The GLP-1 receptor herbal agonist morroniside attenuates neuropathic pain via spinal microglial expression of IL-10 and β-endorphin

Objectives: To assess the protective effect of the glucagon-like peptide-1 receptor (GLP-1R) agonist morroniside against neuropathic pain and its downstream mechanisms of activating microglial GLP-1R/interleukin-10 (IL-10)/beta-endorphin antinociceptive pathway. Methods: Spinal nerve ligation-induced neuropathic pain rats were intrathecally injected with morroniside, with mechanical paw withdrawal threshold being assessed. The expression of spinal and cultured microglia IL-10 and beta-endorphin were detected with qRT-PCR. Key findings: Morroniside alleviated mechanical allodynia in neuropathic rats, which was blocked by inhibiting or depleting microglia. In addition, neutralizing spinal IL-10 or beta-endorphin with specialized antibodies or blocking the m-opioid receptor was able to fully reverse the morroniside-induced mechanical antiallodynia. Morroniside treatment stimulated the gene expression of IL-10 and beta-endorphin in the spinal lumbar enlargements of neuropathic rats as well as in primary cultured microglia. Furthermore, pretreatment with the IL-10 antibody blocked morroniside-stimulated beta-endorphin expression in the spinal cords of neuropathic rats and cultured primary microglia, whereas the beta-endorphin antibody failed to affect morroniside-stimulated gene expression of IL-10. Conclusions: These results reveal that morroniside produces therapeutic effects in neuropathy through spinal microglial expression of IL-10 and subsequent beta-endorphin after activation of GLP-1R. Development of biosensor for measuring oxidative stress of fish

Summary: This study developed a biosensor that can rapidly quantify superoxide concentrations in biological samples by immobilizing cytochrome c on the surface of a gold electrode. The sensor exhibited good correlation with superoxide concentrations in the range of 0.58-9.33 μM and showed acceptable performance in Nile tilapia eyeball interstitial sclera fluid.

Summary: A discrimination method for adulterated milk based on temperature-perturbed 2D infrared correlation spectroscopy and NPLS-DA is proposed. The method can accurately classify pure and adulterated milk by calculating 2D correlation spectra and building discrimination models. Compared to the conventional 3D stacked map, this method achieves higher discrimination accuracy.

Temperature-perturbed two-dimensional generalized correlation characteristic slice spectra combined with multivariate method to identify adulterated milk

Summary: To improve the accuracy of adulterated milk discrimination, a detection method based on temperature-perturbed generalized two-dimensional (2D) correlation characteristic slice spectra was proposed. A total of 240 samples were prepared and the infrared attenuated total reflection spectra were collected at different temperatures. Discrimination models based on 2D correlation characteristics slice spectra were established using N-way partial least squares discriminant analysis (NPLS-DA). The results showed that the proposed method effectively extracted characteristic information and improved the accuracy of discrimination.

Slice spectra approach to synchronous Two-dimensional correlation spectroscopy analysis for milk adulteration discriminate

Summary: The discrimination approach of adulterated milk was proposed using synchronous two-trace two-dimensional (2T2D) correlation slice spectra and multivariate method. The method was demonstrated to be useful for analyzing two common adulterants, melamine and urea. The study showed that the prediction accuracies of the slice spectral models were higher than those of the 1D whole models, indicating that the 2T2D slice spectra improved the discrimination accuracy of adulterants in milk. The complexity of the slice spectra was significantly reduced compared to the whole matrix of synchronous 2T2D correlation spectra, making it a more efficient method for adulterated milk detection.

Identification of adulterated milk based on auto-correlation spectra

Summary: A qualitative analysis method for melamine-adulterated milk based on two-trace two-dimensional auto-correlation spectra was proposed. Infrared spectroscopy was used to measure the spectral data of pure milk and melamine-adulterated milk. The intensity of auto-correlation peaks at specific wave numbers was selected as independent variables for modeling. The method achieved 100% accuracy for individual brands and 99.05% accuracy for all four brands combined.

Effect of water change on quality deterioration of Pacific white shrimp (Litopenaeus vannamei) during partial freezing storage

Summary: This study evaluated the correlation between water changes and quality deterioration of Litopenaeus vannamei during partial freezing storage. The results showed significant increases in cross-sectional area and equivalent diameter, but irregular growth in roundness and longiness of the ice crystals. Bound water and immobilized water decreased significantly, while free water increased significantly during storage. Quality determination revealed a significant decrease in total sulfhydryl and Ca2+-ATPase, but a significant increase in disulfide bonds. Correlation analysis showed significant correlations between cross-sectional area and total sulfhydryl, Ca2+-ATPase, and disulfide bonds, respectively. Additionally, significant correlations were found between water distribution index and Ca2+-ATPase, disulfide bonds. Predictive models for ice crystal growth were developed based on cross-sectional area and equivalent diameter using the Arrhenius model.

Study on the development of carbon nanotube enhanced biosensor for gender determination of fish

Summary: Efficient production of seedlings in aquaculture farms requires control of male and female ratios through sex distinguishing. This study aims to develop a highly sensitive biosensor system utilizing single-walled carbon nanotubes (SWCNTs) to improve the efficiency and accuracy of fish gender determination. The concentration of 11-ketotestosterone (11-KT) in plasma is utilized as an indicator. The biosensor was fabricated by immobilizing an anti-11-KT antibody onto a disk-type gold electrode surface modified with a self-assembled monolayer (SAM) and SWCNTs.

A polyaniline-modified immunosensor for E.coli O157:H7 detection based on electrical parameters and support vector regression

Summary: An immunochromatographic test strip for quantitative detection of E.coli O157:H7 was developed using electrochemical impedance technology and the support vector regression (SVR) model, with modified antibodies and a sandwich immune complex formed to change the conductivity of the capture pad. The SVR model showed high correlation coefficients for predicting E.coli O157:H7, demonstrating the feasibility of using impedance electrical parameters and SVR technology for quantitative prediction.

TP53INP2 modulates the malignant progression of colorectal cancer by reducing the inactive form of β-catenin

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

The four subunits of rabbit skeletal muscle lactate dehydrogenase do not exert their catalytic action additively

Summary: Oligomeric enzymes are known for their higher catalytic rates compared to monomeric enzymes, but the extent of additivity in their activity is still not well understood. This study used tetrameric rabbit lactate dehydrogenase as a model to examine the kinetics of its catalytic action. Surprisingly, when the concentration of the limiting reactant exceeded that of a single subunit, there was a significant slowdown in the enzyme's conformational rearrangements.

The results showed that indium curcumin and vanadyl curcumin exhibited higher binding affinities and stronger inhibitory effects on amyloid fibrillation compared to gallium curcumin.

The effects of noise exposure on hippocampal cognition in C57BL/6 mice via transcriptomics
