Summary: The study identified common host factors for three coronaviruses and critical host pathways supporting infection by all of them, including phosphatidylinositol phosphate biosynthesis and cholesterol homeostasis. Inhibition of these pathways reduced replication of all three coronaviruses, while the lysosomal protein TMEM106B appeared unique to SARS-CoV-2 infection. These findings provide important insights for understanding the coronavirus life cycle and developing host-directed therapies.

Summary: Traditional Chinese medicine (TCM) has a long history in treating infectious diseases, particularly the common cold and flu. It is difficult to differentiate between these two based on symptoms alone. While there is a flu vaccine, there is no specific vaccine or medication for the common cold. This review systematically evaluates the scientific evidence supporting the efficacy of TCM in treating colds for the first time, considering theoretical principles, clinical research, pharmacological perspectives, and underlying mechanisms. The findings suggest that TCM is effective and safe for treating colds, and may be used as a complementary or alternative approach. Further high-quality clinical trials and pharmacological studies are needed for verification and optimization of TCM's clinical practice and scientific research for cold treatment.

Summary: This study evaluated the effectiveness of antihistamine-decongestant-analgesic combinations compared with placebo or other active controls in reducing the duration and alleviating the symptoms of the common cold. The results showed that these combinations had some general benefit in adults and older children, but the effect on individual symptoms may be too small to be clinically relevant. Adverse effects were more common in the antihistamine-decongestant and analgesic-decongestant groups compared to the control group. There was no evidence of effectiveness in young children.

Summary: Lung fibroblasts, expressing IRAK-M, have been shown to play a crucial role in subepithelial fibrosis and airway remodeling. In this study, the effect of IRAK-M on lung fibroblasts was investigated through silencing or overexpressing IRAK-M. The results revealed that silencing IRAK-M enhanced the proliferation, migration, and invasiveness of lung fibroblasts, while overexpression of IRAK-M inhibited these cellular phenotypes. Modulation of IRAK-M expression also affected the production of inflammatory and fibrotic cytokines by the fibroblasts. These findings suggest that targeting IRAK-M could be a potential strategy to slow down airway remodeling.

Summary: By using single-cell RNA sequencing, whole-cell patch clamp, and calcium transient techniques, we investigated the effects of propofol and sevoflurane on the gene transcriptome and ion channels in human fetal prefrontal cortex cells. Both propofol and sevoflurane promoted microgliosis, but only sevoflurane decreased the transcriptional similarity of microglia. Both anesthetics transiently altered the transcriptome profiling of various cell types, such as microglia, excitatory neurons, interneurons, astrocytes, and oligodendrocyte progenitor cells, for less than 2 hours. Our study provides valuable insights for future mechanistic studies of general anesthetics in clinical settings.
