IRAK-M Regulates Proliferative and Invasive Phenotypes of Lung Fibroblasts

Summary: Lung fibroblasts, expressing IRAK-M, have been shown to play a crucial role in subepithelial fibrosis and airway remodeling. In this study, the effect of IRAK-M on lung fibroblasts was investigated through silencing or overexpressing IRAK-M. The results revealed that silencing IRAK-M enhanced the proliferation, migration, and invasiveness of lung fibroblasts, while overexpression of IRAK-M inhibited these cellular phenotypes. Modulation of IRAK-M expression also affected the production of inflammatory and fibrotic cytokines by the fibroblasts. These findings suggest that targeting IRAK-M could be a potential strategy to slow down airway remodeling.

Summary: By using single-cell RNA sequencing, whole-cell patch clamp, and calcium transient techniques, we investigated the effects of propofol and sevoflurane on the gene transcriptome and ion channels in human fetal prefrontal cortex cells. Both propofol and sevoflurane promoted microgliosis, but only sevoflurane decreased the transcriptional similarity of microglia. Both anesthetics transiently altered the transcriptome profiling of various cell types, such as microglia, excitatory neurons, interneurons, astrocytes, and oligodendrocyte progenitor cells, for less than 2 hours. Our study provides valuable insights for future mechanistic studies of general anesthetics in clinical settings.

IRAK-M has effects in regulation of lung epithelial inflammation

Summary: This study found that IRAK-M regulates epithelial immune response in lung epithelial cells. IRAK-M knockdown leads to increased secretion of inflammatory cytokines and chemokines, and the secretion of CXCL10 is partly mediated through JNK and p38 MAPK pathways. Additionally, asthma patients carrying G/G genotypes have significantly higher levels of serum CXCL10 than those carrying A/A genotypes.
