SLAMF1 signaling inducesMycobacterium tuberculosisuptake leading to endolysosomal maturation in human macrophages

Angela Maria Barbero, Aldana Trotta, Melanie Genoula, Rodrigo Emanuel Hernandez Del Pino, Martin Andres Estermann, Josefina Celano, Federico Fuentes, Veronica Edith Garcia, Luciana Balboa, Paula Barrionuevo, Virginia Pasquinelli

Summary: SLAMF1 enhances the defensive response of macrophages against Mycobacterium tuberculosis infection, facilitating increased internalization of the bacteria. Additionally, SLAMF1 is involved in the maturation process of endolysosomes and shows active function at the site of infection in tuberculosis patients.

McLaren, Amalio Telenti, Pere-Joan Cardona, Bonaventura Clotet, Cristina Vilaplana, Javier Martinez-Picado, Nuria Izquierdo-Useros

Summary: The SIGLEC1 null variant is significantly associated with extrapulmonary dissemination of Mycobacterium tuberculosis, potentially leading to delayed immunity and local bacterial spread. Lack of Siglec-1 limits antigen exchange, increasing the risk of extrapulmonary dissemination.

Dysbiosis, malnutrition and enhanced gut-lung axis contribute to age-related respiratory diseases

Vinciane Saint-Criq, Geanncarlo Lugo-Villarino, Muriel Thomas

Summary: Older people are at a higher risk of developing respiratory diseases due to aging, changes in intestinal and lung microbiota, and malnutrition. As communication along the gut-lung axis becomes more permissive with age, the gut and lung microbiota play key roles in maintaining healthy lungs. Nutrition-based preventive strategies targeting the microbiota may be effective in preventing lung disease in elderly populations.

The Cholinergic System Contributes to the Immunopathological Progression of Experimental Pulmonary Tuberculosis

Leon Islas-Weinstein, Brenda Marquina-Castillo, Dulce Mata-Espinosa, Iris S. Paredes-Gonzalez, Jaime Chavez, Luciana Balboa, Jose Luis Marin Franco, Daniel Guerrero-Romero, Jorge Alberto Barrios-Payan, Rogelio Hernandez-Pando

Summary: The cholinergic system is involved in regulating inflammation during bacterial respiratory infections, and studies have shown its presence and role in the immunopathogenesis of pulmonary tuberculosis. Upregulation of the cholinergic system, particularly during late progressive TB, can aid in bacterial growth and immunomodulation within the lung. Manipulating this system therapeutically may offer potential for treating the disease.

Human macrophage polarization shapes B. pertussis intracellular persistence

Hugo A. Valdez, Jose L. Marin Franco, Juan P. Gorgojo, Jimena Alvarez Hayes, Luciana Balboa, Marcelo Fernandez Lahore, Maria C. Sasiain, Maria Eugenia Rodriguez

Summary: The study found that macrophage polarization influences the development of intracellular Bordetella pertussis infections. M1 macrophages exhibited proinflammatory responses, while M2a and M2c macrophages did not induce proinflammatory cytokines but showed increased expression of M2 markers.

Mycobacteria-host interactions in human bronchiolar airway organoids

Nino Iakobachvili, Stephen Adonai Leon-Icaza, Kevin Knoops, Norman Sachs, Serge Mazeres, Roxane Simeone, Antonio Peixoto, Celia Bernard, Marlene Murris-Espin, Julien Mazieres, Kaymeuang Cam, Christian Chalut, Christophe Guilhot, Carmen Lopez-Iglesias, Raimond B. G. Ravelli, Olivier Neyrolles, Etienne Meunier, Geanncarlo Lugo-Villarino, Hans Clevers, Celine Cougoule, Peter J. Peters

Summary: The study found that Mycobacterium tuberculosis and Mycobacterium abscessus mainly exist as extracellular bacteria in human airway organoids, infecting epithelial cells with low efficiency. The organoids respond to infection by modulating cytokine, antimicrobial peptide, and mucin gene expression. Adult stem cell-derived airway organoids can be used to elucidate early events of mycobacterial infection in human settings.

Dysregulation of the IFN-I signaling pathway by Mycobacterium tuberculosis leads to exacerbation of HIV-1 infection of macrophages

Maeva Dupont, Stella Rousset, Thien-Phong Vu Manh, Sarah Catherine Monard, Karine Pingris, Shanti Souriant, Zoi Vahlas, Tomas Velez, Renaud Poincloux, Isabelle I. Maridonneau-Parini, Olivier Neyrolles, Geanncarlo Lugo-Villarino, Christel Verollet

Summary: While tuberculosis is a risk factor for HIV-1-infected individuals, the mechanisms by which Mycobacterium tuberculosis worsens HIV-1 pathogenesis still need to be fully understood. Recent research has shown that HIV-1 infection and spread are exacerbated in macrophages exposed to TB-associated microenvironments.

Brucella abortus RNA does not polarize macrophages to a particular profile but interferes with M1 polarization

Agustina Serafino, Jose L. Marin Franco, Mariano Maio, Aldana Trotta, Melanie Genoula, Luis A. Castillo, Federico Birnberg Weiss, Jose R. Pittaluga, Luciana Balboa, Paula Barrionuevo, M. Ayelen Milillo

Summary: This study found that the RNA molecule of Brucella abortus can interfere with the polarization process of macrophages, thereby affecting the host's immune response to the bacteria. Specifically, this RNA molecule decreased the expression of M1 surface markers on macrophages and reduced the production of Nitrogen Reactive Species. These findings are important for understanding the mechanisms of chronic infection caused by Brucella.

Druggable redox pathways against Mycobacterium abscessus in cystic fibrosis patient-derived airway organoids

Peters, Virginie Molle, Geanncarlo Lugo-Villarino, Kaymeuang Cam, Laurence Berry, Etienne Meunier, Celine Cougoule

Summary: Although Mabs infection can induce oxidative stress, pharmacological activation of antioxidant pathways can better control Mabs growth and reduce its virulence. Genetic and pharmacological inhibition of CFTR is associated with improved Mabs growth and increased virulence. Pharmacological activation of antioxidant pathways can inhibit Mabs growth and improve efficacy when combined with cefoxitin.
