Summary: This study isolated arterial feto-placental endothelial cells from control and preeclamptic placentas to investigate the potential protective effects of hydroxychloroquine (HCQ) in an in vitro model of endothelial activation. The results showed that HCQ did not adversely affect wound healing and endothelial barrier integrity, and it had a slight regulatory effect on IL-8 and VCAM-1 gene expression in the presence of inflammation. These findings support the safety of HCQ and highlight the need for clinical trials to evaluate its therapeutic effects in preeclampsia.

Summary: Excessive nutrient consumption and obesity cause inflammation, which spreads to the central nervous system and leads to oxidative stress and mitochondrial dysfunction, resulting in brain damage. Current treatments for obesity, such as lifestyle changes and pharmacotherapy, may have adverse effects on health, so it is necessary to search for new therapeutic alternatives. L-carnitine (LC) and acetyl-L-carnitine (LAC) have been found to have neuroprotective effects against oxidative stress and mitochondrial dysfunction in obesity. Therefore, this study conducted a literature review on the effect of LC and LAC on brain damage caused by obesity, highlighting their potential as promising treatments for restoring metabolic homeostasis in the CNS.

Summary: This study found that the level of N6-methyladenosine (m6A) in placental tissue of early-onset preeclampsia (ePE) patients was reduced. Wilms' tumor 1-associating protein (WTAP), a regulator of m6A, was found to regulate trophoblast cell migration and invasion. The study also revealed the interaction between WTAP and the high mobility group nucleosomal binding domain 3 (HMGN3) gene, and the effect of m6A on the stability of HMGN3 mRNA. WTAP enhanced the stability of HMGN3 mRNA by binding to its m6A site, and HMGN3 was recognized and regulated by insulin-like growth factor 2 mRNA-binding protein 1 (IGF2BP1). The results suggest that m6A modification plays an important role in the pathogenesis of ePE.

Summary: Mitochondrial health declines with age, making older patients more susceptible to adverse drug reactions. This study investigated the effectiveness of l-carnitine and acetylcarnitine as biomarkers for age-related, drug-induced changes in mitochondrial metabolism. The results showed that drug-induced alterations in mitochondrial metabolism were not reflected in the blood by l-carnitine or acetylcarnitine levels, suggesting that changes in muscle function may be more relevant for identifying individuals at increased risk for adverse drug reactions.

In vitro studies suggest that hypoxia and inflammation may contribute to the downregulation of PEG10. Summary: This study examined factors associated with the antihypertensive effects of esaxerenone and the incidence of serum potassium elevation in patients with hypertension. Results showed that factors such as sex, weight, hypertension duration, prior antihypertensive treatment, plasma aldosterone concentration, urinary albumin-to-creatinine ratio, and smoking were associated with changes in blood pressure. Factors significantly associated with serum potassium elevation included baseline serum potassium, renal function, urinary albumin-to-creatinine ratio, blood pressure, and hypertension grade. The study concluded that esaxerenone is effective for patients with various backgrounds, but careful management is needed for those at risk of elevated serum potassium.

Summary: p-Coumaric acid (p-CA) is a plant compound with anti-cancer activities. This study designed a nano-liposomal carrier containing p-CA to enhance its effectiveness against melanoma cells. The findings showed that the liposomal form of p-CA had a greater impact on the cells. Kinetic modeling indicated that the best fitting model was zero-order.

Summary: This study investigated the expression of ncRNAs other than miRNAs in different histologic subtypes of canine mammary gland tumors (MGT). Three aberrantly expressed ncRNAs were identified as potential biomarkers for differentiating MGT subtypes. YRNA and tRFs expression levels were found to be decreased in metastatic compared to primary MGT cell lines.

Summary: In this study, the researchers used signal analysis to study the instantaneous amplitude and phase of sarcomeric oscillations in skeletal muscle. They identified two types of oscillations, sarcomeric oscillations and sarcosynced oscillations, and visualized their behavior during propagating waves. The researchers discovered the presence of sarcomeric defect holes and sarcomeric collision holes, which are important indicators for understanding the oscillation properties of sarcomeres. This finding has important implications for improving our understanding of muscle function and its regulatory mechanisms.

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

Summary: Oligomeric enzymes are known for their higher catalytic rates compared to monomeric enzymes, but the extent of additivity in their activity is still not well understood. This study used tetrameric rabbit lactate dehydrogenase as a model to examine the kinetics of its catalytic action. Surprisingly, when the concentration of the limiting reactant exceeded that of a single subunit, there was a significant slowdown in the enzyme's conformational rearrangements.

Summary: This study explored the inhibitory effects of gallium curcumin, indium curcumin, and vanadyl curcumin on the amyloid fibrillation of hen egg white lysozyme, as well as the binding interactions of these metal complexes with the enzyme. The results showed that indium curcumin and vanadyl curcumin exhibited higher binding affinities and stronger inhibitory effects on amyloid fibrillation compared to gallium curcumin.
