Pharmaceutical perspective on bioactives from Alstonia scholaris: ethnomedicinal knowledge, phytochemistry, clinical status, patent space, and future directions

Alstonia scholaris (L.) R. Br (Apocynaceae), a tropical tree native to Indian subcontinent, Australasia, and Malay Peninsula is well documented in Traditional Chinese Medicine and Ayurveda to cure fever, pain, inflammation, cancer, respiratory, and skin disorders. Nearly 169 alkaloids have been reported along with iridoids, coumarins, flavonoids and steroids from this plant. The highlights of current review are demonstrated in six tables encompassing ethnomedicinal values, pharmacological activities, phytochemistry, clinical significances, and patents associated with A. scholaris. The data analysis of these tables revealed the requirements to convert existed knowledge into finished products. For example, strictamine isolated from this plant has shown an equipotent antiviral activity to that of acyclovir. However, the discovery of such new potent antiviral indole alkaloids will require structure activity relationship studies with natural analogues of this scaffold. In conclusion, despite of over 100 patents and numerous pharmacological activities of A. scholaris, only one product has reached the market. The report stipulates the gap between the existing knowledge and the requisites for commercialization of A. scholaris derived products. Graphic abstract

Exophiarin, an isocoumarin from the fungus Exophiala sp. with antihyperglycemic activity

Summary: The chemical characterization of ethyl acetate extract of Exophiala sp. led to the isolation of three compounds, including a new isocoumarin named exophiarin. The structure was elucidated using 1D, 2D NMR and HRMS data, and the compound showed moderate improvement in glucose uptake activity in rat skeletal muscle cell line L6 when tested.

Isolation and characterization of a novel flavanone glycoside from an endemic plant Haplanthodes neilgherryensis

Summary: Lichens have been used in traditional medicines worldwide for wound healing and treating skin disorders and respiratory diseases. In this study, the researchers investigated 31 endolichenic fungi isolated from 32 lichens found on mangrove plants in Sri Lanka. They identified three cytotoxic molecules from one of the fungi, Talarotnyces pinophilus, and evaluated their anticancer potential against various cancer cell lines.

Exploration of Potent Cytotoxic Molecules from Fungi in Recent Past to Discover Plausible Anticancer Scaffolds

Summary: Fungi can produce various scaffolds with unique biological activities, but so far, no molecule derived from fungi has become an anti-cancer drug on the market. Every year, numerous cytotoxic molecules of fungal origin are published, making it necessary to critically analyze these compounds to identify the potent ones. A review published in 2014 mentioned the best cytotoxic fungal metabolites and their status in drug development. In this report, we included 176 cytotoxic molecules isolated from fungi after 2014 and categorized them based on their potencies. Emphasis was given to discuss the 42 molecules that showed IC50 values below 1 mu M. This review provides potent fungal scaffolds that can be prioritized as drug candidates for cancer therapeutics.

Bioprospecting of an Endolichenic Fungus Phanerochaete sordida Isolated from Mangrove-Associated Lichen Bactrospora myriadea

Summary: Bioassay-guided fractionation of the ethyl acetate extract from the endolichenic fungus Phanerochaete sordida led to the isolation of a bioactive compound. The compound exhibited high antioxidant activity, moderate anti-inflammatory and antibacterial activity, and moderate tyrosinase inhibition and cytotoxicity. In vitro and in silico experiments confirmed the bioactivities and drug-like properties of the compound.

Summary: Parkinson's disease (PD) is a progressive neurodegenerative disease with no permanent cure. The accumulation of toxic alpha-synuclein and neuronal inflammation are key factors in its pathology. Swertiamarin (SWE), a natural product, has been found to have anti-inflammatory effects and a potential reversal effect on alpha-syn accumulation. In this study, SWE showed anti-inflammatory activity in glial cells and neuroprotective effects in a mouse model of PD. Further research is needed to fully understand the mechanisms involved.

However, redundant isolation of metabolites hinders discovery rates. To trigger biosynthetic genes in fungi, induction strategies beyond standard conditions are necessary. Advancements in computational tools aid in connecting gene clusters and their metabolite production, leading to the identification of numerous cryptic metabolites.
