Mesoporous Silica Nanoparticles Trigger Liver and Kidney Injury and Fibrosis Via Altering TLR4/NF-κB, JAK2/STAT3 and Nrf2/HO-1 Signaling in Rats

Mesoporous silica nanoparticles (MSNs) represent a promising inorganic platform for multiple biomedical applications. Previous studies have reported MSNs-induced hepatic and renal toxicity; however, the toxic mechanism remains unclear. This study aimed to investigate MSNs-induced hepatic and nephrotoxicity and test the hypothesis that altered TLR4/MyD88/NF-kappa B, JAK2/STAT3, and Nrf2/ARE/HO-1 signaling pathways mediate oxidative stress, inflammation, and fibrosis induced by MSNs. Rats were administered 25, 50, 100, and 200 mg/kg MSNs for 30 days, and samples were collected for analyses. MSNs induced functional and histologic alterations, increased the levels of reactive oxygen species (ROS), lipid peroxidation and nitric oxide, suppressed antioxidants, and Nrf2/HO-1 signaling in the liver and kidney of rats. MSNs up-regulated the expression of liver and kidney TLR4, MyD88, NF-kappa B p65, and caspase-3 and increased serum pro-inflammatory cytokines. In addition, MSNs activated the JAK2/STAT3 signaling pathway, down-regulated peroxisome proliferator activated receptor gamma (PPAR gamma), and promoted fibrosis evidenced by the increased collagen expression and deposition. In conclusion, this study conferred novel information on the role of ROS and deregulated TLR4/MyD88/NF-kappa B, JAK2/STAT3, PPAR gamma, and Nrf2/ARE/HO-1 signaling pathways in MSNs hepatic and nephrotoxicity. These findings provide experimental evidence for further studies employing genetic and pharmacological strategies to evaluate the safety of MSNs for their use in nanomedicine.

Summary: This study evaluated the levels of heavy metals in Plantago ovata and domestic goats in the eastern region of Saudi Arabia, which were influenced by the petroleum industry. The results showed that the petroleum industry increased the levels of lead, cadmium, nickel, and vanadium in both the plant and animal samples, leading to liver and kidney injury, oxidative stress, and upregulation of pro-inflammatory and apoptosis markers.

Formulation of Osimertinib Nano Lipid Carriers: Optimization, Characterization and Cytotoxicity Assessment

Summary: In this study, orally administered Osimertinib nanostructured lipid carriers (OSM-NLCs) were successfully prepared and evaluated for their drug release, permeation, and cytotoxicity against lung cancer cells. The results showed that OSM-NLCs exhibited high drug release and permeation flux, as well as lower cytotoxicity compared to pure OSM.

Summary: The present study aimed to develop MZBSo bilosomes and MZBSo gel. MZBSo presented nano-sized vesicles with high entrapment efficiency and suitable ζ potential, while MZBSoG2 showed good pH, viscosity, spreadability, and a slower drug release. MZBSoG2 was non-irritant and exhibited significant antifungal activity. Stability study results showed no significant changes. MZ-loaded gels could improve therapeutic efficacy.

Summary: The qualitative study of copper-cobalt ingot sample was conducted using laser induced breakdown spectroscopy (LIBS), and CuCo2O4 nanoparticles were synthesized using the alloy as a target material for Nd:YAG laser ablation. The composition and morphology of the synthesized nanoparticles were determined using various techniques including X-ray diffraction (XRD), energy dispersive X-ray (EDX) analysis, transmission electron microscopy (TEM), scanning electron microscopy (SEM), and dynamic light scattering (DLS). The synthesized nanoparticles exhibited antibacterial activity and potential for magnetic hyperthermia.

Photothermal Hyperthermia Study of Ag/Ni and Ag/Fe Plasmonic Particles Synthesized Using Dual-Pulsed Laser

Summary: Magneto-plasmonic Ag/Ni and Ag/Fe nanoparticles were synthesized using a dual-pulsed Q-switched Nd:YAG 1064 nm laser ablation method. The optical and magnetic properties were investigated, and the morphologies and phase structures of the nanoparticles were characterized. The synthesized nanoparticles exhibited plasmonic hyperthermia properties, reaching high temperatures when irradiated with a NIR 808 nm laser. These findings suggest the potential use of silver-based plasmonic nanoparticles for hyperthermia treatment.

Effective Removal of Methylene Blue by Mn3O4/NiO Nanocomposite under Visible Light

Summary: Wastewater treatment is indispensable to prevent adverse health effects and deterioration of the quality of life on earth. Photocatalysis using nanocomposites of manganese oxide (Mn3O4) and nickel oxide (NiO) has been studied for its effectiveness in purifying wastewater. The synthesized nanomaterials were characterized using various techniques and the photocatalytic performance of the Mn3O4/NiO nanocomposite (NC) was evaluated. The results show that the NCs have a high photocatalytic degradation efficiency of 95% for removing methylene blue (MB) dye from wastewater.

Cadmium cardiotoxicity is associated with oxidative stress and upregulated TLR-4/NF-kB pathway in rats; protective role of agomelatine

Summary: This study evaluated the protective effects of agomelatine (AGM) against cadmium (Cd)-induced cardiotoxicity in rats and found that AGM modulated the TLR-4/NF-kB pathway and HSP70. The results showed that AGM alleviated Cd-induced myocardial injury, reduced oxidative stress, and enhanced antioxidant effects.

Summary: The biocompatible chitosan/zeolite-A (CS/ZA) and β-cyclodextrin/zeolite-A (CD/ZA) biocomposites were produced by hybridizing synthetic zeolite-A (ZA) with chitosan and β-cyclodextrin. The hybridization with the two biopolymers resulted in significant increases in the loading capacities of the 5-fluorouracil drug (5-Fu), with CD/ZA exhibiting the highest loading capacity. The release activities of 5-Fu from both CS/ZA and CD/ZA showed continuous and controlled profiles, with CD/ZA releasing the drug at a faster rate.

Summary: In this study, natural bentonite clay was modified to enhance its drug loading capabilities for 5-fluorouracil. The modification process involved exfoliation of the clay into nanosheets and further functionalization with methanol. The modified carrier (Mth/EXBE) showed higher loading site density and faster drug release compared to the raw clay. Additionally, the cytotoxicity of 5-fluorouracil was significantly enhanced by the modified carriers.

Summary: Natural kaolinite can be modified into separated single nanosheets (KNs) and scrolled nanoparticles as nanotubes (KNTs). The synthetic nanostructures show advanced oxaliplatin-medication (OXAP) delivery systems. The loading capacity of KNs and KNTs is significantly increased compared to raw kaolinite, and they exhibit classic kinetic and isotherm behaviors. The release of OXAP from KNs and KNTs shows continuous and regulated profiles, with KNTs demonstrating faster release. The cytotoxicity of the synthesized structures is significantly enhanced on cancer cell lines, and they greatly increase the cytotoxicity of OXAP.
