2D Visualization of the Psoriasis Transcriptome Fails to Support the Existence of Dual-Secreting IL-17A/IL-22 Th17 T Cells

The present paradigm of psoriasis pathogenesis revolves around the IL-23/IL-17A axis. Dual-secreting Th17 T cells presumably are the predominant sources of the psoriasis phenotype-driving cytokines, IL-17A and IL-22. We thus conducted a meta-analysis of independently acquired RNA-seq psoriasis datasets to explore the relationship between the expression of 1L17A and IL22. This analysis failed to support the existence of dual secreting IL-17A/IL-22 Th17 cells as a major source of these cytokines. However, variable relationships amongst the expression of psoriasis susceptibility genes and of IL17A, IL22, and IL23A were identified. Additionally, to shed light on gene expression relationships in psoriasis, we applied a machine learning nonlinear dimensionality reduction strategy (t-SNE) to display the entire psoriasis transcriptome as a 2-dimensonal image. This analysis revealed a variety of gene clusters, relevant to psoriasis pathophysiology but failed to support a relationship between IL17A and IL22. These results support existing theories on alternative sources of IL-17A and IL-22 in psoriasis such as a Th22 cells and non-T cell populations.

A study identified a novel DDX58 variant, expanding the genotypic spectrum of the disease. Furthermore, detailed ocular and dermatological phenotypes were described for the first time, shedding light on the molecular pathogenesis of SGMRT.

Cytokine responses in nonlesional psoriatic skin as clinical predictor to anti-TNF agents

Summary: This study evaluates the use of baseline molecular expression profiling to assess treatment outcome for patients with psoriasis. The results show a correlation between clinical response and molecular changes, particularly for genes responding to IL-17A/TNF in keratinocytes. Baseline gene expressions in nonlesional skin were found to be the best marker of treatment response, and cytokine gene signatures enriched in baseline nonlesional skin had strong correlations with treatment improvement.

Summary: This study demonstrates that tape stripping with optimized high-resolution transcriptomic profiling can effectively assess and characterize inflammatory responses in the skin. By comparing different samples and sampling methods, the study shows that tape stripping can capture the transcriptome of the upper layers of the skin, providing insights into the molecular components of the immune amplification pathway in psoriasis. Additionally, the study suggests that nonlesional psoriatic skin exhibits activated, proinflammatory changes, which are not captured by full-thickness skin biopsy transcriptome profiling.

Alcohol consumption and smoking in relation to psoriasis: a Mendelian randomization study

Summary: This study aimed to assess the causal associations of alcohol consumption and smoking with psoriasis risk. The findings revealed a causal effect of smoking on psoriasis risk, while no causal relationship was found between alcohol consumption and psoriasis. Restricting smoking could be helpful in reducing the burden of psoriasis.

Enhanced rare disease mapping for phenome-wide genetic association in the UK Biobank

Summary: This study demonstrates the utility of the UK Biobank as a population assay for rare diseases. By constructing a consensus mapping between ICD-10 codes and ORPHA codes and performing genetic association analysis, the researchers identified and characterized 420 rare diseases affecting individuals in the UK Biobank. Significant genetic associations for specific diseases were also identified. The study also highlights the transferability of the mapping method to other databases.

Summary: Using RNA-Seq-based variant discovery, a possible psoriasis-susceptibility locus was identified within the PCSK9 gene. Single-cell RNA-Seq analysis revealed that PCSK9 is primarily expressed in keratinocytes in human skin. The expression of PCSK9 varied across different keratinocyte subpopulations and showed an inverse relationship with IL36G expression. Furthermore, other immune genes were also found to be linked to PCSK9 expression.

Shared genetic risk factors and causal association between psoriasis and coronary artery disease

Summary: ERAP1, ERAP2, and LNPEP, which are aminopeptidases, play a role in autoimmune diseases. The expression of ERAP2 is increased while ERAP1 is decreased in psoriasis patients with autoimmune-linked variants of ERAP. Different layers of skin cells show different expression patterns of aminopeptidases, and in psoriasis, basal keratinocytes also express CCL2 and IL-15. In addition, ERAP2-high skin fibroblasts also express more CXCL14.
