature of 4 degrees C was the most effective in maintaining the quality and reducing microbial counts.

Efficacy of soil-borne entomopathogenic fungi against subterranean termite, Coptotermes curvignathus Holmgren (Isoptera: Rhinotermitidae)

Summary: This study isolated and identified 11 potential entomopathogenic fungi with insecticidal activity against C. curvignathus. Among them, UPM-A2C-1 M. anisopliae var anisopliae showed the best efficacy and had high virulence score and mycelia formation rate against the subterranean termite.

Summary: This study aimed to identify novel indigenous isolates of entomopathogenic fungi (EPF) in oil palm soil as part of integrated pest management (IPM) of oil palm insect pests. Seven Metarhizium anisopliae and two Purpureocillium lilacinum isolates were identified and could be potentially considered as biological control candidates for major insect pests in oil palm.
