Vaccination with dendritic cells pulsed ex vivo with gp100 peptide-decorated liposomes enhances the efficacy of anti PD-1 therapy in a mouse model of melanoma

Background: Targeting antigens to dendritic cells (DCs) via nanoparticles is a powerful strategy which improves the efficacy of ex vivo antigen-pulsed DC vaccines. Methods: In this study, liposomes were first decorated with gp100(25-33) self-antigen and then characterized. Then, DCs were pulsed ex vivo with liposomal gp100 and injected subcutaneously in mice bearing B16F10 established melanoma tumors in combination with anti-PD-1 therapy. Results: Treatment with liposomal pulsed DC vaccine elicited the strongest anticancer immunity and enhanced intratumoral immune responses based on infiltration of gp100-specific CD4+ and CD8+ T cells to the tumor leading to significant tumor growth regression and prolonged survival rate. Treatment with liposomal pulsed DC vaccine also markedly enhanced specific cytotoxic T lymphocytes (CTL) responses with a significant higher titer of IFN-gamma in the spleen. Moreover, a significant increase of PD-1 expressing CD8+ tumor infiltrating lymphocytes (TILs) was detected in tumors. Conclusion: Our results demonstrate an optimum dose of liposomal gp100 significantly increases the efficacy of anti-PD-1 therapy in mice and might be an effective strategy to overcome resistance to anti-PD-1 therapy. Summary: Despite the global recommendation of the safe and effective measles vaccine since 1974, there were over 17 million cases of measles in 2017, with the majority occurring in low- and middle-income countries. A study from 2000 to 2019 in 101 LMICs showed that after gains in measles vaccine coverage from 2000 to 2010, there was regression in coverage in over half of the districts from 2010 to 2019, with rural areas having lower coverage than urban areas.

Mesenchymal stem cell (MSC)-derived exosomes as novel vehicles for delivery of miRNAs in cancer therapy

Summary: Mesenchymal stem cells (MSCs) have potential in cancer therapy and can be used as vehicles in cancer gene therapy. MSC-derived exosomes, as novel cell-free alternatives to MSC-based cell therapy, have advantages in safety, stability, and convenience. Unmodified MSC-derived exosomes can promote or inhibit tumors, while modified exosomes can suppress cancer development by delivering therapeutic molecules.

Assessment of the association between TNIP1 polymorphism with clinical features and risk of systemic lupus erythematosus

Summary: This study evaluated the association between TNIP1 gene rs6889239 polymorphism and the risk and clinical characteristics of RA and SLE in the Iranian population. The results showed a significant association between the CC genotype and C allele of rs6889239 with an increased risk of SLE. However, there was no significant association between genotypes and allele frequencies of rs6889239 with the occurrence risk of RA. Stratification analysis also revealed a link between the C allele in rs6889239 and renal involvement in SLE patients and lower age of onset in the RA group.

Cinnamaldehyde Regulates Insulin and Caspase-3 Signaling Pathways in the Sporadic Alzheimer's Disease Model: Involvement of Hippocampal Function via IRS-1, Akt, and GSK-3β Phosphorylation

Summary: This study investigated the regulatory effects of cinnamaldehyde on insulin and caspase-3 signaling pathways in an Alzheimer's disease (AD) rat model. The results demonstrated that cinnamaldehyde could improve memory deficits and anxiety-like behavior, and modulate the IRS-1/AKT/GSK-3β signaling pathway and inhibit Aβ aggregation and caspase-3 cleavage in the hippocampus. These findings suggest that cinnamaldehyde may have therapeutic potential for AD treatment.

Study of frequency and inheritance model of ACE1 I/D and ACE2 rs2285666 polymorphisms in COVID-19 patients with varying severity of lung involvement and its effect on serum cytokines levels

Summary: This study aimed to evaluate the frequency and inheritance of ACE1 I/D and ACE2 rs2285666 polymorphisms in COVID-19 patients and their impact on disease severity and cytokine levels. The results showed no significant association between ACE2 rs2285666 polymorphism genotypes and ACE1 I/D with the severity of lung involvement. However, there was a significant association between ACE1 I/D polymorphism genotypes and IL-6, WBC, and NLR levels.

Summary: Vitamin D has been found to be associated with substance abuse and dependence, potentially through its effect on dopamine signaling pathway. The study suggests that vitamin D can be considered as an effective therapeutic approach for narcotic addiction and substance abuse. These findings have important implications for the treatment of drug addiction.

Silymarin pretreatment protects against ethanol-induced memory impairment: Biochemical and histopathological evidence

Summary: This study investigated the effects of ethanol on memory impairment and the neuroprotective role of silymarin. The results showed that ethanol caused memory deficits and increased oxidative stress and hippocampal apoptosis. However, administration of silymarin alleviated these adverse effects induced by ethanol.

Molecular typing of clinical and environmental isolates of Klebsiella pneumoniae producing ESBLs by PFGE

Summary: This study investigated the molecular typing of extended-spectrum beta-lactamase (ESBL) producing Klebsiella pneumoniae isolates from Beheshti Hospital in Iran. The results showed that there was a common molecular signature among both clinical and environmental K. pneumoniae isolates, indicating a similar genotype and likely a common origin. Therefore, it is important for hospitals to implement effective infection control systems to reduce the spread of ESBL strains.
