Cultured mouse peritoneal macrophages release large numbers of similar to 30-nm cholesterol-rich particles. Here, we show that those particles represent fragments of the plasma membrane that are pulled away and left behind during the projection and retraction of filopodia and lamellipodia. Consistent with this finding, the particles are enriched in proteins found in focal adhesions, which attach macrophages to the substrate. The release of particles is abolished by blocking cell movement (either by depolymerizing actin with latrunculin A or by inhibiting myosin II with blebbistatin). Confocal microscopy and NanoSIMS imaging studies revealed that the plasma membrane-derived particles are enriched in 'accessible cholesterol' (a mobile pool of cholesterol detectable with the modified cytolysin ALO-D4) but not in sphingolipid-sequestered cholesterol [a pool detectable with ostreolysin A (OlyA)]. The discovery that macrophages release cholesterol-rich particles during cellular locomotion is likely relevant to cholesterol efflux and could contribute to extracellular cholesterol deposition in atherosclerotic plaques.

Intra- and Intercellular Silver Nanoparticle Translocation and Transformation in Oyster Gill Filaments: Coupling Nanoscale Secondary Ion Mass Spectrometry and Dual Stable Isotope Tracing Study

Zishuang Shao, Paul Guagliardo, Haibo Jiang, Wen-Xiong Wang

Summary: This study utilized high-resolution nanoscale secondary ion mass spectrometry and scanning electron microscopy to visualize the subcellular localization and potential toxicity effects of AgNPs in oyster gill filaments. The findings demonstrated an endocytosis-based entry of AgNPs into cells and identified AgNP-induced autophagy in oyster gill cells. Additionally, the study revealed the transport and sequestration roles of different hemocytes in AgNP distribution.

Summary: Nucleic acid therapeutics have shown promise in modifying gene expression, but there is limited understanding of their cellular uptake and tissue distribution. Utilizing NanoSIMS imaging allows for high sensitivity and spatial resolution in defining the distribution of antisense oligonucleotides, potentially improving the efficacy and delivery of nucleic acid therapeutics for treating human diseases.

Evidence of Carrier Localization in AlGaN/GaN-Based UV Multiple Quantum Wells with Opposite Polarity Domains Provided by Nanoscale Imaging

Mei Cui, Wei Guo, Houqiang Xu, Jie'an Jiang, Li Chen, Somak Mitra, Iman S. Roqan, Haibo Jiang, Xiaohang Li, Jichun Ye

Summary: The research demonstrates that introducing opposite polarity domains in AlGaN-based multiple quantum wells can improve the optical properties of UV emitters, despite the inferior crystalline quality in N-polar domains. By combining space-resolved luminescence peak position and Raman spectroscopy analysis, it is confirmed that carrier localization effect exists in the MQWs.

NanoSIMS Imaging of Bioaccumulation and Subcellular Distribution of Manganese During Oyster Gametogenesis

Nanyan Weng, Paul Guagliardo, Haibo Jiang, Wen-Xiong Wang

Summary: This study investigated the distribution and accumulation of manganese in oyster oocytes and sperm cells, revealing significant differences in subcellular localization and accumulation capacity between the two cell types. Manganese was found to mainly accumulate in cortical granules of oocytes, while it was primarily distributed in the nucleus of sperm cells, indicating sex differentiation in susceptibility to manganese stress. This highlights the importance of considering gender differences in future biomonitoring and ecotoxicological studies of manganese in marine bivalves.
