High Levels of Apoptosis Are Induced in the Human Colon Cancer HT-29 Cell Line by Co-Administration of Sulforaphane and a Peptide Nucleic Acid Targeting miR-15b-5p

Sulforaphane (SFN) is one of most important dietary constituents of broccoli (Brassica oleracea) and other cruciferous vegetables, which have been reported to exhibit health benefits, including prevention and therapy of cancer, such as colorectal carcinoma (CRC). The objective of this study was to determine whether the anticancer effect of SFN on colon cancer HT-29 cell line could be improved by the combined treatment with molecules inhibiting microRNAs (miRNAs) involved in CRC. As miRNA inhibiting molecules we focused on peptide-nucleic acids (PNAs). As miRNA to be targeted, miR-15b-5p was selected on the basis of several information present in the literature and confirming that miR-15b-5p is overexpressed in colon cancer patients, and that its targeting decreases cell migration and metastasis in colorectal cancer. In this article, we described for the first time the efficacy of targeting miR-15b-5p by using a PNA against miR-15b-5p (R8-PNA-a15b), functionalized with an octoarginine peptide (R8) for maximizing cellular uptake. The miR-15b-5p downregulation in the colon cancer HT-29 cell line was associated with inhibition of in vitro cell growth and activation of the proapoptotic pathway, demonstrated by a sharp increase of late apoptotic cells in HT-29-treated cell populations. A second conclusion of this study is that the R8-PNA-a15b might be proposed in combo-therapy associated with SFN. To our knowledge, no report is available in the literature on a combination between SFN and miRNA-targeting molecules. Our data demonstrate that this combined treatment leads to a very high proportion of apoptotic HT-29 cells (over 85%), a value higher than the sum of the values of apoptotic cells obtained after singularly administered regents (either SFN or R8-PNA-a15b).

Summary: Glioblastoma multiforme (GBM) is a highly lethal tumor with no curative treatment available. This study investigated the potential of a combined therapy using sulforaphane (SFN) and a peptide nucleic acid (PNA) to target miR-15b-5p in GBM cells. The results showed that SFN and PNA-a15b synergistically induced apoptosis in GBM cells, suggesting the feasibility of using combined treatments based on PNAs and nutraceuticals to stimulate apoptosis in GBM.

Testing Sirolimus and other geroprotective drugs might be a key strategy to improve the overall health resilience of aged populations. This study analyzed the activity of CD4(+) and CD8(+) T cells in beta-Thalassemia patients treated with Sirolimus and found that it positively impacted the biological activity and number of memory T cells.

Production and Characterization of K562 Cellular Clones Hyper-Expressing the Gene Encoding α-Globin: Preliminary Analysis of Biomarkers Associated with Autophagy

Summary: The objective of this study was to develop an experimental model system for studying the reduction of toxic alpha-globin aggregates in beta-thalassemia. K562 cellular clones were produced that stably produced high levels of alpha-globin protein and showed increased apoptosis and autophagy. This model system can be used to screen pharmacological agents that activate autophagy and reduce alpha-globin accumulation, as well as for new therapeutic approaches targeting the reduction of alpha-globin gene expression.

Summary: We report the co-existence of parallel and antiparallel conformations of E3/K3 and E3/R3 heterodimeric coiled-coil systems in solution for the first time. Introducing a furanylated amino acid in the (EIAALEK)(3) sequence enabled the freezing of the respective coiled-coil complexes through photo-induced covalent crosslinking. Computational simulations and fluorescence experiments based on pyrene-pyrene stacking further confirmed the presence of both conformations in solution.

Summary: Starting from a list of nine dysregulated miRNAs in early-stage CRC patients, the study verified the therapeutic effects of anti-miRNA PNAs targeting the most relevant up-regulated miRNAs. PNAs down-regulated these target miRNAs and induced apoptosis in CRC cellular models. Combining anti-miRNA PNAs with pro-apoptotic compounds like sulforaphane resulted in high levels of apoptosis induction.

Methacrylated Silk Fibroin Additive Manufacturing of Shape Memory Constructs with Possible Application in Bone Regeneration

Summary: In this study, a crosslinked methacrylated silk (Sil-MA) grid structure was developed, which can recover its shape under UV light and has biocompatibility and osteogenic support. Sil-MA was chemically and physically characterized, and tested for its biocompatibility according to ISO protocols. Digital Light Processing and pneumatic extrusion techniques were used to manufacture the Sil-MA grid structure, demonstrating its shape memory property and potential application in bone regeneration.
