Prostaglandin E2/EP4 axis is upregulated in Spondyloarthritis and contributes to radiographic progression

Daniele Mauro, Archita Srinath, Giuliana Guggino, Vicky Nicolaidou, Stefania Raimondo, Jonathan J. Ellis, Jessica Whyte, Maria Maddalena Nicoletti, Marco Romano, Tony John Kenna, Juan D. Canete, Riccardo Alessandro, Aroldo Rizzo, Matthew Arthur Brown, Nicole J. Horwood, Nigil Haroon, Francesco Ciccia

Summary: Ankylosing spondylitis (AS) is an inflammatory disease that leads to spine ankylosis. The mechanisms of new bone formation in AS are not fully understood, but Single Nucleotide Polymorphisms (SNPs) in the PTGER4 gene have been associated with AS. This study found that the PGE2-EP4 axis, which is involved in inflammation and bone metabolism, may contribute to radiographic progression in AS. Baseline serum PGE2 levels and the PTGER4 SNP rs6896969 were found to predict progression in AS patients. Increased expression of EP4/PTGER4 was observed in immune cells, synovial tissue, and bone marrow of AS patients. The PGE2/EP4 axis also induced bone formation when monocytes were cocultured with mesenchymal stem cells.

UBE2L3 regulates TLR7-induced B cell autoreactivity in Systemic Lupus Erythematosus

Daniele Mauro, Sotiria Manou-Stathopoulou, Felice Rivellese, Elisabetta Sciacca, Katriona Goldmann, Victoria Tsang, Isabelle Lucey-Clayton, Sara Pagani, Farah Alam, Debasish Pyne, Ravindra Rajakariar, Patrick A. Gordon, James Whiteford, Michele Bombardieri, Costantino Pitzalis, Myles J. Lewis

Summary: Study finds that UBE2L3 plays a critical role in TLR7-mediated NF-??B activation in Systemic Lupus Erythematosus (SLE). Inhibition of UBE2L3 by Dimethyl Fumarate (DMF) can effectively suppress TLR7-induced NF-??B activation, B cell differentiation, and autoantibody production in SLE. This suggests that UBE2L3 inhibition could be a potential therapeutic strategy for SLE by repurposing DMF.

Sexual dysfunctions of rheumatological patients are a neglected issue: Results from a national survey of Italian Society of Rheumatology

Luigi Napolitano, Ilenia Pantano, Lorenzo Romano, Luigi Cirillo, Celeste Manfredi, Francesco Mastrangelo, Giovanni Maria Fusco, Daniele Mauro, Lorenzo Spirito, Roberto La Rocca, Davide Arcaniolo, Corrado Aniello Franzese, Carmine Sciorio, Marco Romano, Marco De Sio, Vincenzo Mirone, Francesco Ciccia

Summary: This study assessed the attention, knowledge, and barriers to discussing sexual dysfunction (SD) in patients with rheumatologic diseases among members of the Italian Society of Rheumatology. The results showed that only half of the respondents occasionally asked patients about SD related to their rheumatologic pathologies, and a significant percentage never did so. The main barriers to discussing sexual health were lack of time, patient discomfort, and lack of knowledge/experience. However, most respondents expressed the potential usefulness of attending SD courses to improve knowledge about these conditions.
