Introduction. Breast cancer is the most common cancer occurring in women and causing the highest number of deaths among them. The role of xenoestrogens has been the subject of many studies in the pathogenesis of breast cancer. Less is known about the impact of xenoestrogens on the effectiveness of hormone therapy used to treat breast cancer, and thus possible drug-xenostrogen interactions. Objective. The aim of this review is to summarize the current state of knowledge and present perspectives for further research on the impact of xenoestrogens on the effectiveness of drugs used in the treatment of hormone-dependent breast cancer. Current state of knowledge. Phytoestrogens, in particular flavonoid genistein, are the best studied group of xenoestrogens in terms of interaction with drugs used in the treatment of breast cancer, due to their frequent use, including their use in alleviating the adverse effects of hormone therapy. Analyzing the current state of knowledge, it seems that phytoestrogens intake should be avoided during conventional anti-cancer treatment. Of the other xenoestrogens, bisphenol A (BPA) is one of the best-tested compounds for interactions with drugs used to treat breast cancer. It has been shown that bisphenol A could reduced therapeutic effect of active tamoxifen metabolite and cytostatics used in breast cancer treatment. Conclusion. Confirmation in clinical trials of the results obtained in vitro and in vivo tests, would enable the creation of specific recommendations for patients undergoing breast cancer treatment, especially hormone therapy. An area requiring further research is the analysis of the effects of xenoestrogens other than phytoestrogens, e.g. metalloestrogens, on the effects of drugs used in the treatment of breast cancer.

Establishing laboratory-specific reference intervals for TSH and fT4 by use of the indirect Hoffman method

Sylwia Placzkowska, Malgorzata Terpinska, Agnieszka Piwowar

Summary: This study aimed to determine reference intervals (RIs) for TSH and fT4 using the indirect Hoffman method and compare them with commonly used RIs. The results showed varying RIs in different age groups of the examined populations. The established RIs for fT4 were similar to the manufacturer and literature data, while RIs for TSH differed between age groups.

Differences in Expression of Selected Interleukins in HIV-Infected Subjects Undergoing Antiretroviral Therapy

Beata Szymanska, Karolina Jurkowska, Brygida Knysz, Agnieszka Piwowar

Summary: This study aimed to examine the effect of combined antiretroviral therapy (cART) on the expression of select cytokines in HIV-infected individuals. The results showed that cART significantly decreased IL-7 levels and increased IL-15 levels in HIV-infected men. Further research in this area may help confirm the role of these cytokines in the disease.

Effect of Combined Antiretroviral Therapy on the Levels of Selected Parameters Reflecting Metabolic and Inflammatory Disturbances in HIV-Infected Patients

Karolina Jurkowska, Beata Szymanska, Brygida Knysz, Agnieszka Piwowar

Summary: This study assessed the impact of combined antiretroviral therapy (cART) on selected parameters related to metabolism, cardiovascular diseases, and inflammation in HIV-infected patients. The results indicated significant changes in the expression of these parameters during HIV infection and cART. The data may contribute to better monitoring of the infection and optimizing therapy to prevent comorbidities.

Summary: This study investigated the activity of superoxide dismutase (SOD) and the levels of copper (Cu) and zinc (Zn) in women with polycystic ovary syndrome (PCOS), as well as the gene polymorphism of SOD1. The results showed lower SOD1 activity and Cu, Zn concentration in women with PCOS compared to the control group. Insulin resistance in PCOS further decreased SOD1 activity and increased Cu concentration and Cu/Zn value.

Aromatase Inhibitors and Risk of Metabolic and Cardiovascular Adverse Effects in Breast Cancer Patients-A Systematic Review and Meta-Analysis

Kamila Boszkiewicz, Agnieszka Piwowar, Pawel Petryszyn

Summary: Aromatase inhibitors (AIs) are commonly used as first-line therapy for hormone-dependent breast cancer patients. However, their use is associated with increased risk of cardiovascular events and the development of insulin resistance and diabetes. This meta-analysis found that AIs are associated with a higher risk of cardiovascular events compared to tamoxifen (TAM), but no significant association was found with hypertension, dyslipidemia, hyperglycemia, or weight gain.

Molecular Mechanism of Lipotoxicity as an Interesting Aspect in the Development of Pathological States-Current View of Knowledge

Katarzyna Lipke, Adriana Kubis-Kubiak, Agnieszka Piwowar

Summary: Free fatty acids (FFAs) play vital roles in the organism but disturbances in fatty acid homeostasis can lead to lipotoxicity, contributing to the development of diseases such as insulin resistance and diabetes. Lipotoxic effects primarily affect organs such as the pancreas, liver, skeletal muscles, cardiac muscle, and kidneys.

The Effect of Metalloestrogens on the Effectiveness of Aromatase Inhibitors in a Hormone-Dependent Breast Cancer Cell Model

Kamila Boszkiewicz, Helena Moreira, Ewa Sawicka, Anna Szyjka, Agnieszka Piwowar

Summary: Progressive industrialization, urbanization, and consumerism have led to increased contamination of the environment with endocrine-disrupting compounds (EDCs), which play a crucial role in the rise of hormone-dependent cancers, including breast cancer. This study aimed to investigate the impact of exposure to specific xenoestrogens, commonly found in daily life (aluminum in antiperspirants and deodorants; chromium (III) in dietary supplements and drugs), on the effectiveness of hormone therapy in breast cancer treatment. In vitro tests were conducted on breast cancer cell lines exposed to xenoestrogens, drugs, and their combinations, and the results showed that such exposure can impair the efficacy of hormone therapy.

HIGH GLUCOSE REDUCES THE ANTI-TUMOR ACTIVITY OF AROMATASE INHIBITORS IN A HORMONE-DEPENDENT BREAST CANCER CELL MODEL

Kamila Boszkiewicz, Agnieszka Piwowar

Summary: One-third of breast cancer patients with diabetes have a 40% higher risk of mortality. Metalloestrogens may reduce the effectiveness of aromatase inhibitors (AIs), but high glucose conditions have a greater impact on their activity. Therefore, controlling hyperglycemia and individualized treatment regimens may have important therapeutic implications for cancer patients with diabetes.

Exploring the Relationship between Antioxidant Enzymes, Oxidative Stress Markers, and Clinical Profile in Relapsing-Remitting Multiple Sclerosis

Anna Bizon, Justyna Chojdak-Lukasiewicz, Slawomir Budrewicz, Anna Pokryszko-Dragan, Agnieszka Piwowar

Summary: This study aimed to investigate the alterations in the antioxidant balance in patients with RRMS and the factors influencing it. The study included 161 patients and 29 healthy individuals, and measured the activity of SOD, GPx, and CAT, as well as the concentration of IL-6, LPO, TOS, and TAS. The results showed decreased GPx activity and increased CAT activity in patients with RRMS compared to the control group, and these activities were influenced by gender and disease-modifying therapies. The findings suggest that enhancing GPx activity may be a potential therapeutic strategy for mitigating oxidative stress in RRMS.

Anna Bizon, Agnieszka Piwowar

Summary: The expression of recombinant paraoxonase 1 (PON1) in the human embryonic kidney (HEK293F) cell line was investigated. It was demonstrated that the HEK293F platform is useful for obtaining recombinant human PON1 protein. The addition of high-density lipoprotein increased serum arylesterase and lactonase activity of PON1, while low-density lipoprotein, copper ions, and iron ions decreased PON1 activity.
