MC-LR is one of major microcystin isoforms with potent hepatotoxicity. In the present study, we aim to: 1) explore the dynamics of MC-LR accumulation and elimination in different tissues of male red swamp crayfish Procambarus clarkii; 2) reveal the mechanisms underlying hepatic antioxidation and detoxification. In the semi-static toxicity tests under the water temperature of 25 +/- 2 degrees C, P. clarkii were exposed to 0.1, 1, 10 and 100 mu g/L MC-LR for 7 days for accumulation and subsequently relocated to freshwater for another 7 days to depurate MC-LR. MC-LR was measured in the hepatopancreas, intestine, abdominal muscle and gill by HPLC. The enzyme activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione S-transferase (GST), content of glutathione (GSH), and transcripts of Mn-sod, cat, gpxl, Mu-gst, heat shock protein90 (hsp90), hsp70 and hsp60 in hepatopancreas were detected. The results showed that P. clarkii accumulated more MC-LR in intestine, and less in abdominal muscle and gill during accumulation period and eliminated the toxin more quickly in gill and abdominal muscle, and comparatively slowly in intestine during depuration period. The fast increase of SOD and CAT activities at early stage, subsequent decrease at later stage of accumulation period and then fast increase during depuration period were partially consistent with the transcriptional changes of their respective genes. GPx was activated by longer MC-LR exposure and gpxl mRNA expression showed uncoordinated regulation pattern compared with its enzyme. Hsp genes were up-regulated when P. clarkii was exposed to MC-LR. Effect of Chronic Exposure to Pesticide Methomyl on Antioxidant Defense System in Testis of Tilapia (Oreochromis niloticus) and Its Recovery Pattern

Shunlong Meng, Xi Chen, Chao Song, Limin Fan, Liping Qiu, Yao Zheng, Jiazhang Chen, Pao Xu

Summary: The study found that lower concentrations of methomyl had no effect on antioxidant levels in the testis of Nile tilapia, while higher concentrations significantly influenced these antioxidants. Recovery tests showed that oxidative damage caused by lower concentrations of methomyl was reversible, whereas damage from higher concentrations was irreversible within 18 days.

Tongtong Yin, Yao Zheng, Tingyan Liu, Xiaofei Wang, Jiancao Gao, Zhijuan Nie, Lili Song, Gangchun Xu, Julin Yuan

Summary: The discharge of wastewater from aquaculture ponds can be damaging to the environment. This study aimed to find the optimal ratio and operating parameters of a wastewater circulation device in order to reduce the discharge of polluted water. The results showed that the removal rates of various pollutants reached satisfactory levels when using the appropriate matching ratio and hydraulic retention time. The second unit of the device exhibited enhanced removal efficiency when microorganisms and activated sludge were added.

Summary: The study investigated the water purification mechanism of microorganism and purification material combinations using various methods. It found that the combination group showed high removal rates of nitrogen, phosphorous, and Mn chemical oxygen demand. The activities of various enzymes increased for different groups, and certain microbial genera served as biomarkers for effective treatment. The addition of purification materials and the presence of specific microorganisms also influenced microbial growth and the purification effects for different substances.

Effect of Methomyl on Growth, Antioxidant System of GIFT (Oreochromis niloticus), and Residue in the Presence of Water Spinach (Ipomoea aquatica Forsk)

Xiaojun Jing, Yao Zheng, James P. Mulbah, Jiazhang Chen, Gangchun Xu

Summary: This study investigated the effects of methomyl on the growth and antioxidant system of GIFT tilapia in the presence of water spinach as a floating bed. The results showed that methomyl exposure increased the feed consumption and body weight of GIFT tilapia. Water spinach helped in reducing the negative impact of methomyl on water quality and altered the hepatic oxidation system of GIFT tilapia to some extent.

Salvianolic Acid B Regulates Oxidative Stress, Autophagy and Apoptosis against Cyclophosphamide-Induced Hepatic Injury in Nile Tilapia (Oreochromis niloticus)

Liping Cao, Guojun Yin, Jinliang Du, Rui Jia, Jiancao Gao, Nailin Shao, Quanjie Li, Haojun Zhu, Yao Zheng, Zhijuan Nie, Weidong Ding, Gangchun Xu

Summary: This study evaluated the protective effect of Sal B on oxidative hepatic injury in Nile tilapia and investigated the potential mechanisms involved. The results showed that Sal B can reduce reactive oxygen species (ROS) levels and resist oxidative injury in hepatocytes induced by CTX. The protective effect of Sal B on Nile tilapia is related to the regulation of Nrf2, AMPK/mTOR, and MAPK pathways.

The Effects of Different Feeding Regimes on Body Composition, Gut Microbial Population, and Susceptibility to Pathogenic Infection in Largemouth Bass

Yao Zheng, Haojun Zhu, Quanjie Li, Gangchun Xu

Summary: This study investigated the effects of different feeds on the metabolic parameters of largemouth bass. Feeding with iced fish increased the content of saturated fatty acids, monounsaturated fatty acids, and some polyunsaturated fatty acids in muscle tissue, while feeding with commercial feed increased the content of polyunsaturated fatty acids and higher fatty acids. Feeding with iced fish led to significant decreases and increases in the abundance of Firmicutes and Tenericutes in the gut microbiota. Feeding with commercial feed enriched pathways related to carbohydrate metabolism and the digestive system, while feeding with iced fish enriched pathways related to disease resistance.
