Melatonin promotes seed germination under salinity and enhances the biosynthesis of steviol glycosides in Stevia rebaudiana Bertoni leaves

Melatonin (MEL) can act as a plant growth regulator and biostimulator in stressful situations. Using MEL in seed pretreatment also affects the future growth of plants. Therefore, this research investigated the effects of MEL on seed germination and seedling growth under NaCl in in vitro conditions. The additional effects of MEL on the accumulation of steviol glycosides (SGs) and on the expression of appropriate genes were also studied. Five mu M of MEL was the best concentration for seed germination, while 20 mu M exerted a positive impact on the biomass of stevia plantlets. NaCl significantly decreased seed germination, but MEL alleviated this effect when seeds were germinated in 50 mM of NaCl. Under salinity, the values of almost all morphological traits decreased as MEL concentration increased. The highest amounts of stevioside and rebaudioside A (Reb A) were observed as a result of treating seeds with 5 and 20 mu M of MEL, respectively. When adding NaCl, positive impacts of MEL on the accumulation of both SGs were also observed. Expression analyses of the genes involved in SGs biosynthesis was explored in seeds and leaves, and the transcripts of key enzymes occurred in both the tissues. However, quantitative polymerase chain reaction (qPCR) analysis showed that all tested genes were upregulated in younger leaves, contrary to older ones. Also in younger, rather than older, leaves SG gene expression varied according to MEL concentration. This study, therefore, presents the promising potential of MEL for improving stevia seed germination under salinity conditions and for enhancing the production of SGs in stevia plants.

The growth dynamics and secondary metabolite accumulation were analyzed, and the antioxidant capacity and antimicrobial potential were evaluated. The results showed variations in antioxidant capacity and antimicrobial activity among the tested species.

Endophytic bacteria from in vitro culture of Leucojum aestivum L. a new source of galanthamine and elicitor of alkaloid biosynthesis

Summary: A novel strain of endophytic bacterium Paenibacillus lautus was isolated from Leucojum aestivum plants and found to have the ability to produce alkaloids and growth regulators. Elicitation with the bacterium increased alkaloid production and had various effects on plant growth and metabolism.

Antioxidative Properties and Acrylamide Content of Functional Wheat-Flour Cookies Enriched with Wild-Grown Fruits

Summary: This study evaluated the impact of using widely grown fruits as functional ingredients in wheat-flour cookies on their antioxidative properties and reduction of acrylamide content. The results showed that fruit-enriched cookies had higher antioxidative properties and significantly reduced acrylamide content compared to control cookies. Wild rose and rowan cookies were the most highly rated and widely accepted by the panelists. This suggests that cookies enriched with wild-grown fruits could be a promising novel snack food.

Summary: This study aimed to determine the content of biologically active substances, total phenolic content, and antioxidant activity of Cordyceps militaris fruiting bodies, mycelia, and food supplements. The results showed that substrate composition and cultivation method influenced the properties of the mushroom materials. An important aspect of the study was the evaluation of bioactive substance content after extraction in a simulated digestive system, which allowed for determining potential bioavailability. The mycelium and fruiting bodies of C. militaris were found to be ideal food supplements and pharmaceutical agents that could provide prohealth substances for humans.

Seeds of Stevia rebaudiana Bertoni as a Source of Plant Growth-Promoting Endophytic Bacteria with the Potential to Synthesize Rebaudioside A

Summary: In this study, a new strain of Pantoea vagans, SRS89, was isolated from surface-sterilized stevia seeds and evaluated using various methods. The strain exhibited certain properties, such as yellow color and Gram-negative classification. Genome sequencing and annotation revealed numerous genes involved in transcription, amino acid and carbohydrate metabolism, as well as carotenoid and terpenoid biosynthesis. The strain also showed the ability to synthesize rebaudioside A, a valuable metabolite.

Hypsizygus marmoreus as a Source of Indole Compounds and Other Bioactive Substances with Health-Promoting Activities

Summary: This study evaluated the chemical composition of Hypsizygus marmoreus fruiting bodies, mycelium, and in vitro cultures, and found that H. marmoreus mycelium is a good source of indole compounds, bioelements, glucans, and lovastatin. In vitro cultures enriched with zinc and magnesium salts showed increased levels of certain compounds, including 5-methyltryptamine, melatonin, protocatechuic acid, sterols, and total glucans.

Summary: This study aimed to evaluate the effects of methyl jasmonate (MeJA) on the production of triterpenes in submerged cultures of Ganoderma applanatum. The results showed that MeJA promoted the production of ganoderic acid A and ganoderic acid C and exhibited significant cytotoxic activity against prostate cancer cells.

Studies on the Accumulation of Secondary Metabolites and Evaluation of Biological Activity of In Vitro Cultures of Ruta montana L. in Temporary Immersion Bioreactors

Summary: This study evaluated the effects of cultivation time and different concentrations of plant growth regulators on the biomass and secondary metabolite accumulation of R. montana. The major secondary metabolites in R. montana cultures were coumarins, with xanthotoxin and bergapten as the dominant compounds. The extract obtained from the biomass grown on the 0.1/0.1 LS medium showed the best antioxidant activity.

Elicitation and Enhancement of Phenolics Synthesis with Zinc Oxide Nanoparticles and LED Light in Lilium candidum L. Cultures In Vitro

Summary: In this study, the phenolic compound content and composition of Lilium candidum adventitious bulbs formed in vitro were determined. The use of zinc oxide nanoparticles (ZnO NPs) in the culture medium, along with LED light spectra dominated by red and blue light, significantly increased the content of phenolic compounds.

The Content of Phenolic Compounds in Stevia rebaudiana (Bertoni) Plants Derived from Melatonin and NaCl Treated Seeds

Summary: This study investigated the content of phenolic acids and flavonoids in stevia leaves as affected by treatment with melatonin and germination in NaCl conditions. The results showed that phenolic acids were more abundant than flavonoids in stevia leaves. Treatment with melatonin increased the content of both phenolic acids and flavonoids, and 500μM melatonin under salinity conditions had the most favorable effect on the synthesis of phenolic acids. The content of phenolic compounds also varied depending on the age of the leaves.

Ruta chalepensis L. In Vitro Cultures as a Source of Bioactive Furanocoumarins and Furoquinoline Alkaloids

Summary: Due to decreasing crop acreages and climate change, there has been increasing interest in alternative sources of bioactive compounds. Plant biotechnology offers the potential for cultivating valuable medicinal plant species using in vitro methods, which are not dependent on climate and soil conditions. This study investigated the ability of Ruta chalepensis bioreactor cultures to produce important linear furanocoumarins and furoquinoline alkaloids.
