Plant mitochondrial Complex I composition and assembly: A review

In the mitochondrial inner membrane, oxidative phosphorylation generates ATP via the operation of several multimeric enzymes. The proton-pumping Complex I (NADH:ubiquinone oxidoreductase) is the first and most complicated enzyme required in this process. Complex I is an L-shaped enzyme consisting of more than 40 subunits, one FMN molecule and eight Fe-S clusters. In recent years, genetic and proteomic analyses of Complex I mutants in various model systems, including plants, have provided valuable insights into the assembly of this multimeric enzyme. Assisted by a number of key players, referred to as assembly factors, the assembly of Complex I takes place in a sequential and modular manner. Although a number of factors have been identified, their precise function in mediating Complex I assembly still remains to be elucidated. This review summarizes our current knowledge of plant Complex I composition and assembly derived from studies in plant model systems such as Arabidopsis thaliana and Chlamydomonas reinhardtii. Plant Complex I is highly conserved and comprises a significant number of subunits also present in mammalian and fungal Complexes I. Plant Complex I also contains additional subunits absent from the mammalian and fungal counterpart, whose function in enzyme activity and assembly is not clearly understood. While 14 assembly factors have been identified for human Complex I, only two proteins, namely GLDH and INDH, have been established as bona fide assembly factors for plant Complex I. Summary: In the process of transitioning towards carbon-neutral economies, the aviation sector is a key contributor to greenhouse gas emissions due to its dependence on fossil fuels. As an alternative, biofuels derived from microalgae are being developed due to their potential for lipid accumulation. This review discusses the development status, opportunities, and challenges of technologies that can transform microalgae into aviation fuels, along with an assessment of the present techno-economic feasibility. Further developments are needed to reduce the costs and improve the overall economics of microalgae cultivation and harvesting.

Cell adaptation of the extremophilic red microalga Galdieria sulphuraria to the availability of carbon sources

Summary: The study found that there was no significant difference in the biomass, fatty acid composition, protein, and storage carbohydrate contents of the extremophilic red microalga Galdieria sulphuraria 074W when grown in heterotrophy compared to photoautotrophic condiitons. However, the presence of glucose resulted in a decrease in pigment content, while the presence of glycerol did not. Transcriptome analysis revealed the transcriptional response of the cells to light and dark environments, as well as the involvement of horizontally transferred gene families in response to the availability of different organic and inorganic sources.

Oscillations of chlorophyll fluorescence after plasma membrane excitation in Chara originate from nonuniform composition of signaling metabolites in the streaming cytoplasm

Summary: Excitable cells of higher plants and characean algae respond to stressful stimuli by generating action potentials that influence chlorophyll fluorescence and photosynthesis for an extended period of time. While plant leaves exhibit a reversible depression in the efficiency of photosystem II reaction after an individual action potential, characean algae show long-lasting oscillations of photosystem II reaction efficiency after firing an action potential. This study investigates the possible mechanisms behind these oscillations and suggests that they are a result of metabolic rearrangements in chloroplasts and the cyclosis cessation-recovery cycle induced by calcium influx during action potentials. The findings also indicate that fluidic communications between different cell regions play a role in these oscillations, and the inhibition of oscillations occurs when these communications are restricted or eliminated.

Summary: The conformation of chromophores in isolated phycobiliproteins is heterogeneous, but not in the entire phycobilisome (PBS). Under low-energy excitation, there is no significant uphill energy transfer from the core to the peripheral rods of the PBS, while transfer from the terminal emitters to bulk allophycocyanin chromophores is highly probable.
