Summary: In this study, we found that fatostatin induces ferroptosis in glioblastoma by inhibiting the AKT/mTORC1/GPX4 signaling pathway. Fatostatin also inhibits cell proliferation and the EMT process through the AKT/mTORC1 signaling pathway. Additionally, we developed a p28-functionalized PLGA nanoparticle loaded with fatostatin, which can penetrate the blood-brain barrier and target GBM.

Summary: This study reveals that radioresistant glioblastoma multiforme (GBM) cells exhibit upregulation of SLC25A22, a mitochondrial glutamate transporter, resulting in accumulation of cytosolic glutamate. The accumulated glutamate enhances glutathione production and proline synthesis in the cells, leading to increased resistance to ionizing radiation and a more invasive phenotype. Genetic inhibition of SLC25A22 reduces radioresistance and aggressiveness of GBM in vitro and in vivo. These findings suggest that SLC25A22 may serve as a significant therapeutic target for overcoming radioresistance in GBM.

Summary: In this study, it was found that Syndecan 1 (SDC1) and Transglutaminase 2 (TGM2) were overexpressed in radioresistant glioblastoma (GBM) cells, contributing to the radioresistance of GBM. Mechanistically, SDC1 transported TGM2 from the cell membrane to the cytoplasm, then to lysosomes by interacting with flotillin 1 (FLOT1). TGM2 recognized betaine homocysteine methyltransferase (BHMT) on autophagosomes, promoting the fusion of lysosomes and autophagosomes.

Targeting Furin may provide a potential therapeutic approach for colorectal cancer stem cells.

The programming of immune cells using different strategies has improved their therapeutic influence and specificity. However, effective immunotherapy must consider the complete complexity of immune cell function.
