Gastric signet ring cell carcinoma (GSRCC) is a unique pathological type of gastric carcinoma that is extremely invasive and has a poor prognosis. Expression of microRNAs (miRNAs) has been closely linked to the carcinogenesis of gastric cancer and has been considered as a powerful prognostic marker. The function of miR-935 has never been reported in cancer before. We found, using microRNA array, that expression of miR-935 in GSRCC cell lines is lower than in non-GSRCC cell lines, and enhanced expression of miR-935 in GSRCC cell-lines inhibit cell proliferation, migration and invasion. We also identified Notch1 as a direct target of miR-935. Knockdown of Notch1 reduced proliferation, migration invasion of GSRCC cells, and overexpression Notch1's activated form (Notch intracellular domain) could rescue miR-935's tumor suppressive effect on GSRCC. Expression of miR-935 was lower in gastric carcinoma tissue than in paired normal tissue samples, and lower in GSRCC than in non-GSRCC. Our results demonstrate the inverse correlation between the expression of miR-935 and Notch1 in gastric tissues. We conclude that miR-935 inhibits gastric carcinoma cell proliferation, migration and invasion by targeting Notch1, suggesting potential applications of the miR-935-Notch1 pathway in gastric cancer clinical diagnosis and therapeutics, especially in gastric signet ring cell carcinoma. Summary: This study found that patients with gastric cancer undergoing radical gastrectomy have worse survival outcomes when experiencing significant loss of adipose tissue during neoadjuvant treatment. The loss of visceral adipose tissue and subcutaneous adipose tissue during treatment was associated with poorer overall survival and disease-free survival, while changes in skeletal muscle mass and body weight did not affect survival.

Clinicopathological factors affecting the effect of neoadjuvant chemotherapy in patients with gastric cancer

Lin Jiang, Zhiqiang Ma, Xin Ye, Weiming Kang, Jianchun Yu

Summary: In a retrospective study on gastric cancer patients, factors such as age <60, histological type of poor differentiation or signet-ring cell carcinoma, and weight loss during neoadjuvant chemotherapy were identified as independent risk factors for the effect of neoadjuvant chemotherapy. Patients with weight loss >2.95% during neoadjuvant chemotherapy may have a worse chemotherapy effect. Providing timely nutritional support such as oral nutritional supplements to maintain patients' body weight is crucial for improving the effect of neoadjuvant chemotherapy.

The Sensitivity Prediction of Neoadjuvant Chemotherapy for Gastric Cancer

Summary: Research on predicting the sensitivity of NACT for gastric cancer spans multiple aspects including biomarkers, inflammatory indicators, and imaging assessments. While numerous studies have been conducted on biomarkers, a unified conclusion has not yet been reached. Inflammatory indicators have been shown to be associated with the survival and prognosis of patients undergoing NACT, and imaging assessments such as CT, MRI, and PET may offer unique advantages in early screening for NACT-sensitive patients through careful integration and optimization.

Low urinary iodine is a protective factor of central lymph node metastasis in papillary thyroid cancer: a cross-sectional study

Ziyang Zeng, Kang Li, Xianze Wang, Siwen Ouyang, Zimu Zhang, Zhen Liu, Juan Sun, Xin Ye, Weiming Kang, Jianchun Yu

Summary: An increase in iodine intake has been associated with the rising incidence of papillary thyroid cancer (PTC). However, lower iodine intake was found to be a protective factor for central lymph node metastasis (CLNM) in PTC, particularly in cases with tumors smaller than 1 cm. These results suggest that iodine may play a dual role in PTC development, both as an initiator and a promoter of tumorigenesis.

Hole behavior captured by analysis of instantaneous amplitude and phase of sarcosynced oscillations reveals wave characteristics of sarcomeric oscillations

Seine A. Shintani

Summary: In this study, the researchers used signal analysis to study the instantaneous amplitude and phase of sarcomeric oscillations in skeletal muscle. They identified two types of oscillations, sarcomeric oscillations and sarcosynced oscillations, and visualized their behavior during propagating waves. The researchers discovered the presence of sarcomeric defect holes and sarcomeric collision holes, which are important indicators for understanding the oscillation properties of sarcomeres. This finding has important implications for improving our understanding of muscle function and its regulatory mechanisms.

TP53INP2 modulates the malignant progression of colorectal cancer by reducing the inactive form of β-catenin

Ke Shi, Yunlong Shan, Xiao Sun, Kuida Chen, Qiong Luo, Qiang Xu

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

The four subunits of rabbit skeletal muscle lactate dehydrogenase do not exert their catalytic action additively

Martina Rossi, Fabio Tomaselli, Alejandro Hochkoeppler

Summary: Oligomeric enzymes are known for their higher catalytic rates compared to monomeric enzymes, but the extent of additivity in their activity is still not well understood. This study used tetrameric rabbit lactate dehydrogenase as a model to examine the kinetics of its catalytic action. Surprisingly, when the concentration of the limiting reactant exceeded that of a single subunit, there was a significant slowdown in the enzyme's conformational rearrangements.

Summary: The fatty acid elongation enzyme ELOVL5 plays a critical role in promoting metastasis in prostate cancer. Knocking down ELOVL5 leads to the accumulation of malonyl-CoA, which inhibits fatty acid oxidation in mitochondria.
