Osteocalcin (OC) is a bone-derived hormone that regulates energy metabolism. OC exists in two forms, carboxylated (GlaOC) and uncaboxylated (GluOC), but only the latter appears to have an endocrine function. In this study, we prepared an extract containing both Gla- and GluOC from boiled pork bone using 0.2M carbonate buffer at pH 9.5, and tested whether the extract had beneficial effects on improving metabolic parameters in obese mice. The extract equivalent of 1.2g of GluOC/mouse was orally administrated to C57BL/6 female mice fed a high-fat, high-sucrose diet. Daily oral administration of the extract for four weeks decreased blood glucose levels and promoted glucose tolerance as well as insulin sensitivity. Our study shows for the first time that boiled pork bones are a source material for osteocalcin in the large-scale production of supplements designed to improve glucose metabolism.

Volume-regulated chloride channel regulates cell proliferation and is involved in the possible interaction between TMEM16A and LRRC8A in human metastatic oral squamous cell carcinoma cells

Shohei Yoshimoto, Miho Matsuda, Kenichi Kato, Eijiro Jimi, Hiroshi Takeuchi, Shuji Nakano, Shunichi Kajioka, Etsuko Matsuzaki, Takao Hirofuji, Ryuji Inoue, Masato Hirata, Hiromitsu Morita

Summary: The study found that VRAC is highly functional and associated with proliferation in HST-1 cells, but not in HaCaT cells. The pharmacological profile of VRAC in HST-1 was similar to previous reports, and inhibition of VRAC with DCPIB led to apoptosis in HST-1 cells. Knockdown of TMEM16A and LRRC8A attenuated VRAC activity in HST-1 cells.

Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry Imaging of Tissues via the Formation of Reproducible Matrix Crystals by the Fluorescence-Assisted Spraying Method: A Quantification Approach

Summary: A fluorescence-assisted spraying method was developed to overcome the variability in matrix crystals and improve the reproducibility of analyte intensity in matrix-assisted laser desorption/ionization mass spectrometry imaging. The method utilized a fluorescent reagent, rhodamine 6G, to achieve a constant fluorescence intensity. The proposed method successfully quantified nifedipine and ferulic acid in a rat kidney section and visually determined the accumulation of ferulic acid in the kidney over time.

Summary: In this study, the interaction of cyclic naphthalene diimide derivatives with G-quartet DNA was investigated under dilute or molecular crowding condition. The results showed that compound 4 exhibited the highest stability towards the TA-core. Additionally, compound 3 demonstrated specific growth inhibition of cancer cell line Ca9-22 at IC50 <0.03 μM, with the highest expression level of telomerase mRNA.

Summary: The study established a novel analytical assay for peptide metabolites from glycinin, using MALDI-MS combined with amine derivatization with coumarin. The use of coumarin derivatization under specific conditions led to improved MS detection of peptides, allowing for successful characterization of glycinin-derived metabolites in rats.

Summary: Anthocyanins are natural color pigments found in plants, such as berries, vegetables, and grapes. They have various physiological effects and are considered promising functional food ingredients. However, the bioavailability of anthocyanins, especially their absorption behaviors, is still unclear.
