v-Src Oncogene Induces Trop2 Proteolytic Activation via Cyclin D1

Proteomic analysis of castration-resistant prostate cancer demonstrated the enrichment of Src tyrosine kinase activity in approximately 90% of patients. Src is known to induce cyclin D1, and a cyclin D1-regulated gene expression module predicts poor outcome in human prostate cancer. The tumor-associated calcium signal transducer 2 (TACSTD2/Trop2/M1S1) is enriched in the prostate, promoting prostate stem cell self-renewal upon proteolytic activation via a g-secretase cleavage complex (PS1, PS2) and TACE (ADAM17), which releases the Trop2 intracellular domain (Trop2 ICD). Herein, v-Src transformation of primary murine prostate epithelial cells increased the proportion of prostate cancer stem cells as characterized by gene expression, epitope characteristics, and prostatosphere formation. Cyclin D1 was induced by v-Src, and Src kinase induction of Trop2 ICD nuclear accumulation required cyclin D1. Cyclin D1 induced abundance of the Trop2 proteolytic cleavage activation components (PS2, TACE) and restrained expression of the inhibitory component of the Trop2 proteolytic complex (Numb). Patients with prostate cancer with increased nuclear Trop2 ICD and cyclin D1, and reduced Numb, had reduced recurrence-free survival probability (HR = 4.35). Cyclin D1, therefore, serves as a transducer of v-Src-mediated induction of Trop2 ICD by enhancing abundance of the Trop2 proteolytic activation complex.

Findings revealed a significant amount of unused opioids, and a prescription of approximately 225 morphine milligram equivalents provided appropriate analgesia, but larger initial prescriptions may lead to increased opioid consumption without reducing refill requests.

Summary: The alpha V beta 6 integrin is upregulated during prostate cancer progression and specifically expressed in androgen receptor-negative tumors. By delivering ITGB6-targeting siRNAs into PrCa cells using sEVs, the expression of beta 6 subunit can be downregulated, leading to reduced cell adhesion and migration in PrCa cells.

Summary: The study reveals that many human tumors have reduced levels of Mic60, an essential scaffold of mitochondrial structure, which leads to the disruption of mitochondrial integrity. Surprisingly, these tumors show decreased cell proliferation, resistance to cell death, and activation of a gene expression program related to innate immunity and cytokine/chemokine signaling. This process induces epithelial-mesenchymal transition, tumor cell movement, and promotes metastatic dissemination.

Increased incidence of marginal ulceration following conversion of sleeve gastrectomy to Roux-en-Y gastric bypass: a multi-institutional experience

Summary: This study evaluated the incidence of marginal ulcers (MUs) after primary and secondary Roux-en-Y Gastric Bypass (RYGB). The results showed that patients who underwent conversion of Sleeve Gastrectomy to RYGB had a higher incidence of MUs compared to those who had primary RYGB or Gastric Banding to RYGB. Furthermore, the time to MU development was significantly shorter in patients who underwent conversion surgery.

Summary: Despite therapeutic advances in prostate cancer treatment, metastatic castration-resistant prostate cancer remains a challenge. Highly expressed proteins STEAP1-4 have been identified as significant drivers of prostate cancer aggressiveness and metastasis. However, their other biological functions and understanding of heterotrimers and homotrimers are limited. This review highlights the importance of further research on the role of STEAP1-4 in prostate cancer and their potential clinical applications.

TOMM20 as a Potential Prognostic Biomarker in Chordoma: Results From a High-Volume, Single-Center Study

Summary: The objective of this study was to identify novel signaling pathways and biomarkers associated with chordoma using a large tumor bank. Clinical and pathologic data were collected from 73 patients, and tumor microarrays were constructed from 61 chordoma specimens. The study found that higher expression of TOMM20 was associated with recurrent and metastatic chordomas. TIGAR and TOMM20 expressions were significantly higher in primary lesions, and a TOMM20 composite score of 3 or higher decreased overall and recurrence-free survival.

General surgeons' comfort and urologists' perceptions of bladder trauma management

Summary: Using a physician-reported survey, this study assesses the comfort level of general surgeons in managing bladder trauma. The results show that general surgeons have higher reported comfort levels compared to urologists. Factors such as preoperative diagnosis and patient presentation timing do not significantly affect general surgeons' decision to consult urology.
