Immunogenicity of Anti-SARS-CoV-2 Vaccines in Common Variable Immunodeficiency

Daniel Arroyo-Sanchez, Oscar Cabrera-Marante, Rocio Laguna-Goya, Patricia Almendro-Vazquez, Octavio Carretero, Francisco Javier Gil-Etayo, Patricia Suarez-Fernandez, Pilar Perez-Romero, Edgard Rodriguez de Frias, Antonio Serrano, Luis M. Allende, Daniel Pleguezuelo, Estela Paz-Artal

Summary: Common Variable Immunodeficiency (CVID) patients show slower humoral and cellular responses to COVID-19 vaccination compared to healthy controls. Despite most CVID patients developing S1-specific antibodies and T cells after vaccination, neutralizing antibodies were only detected in half of the patients. Those with absent or discordant responses often had a history of autoimmunity or lymphoproliferation, while recent anti-B cell antibody therapy may also weaken the response. Further studies are necessary to assess the protective efficacy of COVID-19 vaccination in CVID patients, with consideration for a third dose to achieve full immune response.

An Early Th1 Response Is a Key Factor for a Favorable COVID-19 Evolution

Allende, Pablo Morales-Perez, Maria Jose Castro-Panete, Antonio Lalueza, Carlos Lumbreras, Estela Paz-Artal, Antonio Serrano

Summary: The Th1/Th2 balance plays a crucial role in the progression and evolution of infectious diseases. The immune response and T-helper subsets in COVID-19 patients are closely related to the severity of the disease. Asymptomatic individuals show a strong Th1 immune response, while severe patients have a Th17-skewed immune response.

Overcoming CAR-Mediated CD19 Downmodulation and Leukemia Relapse with T Lymphocytes Secreting Anti-CD19 T-cell Engagers

Belen Blanco, Angel Ramirez-Fernandez, Clara Bueno, Lidia Argemi-Muntadas, Patricia Fuentes, Oscar Aguilar-Sopena, Francisco Gutierrez-Aguera, Samanta Romina Zanetti, Antonio Tapia-Galisteo, Laura Diez-Alonso, Alejandro Segura-Tudela, Maria Castella, Berta Marzal, Sergi Betriu, Seandean L. Toribio, Pedro Roda-Navarro, Manel Juan, Pablo Menendez, Luis Alvarez-Vallina

Summary: This study compares the efficacy of CAR-T19 cells and STAb-T19 cells and finds that STAb-T19 cells are more effective in inducing cytotoxicity, avoiding leukemia escape, and preventing relapse. CAR-T19 cells induce leukemia escape through rapid CD19 internalization and degradation, while STAb-T19 cells prevent CD19 downmodulation by forming canonical immunologic synapses. In a long-term patient-derived xenograft mouse model, STAb-T19 cells efficiently eradicate leukemia cells, while leukemia relapse occurs after CAR-T19 therapy.

Synapse topology and downmodulation events determine the functional outcome of anti-CD19 T cell-redirecting strategies

Angel Ramirez-Fernandez, Oscar Aguilar-Sopena, Laura Diez-Alonso, Alejandro Segura-Tudela, Carmen Dominguez-Alonso, Pedro Roda-Navarro, Luis alvarez-Vallina, Belen Blanco

Summary: Cancer immunotherapy strategies based on STAb-T cells may be more effective and safer than CAR-T approaches, as STAb-T cells form artificial immunological synapses that closely resemble physiological synapses and do not cause acute loss of CD19 in target cells.

Tumor targeted 4-1BB agonist antibody-albumin fusions with high affinity to FcRn induce anti-tumor immunity without toxicity

Oana Hangiu, Marta Compte, Anders Dinesen, Rocio Navarro, Antonio Tapia-Galisteo, Ole A. Mandrup, Ainhoa Erce-Llamazares, Rodrigo Lazaro-Gorines, Daniel Nehme-Alvarez, Carmen Dominguez-Alonso, Seandean L. Harwood, Carlos Alfonso, Belen Blanco, Laura Rubio-Perez, Anais Jimenez-Reinoso, Laura Diez-Alonso, Francisco J. Blanco, Laura Sanz, Kenneth A. Howard, Luis Alvarez-Vallina

Summary: This study introduces bispecific antibodies fused with human and mouse sequences that specifically target the tumor microenvironment to provide 4-1BB costimulation. These antibodies show specific targeting of the tumor cells and provide costimulatory activity, with prolonged circulatory half-life and tumor inhibition effects. Combining these antibodies with PD-1-blocking antibodies enhances the therapeutic effect.

First case report of multivisceral transplant from a deceased cardiac death donor

Ane M. Andres, Jose Luis Encinas, Alba Sanchez-Galan, Javier Serradilla Rodriguez, Karla Estefania, Rocio Gonzalez Sacristan, Alida Alcolea, Pilar Serrano, Belen Estebanez, Inigo Velasco Leon, Paula Burgos, Alvaro Gonzalez Rocafort, Bunty Ramchandani, Belen Calderon, Cristina Verdu, Esperanza Jimenez, Paloma Talayero, Pablo Stringa, Itziar de la Pena Navarro, Esther Ramos, Francisco Hernandez Oliveros

Summary: The shortage of pediatric multivisceral donors leads to long waiting times and high mortality rates for pediatric patients. The use of donors after cardiac death, particularly with normothermic regional perfusion, has increased for solid organ transplantation, except for the intestine. This case demonstrates the possibility of reducing waiting time for patients through multivisceral transplantation from donors after cardiac death.

COVID-19 and the antiphospholipid syndrome

Manuel Serrano, Gerard Espinosa, Antonio Serrano, Ricard Cervera

Summary: COVID-19 not only affects the respiratory system, but also has an impact on the cardiovascular system, causing coagulation abnormalities and potentially increasing the risk of thrombotic events and mortality. This multiorgan vascular disease overlaps with other known microangiopathies and may be associated with inflammatory response and coagulopathy.

Summary: In this study, a PD-L1/EGFR symmetric bispecific antibody was developed, which could simultaneously inhibit EGFR-mediated proliferation, effectively block PD-1/PD-L1 interaction, and induce strong antigen-specific antibody-dependent cellular cytotoxicity activity in vitro. Potent therapeutic efficacies of this antibody were observed in two different humanized mouse models.

Bi- and trispecific immune cell engagers for immunotherapy of hematological malignancies

Antonio Tapia-Galisteo, Luis alvarez-Vallina, Laura Sanz

Summary: Immune cell engagers are modified antibodies that have one arm binding a tumor-associated antigen and another arm binding an activating receptor in immune effector cells. These engagers have the potential to revolutionize the treatment of hematological malignancies and are more potent than conventional monoclonal antibodies. The field is growing rapidly, with multiple formats and targets currently in clinical trials, and trispecific antibodies show even more promise by targeting additional tumor-associated antigens or costimulatory receptors.
