In vivo antitumour properties of tribenzyltin carboxylates in a 4T1 murine metastatic mammary tumour model: Enhanced efficacy by PLGA nanoparticles

This study reports the in vivo performance of two tribenzyltin carboxylate complexes, tri(4-fluorobenzyl)tin [(N,N-diisopropylcarbamothioyesulfanyll acetate (C1) and tribenzyltin isonicotinate (C9), in their native form as well as in a poly(lactic-co-glycolic acid) (PLGA)-based nanoformulation, to assess their potential to be translated into clinically useful agents. In a 4T1 murine metastatic mammary tumour model, single intravenous administration of C1 (2.7 mg/kg) and C9 (2.1 mg/kg; 2.1 mg/kg C9 is equivalent to 2.7 mg/kg C1) induced greater tumour growth delay than cisplatin and doxorubicin at equivalent doses, while a double-dose regimen demonstrated a much greater tumour growth delay than the single-dose treated groups. To improve the efficacy of the complexes in vivo, C1 and C9 were further integrated into PLGA nanoparticles to yield nanosized PLGA-C1 (183.7 +/- 0.8 nm) and PLGA-C9 (163.2 +/- 1.2 nm), respectively. Single intravenous administration of PLGA-C1 (2.7 mg C1 equivalent/kg) and PLGA-C9 (2.1 mg C9 equivalent/kg) induced greater tumour growth delay (33% reduction in the area under curve compared to that of free C1 and C9). Multiple-dose administration of PLGA-C1 (5.4 mg C1 equivalent/kg) and PLGA-C9 (4.2 mg C9 equivalent/kg) induced tumour growth suppression at the end of the study (21.7 and 34.6% reduction relative to the size on day 1 for the double-dose regimen; 73.5 and 79.0% reduction relative to the size on day 1 for the triple-dose regimen, respectively). Such tumour growth suppression was not observed in mice receiving multiple-dose regimens of free C1 and C9. Histopathological analysis revealed that metastasis to the lung and liver was inhibited in mice receiving PLGA-C1 and PLGA-C9. The current study has demonstrated the improved in vivo antitumour efficacies of C1 and C9 compared with conventional chemotherapy drugs and the enhancement of the efficacies of these agents via a robust PLGA-based nanoformulation and multiple-drug administration approach.

Novel Gemcitabine-Re(I) Bisquinolinyl Complex Combinations and Formulations With Liquid Crystalline Nanoparticles for Pancreatic Cancer Photodynamic Therapy

Summary: With a 5-year survival rate of less than 10%, pancreatic ductal adenocarcinoma (PDAC) is a highly lethal cancer. Gemcitabine is commonly used as the first-line treatment, but its efficacy is often reduced due to poor cellular penetration and tumor resistance. This study explores the synergistic use of a transition metal complex photosensitizer, Re(I) bisquinolinyl complex, and gemcitabine against PDAC. The encapsulation of these compounds into liquid crystalline nanoparticles (LCNPs) was found to enhance their therapeutic efficiency. The results suggest the potential use of this combination therapy and the importance of nano-formulation in PDAC treatment.

Self-assembled chitosan-insulin oral nanoparticles-A critical perspective review

Summary: Chitosan is a natural polysaccharide that is biodegradable, bioadhesive, and biocompatible. It has been extensively studied for oral insulin drug delivery systems. Electrostatic interaction between chitosan and insulin leads to the formation of self-assembled nanoparticles, which can protect insulin from degradation and improve intestinal uptake. Functionalized chitosan nanoparticles also offer opportunities for targeted and prolonged delivery. This review provides a comprehensive guide for formulating stable insulin-chitosan nanoparticles using mild ionic interactions.

Evaluation of Folate-Functionalized Nanoparticle Drug Delivery Systems-Effectiveness and Concerns

Summary: Targeting folate receptors is a potential solution for addressing low tumor selectivity in conventional chemotherapeutics. Folate-functionalized nanoparticle drug delivery systems show promise due to their advantages, although none have entered clinical trials. In vitro evidence supports their efficacy compared to non-targeting carriers and free drug formulations. However, in vivo biodistribution studies reveal issues with rapid liver uptake and reduced tumor uptake, potentially influenced by the expression of folate receptor β by activated macrophages in the liver, as well as the route of administration and tumor location. Generalizing nanoparticles reported in various publications is challenging due to different formulations, lack of characterization, and experimental settings, making it difficult to determine the accurate factors influencing targeting effectiveness.

Analysis of stabilization mechanisms in β-lactoglobulin-based amorphous solid dispersions by experimental and computational approaches

Summary: Our previous work demonstrated that beta-lactoglobulin-stabilized amorphous solid dispersion (ASD) loaded with 70 % indomethacin remains stable for over 12 months. We further investigated the stabilization mechanisms by testing five other drug molecules and using experimental techniques and molecular dynamics simulations. The results showed that steric confinement, hydrogen bonding, and the glass transition temperature of the drug molecule play important roles in stabilizing ASDs with high drug loadings.

Do the enantiomers of ketamine bind enantioselectively to human serum albumin?

Summary: The binding of drugs to plasma proteins, such as human serum albumin (HSA), is crucial for determining pharmacokinetic parameters. This study investigated the enantioselective binding of S- and R-ketamine to HSA. It was found that ketamine has weak affinity to HSA, with no significant differences in binding behavior between the individual enantiomers and the racemate. The aromatic ring and N-methyl group were identified as the most strongly involved structural moieties in the binding of ketamine to HSA.

Targeting fusion proteins of the interleukin family: A promising new strategy for the treatment of autoinflammatory diseases

Fusion protein technology, with its ability to enhance therapeutic efficacy, has been explored for developing anti-inflammatory drugs. This review discusses the efficacy of fusion protein drugs developed with ILs or their receptors in treating autoinflammatory diseases, highlighting the potential of this technology in future anti-inflammatory drug development.

The use of polymorphic state modifiers in solid lipid microparticles: The role of structural modifications on drug release performance

Summary: This study investigated the correlation between the structure and release properties of solid lipid microparticles (MPs) with different liquid additives. The additives accelerated the conversion of the unstable alpha-form of tristearin to the stable beta-polymorph and caused structural modifications in the MPs. The presence of additives prolonged the drug release in water and resulted in higher release profiles in biorelevant media. The findings suggest that the release behavior can be influenced by the polymorphism and supramolecular-level structural modification of lipid formulations containing crystal modifiers.

Impact of drug load and polymer molecular weight on the 3D microstructure of printed tablets

This provides a promising direction for the future development of a safe, effective, and minimally invasive perivascular drug delivery method to treat vascular disease.
