Potential Nutraceutical Benefits of In Vivo Grown Saffron (Crocus sativus L.) As Analgesic, Anti-inflammatory, Anticoagulant, and Antidepressant in Mice

Crocus sativus, a medicinally important herbaceous plant, has been traditionally used to cure coughs, colds, insomnia, cramps, asthma, and pain. Moreover, the therapeutic applications of saffron include its immunomodulatory and anticancer properties. The current experimental analysis was performed to explore the potential nutraceutical efficacy of corm, leaf, petal, and stigma of saffron ethanolic extracts as analgesic, anti-inflammatory, anticoagulant, and antidepressant using hot plate, carrageenan-induced paw edema, capillary tube and forced swim test, respectively in mice. The results indicated that among all the extracts, stigma ethanolic extract (SEE) represented maximum latency activity (72.85%) and edema inhibition (77.33%) followed by petal ethanolic extract (PEE) with latency activity and edema inhibition of 64.06 and 70.50%, respectively. Corm ethanolic extract (CEE) and leaf ethanolic extract (LEE) displayed mild analgesic activity of 22.40% and 29.07%, respectively. Additionally, LEE (53.29%) and CEE (47.47%) exhibited mild to moderate response against inflammation. The coagulation time of SEE (101.66 s) was almost equivalent to the standard drug, aspirin (101.66 s), suggesting a strong anticoagulant effect followed by PEE (86.5 s). LEE (66.83 s) represented moderate inhibitory effect on coagulation activity while CEE (42.83 s) showed neutral effect. Additionally, PEE and SEE also expressed itself as potential antidepressants with immobility time <= 76.66 s, while CEE (96.50 s) and LEE (106.83 s) indicated moderate to mild antidepressant efficacy. Based on the in vivo activities, saffron extract, particularly SEE and PEE, can be used as a potential nutraceutical and therapeutic agent due to its significant pharmacological activities.

Downregulation of PI3K/AKT/mTOR Pathway in Juglone-Treated Bovine Oocytes

Summary: In this study, we investigated the toxicity of juglone and its relationship with PI3K/AKT/mTOR signaling during the in vitro maturation (IVM) of oocytes. Results showed that juglone exposure led to a significant decrease in F-actin accumulation and downregulation of various genes related to oocyte development and meiosis progression. Juglone treatment also resulted in decreased expression of markers associated with glycolysis, ATP synthesis, OXPHOS, and oocyte survival, as well as lower expression of PI3K, AKT, and mTOR. Additionally, juglone-exposed oocytes exhibited increased expression of autophagy markers and DNA damage-specific markers.

Summary: Three bis-acyl-thiourea derivatives were synthesized and characterized, and their DNA binding, anti-urease, and anticancer activities were explored. The results showed that these derivatives exhibited binding interactions with DNA, with UP-1 showing better inhibitory efficiency against urease and higher cytotoxicity against cancerous and healthy cell lines.

Molecular Identification of Endophytic Bacteria from Silybum marianum and Their Effect on Brassica napus Growth under Heavy Metal Stress

Summary: In this study, 11 endophytic bacteria were isolated from Silybum marianum collected from heavy metal polluted sites. Three of these isolates showed multiple beneficial features, including heavy metal resistance, plant growth stimulation, IAA production, heavy metal toxicity remediation, and antibacterial activity. These bacteria were also found to promote the translocation and degradation of heavy metals in Brassica napus plants.

Summary: This study investigated the knowledge, attitudes, practices, and information sources of pregnant women in the Malakand region of Pakistan regarding toxoplasmosis. The findings revealed poor knowledge about toxoplasmosis among the participants, with 31.2% having good knowledge, 39.2% having moderate knowledge, and 29.5% having poor knowledge. The number of children within pregnant multipara women was significantly associated with knowledge about toxoplasmosis.

Lycopene attenuates chlorpyrifos-induced hepatotoxicity in rats via activation of Nrf2/HO-1 axis

Summary: The study evaluated the potential hepatoprotective effects of lycopene (LCP) against chlorpyrifos (CPF)-induced hepatotoxicity in rats. The results showed that LCP protected the liver by inhibiting the increase in serum alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, and lactate dehydrogenase induced by CPF. LCP also prevented liver tissue damage by reducing bile duct proliferation and periductal fibrosis caused by CPF.

Parasite Diversity in a Freshwater Ecosystem

Summary: This study examined the prevalence, diversity, and mean intensity of parasites in 9 freshwater fish species. A total of 26 species of parasites were identified, with the highest diversity found in the Okara site. Labeo rohita had the highest number of parasites, while Notopterus notopterus and Sperata seenghala appeared to be resistant species. The study also found that parasite prevalence increased with the length, size, and age of fish.

These findings suggest that ADP may have the potential to improve dyslipidemia and obesity.
