Single-cell RNA-Seq reveals transcriptional heterogeneity and immune subtypes associated with disease activity in human myasthenia gravis

Summary: This study used single-cell sequencing to reveal the transcriptomes of individual cells in patients with myasthenia gravis, identifying 13 major cell clusters and 39 cell subgroups. It was found that B cells, CD4(+) T cells, and monocytes showed more heterogeneity in MG patients, with expanded CD4(+) T cells. Further analyses showed that B cells in MG exhibited higher activity towards plasma cell differentiation, CD4(+) T cell subsets were unbalanced, and monocytes showed highly activated inflammatory pathways. Additionally, a disease-relevant subgroup of CD180(-) B cells was discovered, which was associated with disease activity and anti-AChR antibody levels in MG patients.

Summary: The study identified age, tumor size, surgery, primary site, and tumor stage as main factors affecting the prognosis of skull base chordoma. Resection of the tumor as much as possible while ensuring safety, combined with postoperative radiotherapy may be the optimum treatment for skull base chordoma.

Deep Learning Model for Intracranial Hemangiopericytoma and Meningioma Classification

Summary: Our study found that SFT/HPC showed more invasion to venous sinus (p = 0.001), more cystic components (p < 0.001), and more heterogeneous enhancement patterns (p < 0.001). The deep learning model achieved a high classification accuracy of 0.889 with an AUC of 0.91 in the validation set. Feature maps demonstrated distinct clustering of SFT/HPC and meningioma in the training and test cohorts, and the attention of the deep learning model mainly focused on the tumor bulks representing the solid texture features for discrimination.

Summary: The major obstacles in brain cancer treatment, including the blood-tumor barrier (BTB) and quiescent tumor cells, have been addressed in this study. Sox2+ tumor cells in mouse medulloblastoma (MB) project cellular processes to ensheathe capillaries, which depend on the mechanosensitive ion channel Piezo2. Targeting Piezo2 not only compromises the BTB and reduces the quiescence of tumor cells, but also enhances the response to chemotherapy, improving the treatment outcome for brain cancer patients.

A comprehensive machine-learning model applied to MRI to classify germinomas of the pineal region

Summary: In this study, a computational framework was used to predict the histology of pineal region tumors based on radiomics features and demographics. The models that combined demographics and radiomics features performed the best, with an AUC of 0.88. T1 and T2 sequences were found to be the most informative for differentiating pineal tumors.

Alterations in 3D chromatin organization contribute to tumorigenesis of EGFR-amplified glioblastoma

Summary: In this study, the alterations in 3D chromatin organization of EGFR-amplified glioblastoma were explored, and it was found that the chromatin relaxation and unexpected entanglement of chromosome regions were observed in the glioblastoma cells. The activation of compartments reshaped chromatin accessibility and activated tumorigenesis-related genes. The altered topological associating domains (TADs) and chromatin loop alterations were associated with oncogene activation and tumor repressor deactivation.

Systematic Analysis of 4-gene Prognostic Signature in Patients with Diffuse Gliomas Based on Gene Expression Profiles

Summary: The study identified 67 genes associated with malignant progression of diffuse glioma using WGCNA method and established a 4-gene molecular marker including KDELR2, EMP3, TIMP1, and TAGLN2. The molecular marker served as an independent predictor of prognosis in patients with diffuse gliomas, showing good predictive power and associations with clinical and genetic backgrounds of gliomas.
